We investigated the effects of peripheral injection of sauvagine, a CRF2>CRF1 receptor (corticotropin-releasing factor) agonist compared with CRF, on two sets of tonic colorectal distension (CRDs 30, 40, 50 mmHg, 3-min on/off)-induced visceromotor response (VMR) measured as area under the curve (AUC) of abdominal muscle contraction in conscious female rats. Sauvagine (10 or 20 microg/kg, s.c.) abolished the 226.7+/-64.3% and 90.4+/-38.1% increase in AUC to the 2nd CRD compared with the 1st CRD (performed 30 min before) in female Fisher and Sprague-Dawley (SD) rats, respectively. CRF had no effect while the CRF1 antagonist, antalarmin (20 mg/kg, s.c.), alone or with sauvagine, blocked the enhanced response to the 2nd CRD, performed 60 min after the 1st CRD, and reduced further the AUC by 33.5+/-23.3% and 63.5+/-7.2%, respectively in Fisher rats. These data suggest that peripheral CRF2 receptor activation exerts antinociceptive effects on CRD-induced visceral pain, whereas CRF1 contributes to visceral sensitization.  相似文献

Exposure of oilseed rape (Brassica napus) plants to increasing leaf temperatures between 15 degrees C and 25 degrees C increased photorespiratory NH(4)(+) production from 0.7 to 3.5 micromol m(-2) s(-1). Despite the 5-fold increase in the rate of NH(4)(+) production, the NH(4)(+) concentration in root and leaf tissue water and xylem sap dropped significantly, whereas that in the leaf apoplastic fluid remained constant. The in vitro activity of glutamine synthetase (GS) in both leaves and roots also increased with temperature and in all cases substantially exceeded the observed rates of photorespiratory NH(4)(+) production. The surplus of GS in oilseed rape plants was confirmed using GS2 antisense plants with 50% to 75% lower in vitro leaf GS activity than in the wild type. Despite the substantial reduction in GS activity, there was no tendency for antisense plants to have higher tissue NH(4)(+) concentrations than wild-type plants and no overall correlation between GS activity and tissue NH(4)(+) concentration was observed. Antisense plants exposed to leaf temperatures increasing from 14 degrees C to 27 degrees C or to a trifold increase in the O(2) to CO(2) ratio did not show any change in steady-state leaf tissue NH(4)(+) concentration or in NH(3) emission to the atmosphere. The antisense plants also had similar leaf tissue concentrations of glutamine, glycine, and serine as the wild type, whereas glutamate increased by 38%. It is concluded that photorespiration does not control tissue or apoplastic levels of NH(4)(+) in oilseed rape leaves and, as a consequence, that photorespiration does not exert a direct control on leaf atmosphere NH(3) fluxes.  相似文献

An infection with the rare mycosis Chrysosporium parvum was diagnosed in a European beaver (Castor fiber) shot in northern Sweden. The animal was in normal body condition and no signs of disease were observed. In the lungs a large number of nodules, up to 5 mm diameter, were observed. A large number of adiaspores were observed in the interstitium of the lungs and in the mediastinal lymph node. A chronic inflammatory reaction dominated by mononuclear leukocytes and giant cells was observed around the spores. This is the first report of adiaspiromycosis (Chrysosporium parvum) in the European beaver.  相似文献

Almost homogeneous populations representing different developmental stages of somatic embryos (globular, torpedo-shaped, plantlets) and vacuolated cells were obtained from a cell suspension culture of carrot. The concentrations of S-adenosylmethionine (SAM), S-adenosylhomocysteine (SAH) and methylated DNA were determined in embryos at different developmental stages and were found to increase during somatic embryogenesis. The highest increase during embryogenesis was a 5-fold increase in the level of SAM. A considerable increase in the methylation index (SAM/SAH ratio) was also found. We propose that the levels of SAM and SAH may be involved in the control of somatic embryogenesis by affecting the level of DNA methylation, which in turn might cause differential changes in gene activation. An increase in the level of SAM may be a prerequisite for progression of embryogenesis and the development of complete embryos.  相似文献

The enterochromaffin-like (ECL) cells represent the predominant endocrine cell population in the acid-producing part of the stomach of both experimental animals and man. These cells actively produce and store histamine in addition to an anticipated but as yet unidentified peptide hormone and are under the control of gastrin. An acute gastrin stimulus causes exocytosis of the cytoplasmic granules/vesicles (and release of histamine and activation of the histamine-forming enzyme, histidine decarboxylase), while a more sustained gastrin stimulus causes first hypertrophy and then hyperplasia of the ECL cells in the rat (at most, a fivefold increase in the cell number). These effects can be demonstrated following infusion of gastrin or following an increase in the concentration of circulating gastrin of endogenous origin. The growth of the ECL cells reflects an accelerated self-replication rate. As studied in the rat, the self-replication rate is accelerated quite soon after induction of hypergastrinemia (blockade of acid secretion), the rate is maximally elevated within two weeks and then declines to control values at ten and 20 weeks despite the sustained hypergastrinemia. Lifelong hypergastrinemia in rats is associated not only with ECL-cell hyperplasia but also with an increased incidence of ECL-cell carcinoids. Recently, we could show that alpha-fluoromethylhistidine, which is a suicide inhibitor of histidine decarboxylase, effectively depletes the ECL cells of histamine and that the histamine-depleted ECL cells respond to gastrin with hyperplasia in a manner identical to normal ECL cells. Other factors beside gastrin seem to participate in the control of ECL-cell function and proliferation. Although exogenous somatostatin is known to suppress the activity of the ECL cells, we have failed to obtain evidence that the somatostatin cells in the oxyntic mucosa play a role in the physiological control of the ECL cells. The vagus, however, is important for the ability of the ECL cells to respond to gastrin. This conclusion is based on the observation that vagal denervation suppresses the hyperplastic response of the ECL cells to gastrin. Porta-cava shunting, on the other hand, greatly enhances the responsiveness of the ECL cells to gastrin. The mechanism behind this effect is unknown.  相似文献

The effects of bicarbonate and oxygen on photoinhibition of thylakoid membranes were investigated by varying their concentrations independently of each other. A pretreatment of the thylakoid suspension which lowered the bicarbonate concentration of the medium without affecting its oxygen content, increased the degree of photoinhibition upon illumination. This showed that the normal bicarbonate content of a thylakoid suspension, originating from dissolved carbondioxide from the air, protects against photoinhibition. The resistance against photoinhibition was further increased by addition of extra NaHCO3 up to about 5 mM. The normal oxygen content can be decreased profoundly without affecting the degree of photoinhibition; in contrast, even small changes from the normal bicarbonate content affected photoinhibition.At oxygen concentrations approximately below 25 M, added NaHCO3 not only did not protect, but caused a more severe PS 2 inactivation. Variation in movement across time and space fundamentally shapes the abundance and distribution of populations. Although a variety of approaches model structured population dynamics, they are limited to specific types of spatially structured populations and lack a unifying framework. Here, we propose a unified network‐based framework sufficiently novel in its flexibility to capture a wide variety of spatiotemporal processes including metapopulations and a range of migratory patterns. It can accommodate different kinds of age structures, forms of population growth, dispersal, nomadism and migration, and alternative life‐history strategies. Our objective was to link three general elements common to all spatially structured populations (space, time and movement) under a single mathematical framework. To do this, we adopt a network modeling approach. The spatial structure of a population is represented by a weighted and directed network. Each node and each edge has a set of attributes which vary through time. The dynamics of our network‐based population is modeled with discrete time steps. Using both theoretical and real‐world examples, we show how common elements recur across species with disparate movement strategies and how they can be combined under a unified mathematical framework. We illustrate how metapopulations, various migratory patterns, and nomadism can be represented with this modeling approach. We also apply our network‐based framework to four organisms spanning a wide range of life histories, movement patterns, and carrying capacities. General computer code to implement our framework is provided, which can be applied to almost any spatially structured population. This framework contributes to our theoretical understanding of population dynamics and has practical management applications, including understanding the impact of perturbations on population size, distribution, and movement patterns. By working within a common framework, there is less chance that comparative analyses are colored by model details rather than general principles.  相似文献

Barley (Hordeum vulgare L. cv. Golf) plants were grown at twodifferent relative addition rates; 0.1 and 0.2 d–1 ofnitrate. Three to five days before measurements started theplants were transferred to a nutrient solution with 2 mM nitrateor ammonium. The ammonium-grown plants showed increased ammoniumlevels in both shoots and roots and also increased ammoniumconcentrations in xylem sap. Ammonia emission measured in cuvettes connected to an automaticNH3 monitor was close to zero for nitrate-grown plants but increasedto 0.59 and 0.88 nmol NH3 m–2 S–1 for plants transferredto ammonium after growing at RA=0.2 and 0.1 d–1, respectively.In darkness, NH3 emission decreased together with photosynthesisand transpiration, but increased rapidly when the light wasturned on again. Addition of 0.5 mM methionine sulphoximine (MSO) to the plantscaused an almost complete inhibition of both root and shootglutamine synthetase (GS) activity after 24 h. Ammonia emissionincreased dramatically and photosynthesis and transpirationdecreased in both nitrate- and ammonium-grown plants as a resultof the GS inhibition. At the same time plant tissue and xylemsap ammonium concentrations increased, indicating the importanceof GS in controlling plant ammonium levels and thereby NH3 emissionfrom the leaves. Summary The conjugative transposon Tn916 was used for mutagenesis ofClostridium acetobutylicum ATCC 824. Tetracycline-resistant mutants were screened for loss of granulose synthesis and five classes of granulose mutants, that contained single transposon insertions, were identified on the basis of altered solvent production. Class 1 mutants did not make acetone or butanol, lacked activity of enzymes induced during solventogenesis, and did not sporulate, indicating that they are regulatory mutants. The class 2 mutant strains also did not produce acetone but did form small amounts of butanol and ethanol while the class 3 mutants produced low amounts of all solvents. Class 4 and 5 mutants produced essentially the same or higher amounts of solvents than the parent strain. Transposon insertions in the class 1 mutants were used as markers for in vitro synthesis of flanking chromosomal DNA using Tn916-specific primers. The DNA fragments were labeled to produce specific probes. Transposon insertion sites in the chromosomes of 13 different class 1 regulatory mutants were compared by hybridization of the specific probes to Southern blots of restriction endonuclease-digested parental chromosomal DNA. Insertions in two mutants appeared to be, in the same region of the chromosome. These results predict, that multiple regulatory elements are required to induce solvent production and sporulation.  相似文献
