Determination of surface immunoglobulin density on frozen-thawed mononuclear cells analyzed by the fluorescence-activated cell sorter

Recent data have demonstrated that differences in sIg density on B lymphocytes distinguish functionally distinct subpopulations of these cells. Other reports suggest that cyropreservation may change the frequency of sIg-bearing lymphocytes. To determine if cryopreservation alters either the frequency of sIg cells or the distribution of sIg density, PBM from normals and patients with CLL and LCL were analyzed using the FACS. Aliquots of Ficoll-Hypaque-separated PBM were controlled-rate frozen (1 °C/min) in 7.5% Me₂SO in RPMI 1640 and thawed in a 37 °C water bath on the same day. Fresh and frozen-thawed PBM aliquots were labeled with fluorescein conjugates of F(ab′) fragments of affinity chromatography-purified anti-Fab or class-specific anti-μ, anti-δ, anti-γ, or anti-α. Histograms of relative cell fluorescence, reflecting sIg density, were prepared for each aliquot with the FACS. The frequency of sIg-bearing PBM labeled with each reagent was not significantly altered by freezing. Likewise, FACS profiles demonstrated that the distribution of sIg density on normal and CLL PBM was unchanged after freezing. However, the fluorescence peak produced by frozen-thawed unlabeled cells was occasionally slightly broader than that of fresh cells, suggesting increased autofluorescence induced by freezing. These data indicate that frozen cell preparations may be utilized for the study of B-lymphocyte subsets as determined by sIg density.     

Intensification of alcoholic fermentation upon dehydration-rehydration of the yeast Saccharomyces cerevisiae

Summary In comparison with intact yeast, dehydrated-rehydrated cells of Saccharomyces cerevisiae show significantly higher ethanol production from exogenous substrate under both anaerobic and aerobic conditions, particularly when low concentration (0.1%) of glucose are used. For populations with a higher percentage of viable rehydrated cells (above 70%) a more notable decrease in the Pasteur effect (the difference between the quantity of ethanol formed under anaerobic and aerobic conditions) is observed.     

Effect of continuous application of manures and fertilizers on rhizosphere microflora in arecanut palm

Summary The rhizosphere microflora of arecanut palm under continuous application of organic manures and inorganic fertilizers was studied. The nutrients applied are 100 g N, 40 g P₂O₅ and 140 g K₂O/palm/year in the form of organics and inorganics. The application of organic manure increased the microbial population. The increase in microbial population was observed between the rhizosphere samples collected at 0–30cm and 30–60 cm depths. The surface cultivation of soil increased the microbial population.Trichoderma sp. andAspergillus sp. dominated in therhizosphere of arecanut palm. Contribution No. 208. Osteogenic precursor cells in repair osteogenesis

Histological and electron-microscopic radioautographic investigations of regenerating medullar tissue were performed in rabbits following curettage. Low differentiated connective tissue cells were shown to possess the highest proliferative activity. DNA synthesis mainly took place in low differentiated, endothelial and osteogenic cells. It is suggested that low differentiated cells take part in histogenesis of regenerating medullar tissue, including osteo- and angiogenesis.     

Effect of retinoic acid on the aggregation of erythrocytes and leukocytes in the blood

Human and animal blood smear staining with PAPh has revealed mononuclear leukocyte-erythrocyte aggregates. Administration of retinoic acid increased concentration and dimensions of these aggregates and was followed by preferential accumulation of PAPh-negative osmotically unstable erythrocytes. Similar changes were detected in the blood samples of women engaged in the production of retinoic acid. Aggregate concentration showed positive correlation with erythrocyte sedimentation rate and no correlation with prothrombin time.     

Fatty acid composition of Escherichia coli cells and their viability in air

Experiments on E. coli used as a model have revealed that fatty-acid composition is one of the characteristics which determine the viability of bacteria in the air. The viability of microbial cells in the air has been shown to increase with the increase of the pool of cyclopropane acids and the palmitic acid/palmitoleic acid ratio in the cells, irrespective of their genotype and the phase of their growth.
