Until recently, pansporoblastic microsporidia that produce a variable and large number of sporoblasts from a sporont have been included in a single genus, namely Pleistophora Gurley, 1893. Ultrastructural studies have been used to determine whether the resemblance of these species is fundamental or superficial. The results indicated that the multisporous pansporoblastic forms belong to at least three genera and, thus, that Pleistophora is a “composite genus.” The term pansporoblast was originally used for stages in myxosporidian development. The term sporophorous vesicle adopted from Gurley is suggested for the spore-containing vesicle in the Microspora. Three species were studied: Pleistophora typicalis, the type-species; Pleistophora culicis, for which a new genus Vavraia has already been proposed; and Pleistophora simulii. P. typicalis and V. culicis have isolated nuclei throughout their development, and the sporophorous vesicle wall enveloping the sporoblasts is derived from amorphous secretions laid down during merogony external to the plasmalemma. Pleistophora and Vavraia are differentiated principally in terms of the structure of the sporophorous vesicle wall and mode of division of the sporogonial plasmodium. The nuclei of young sporonts of P. simulii are in diplokaryon arrangement and undergo meiosis to give haploid nuclei in the sporoblasts. The sporophorous vesicle wall is membranoid and is laid down external to the plasmalemma at the onset of sporogony. A new genus, Polydispyrenia n. g., is suggested for this species, the affinities of which are closer to the dimorphic species of microsporidia than to Pleistophora or Vavraia. The terms “merontogenetic sporophorous vesicle” and “sporontogenetic sporophorous vesicle” are used to distinguish between the two groups. 相似文献

Adenosine deaminase from Bacillus cereus is quite unstable, similarly to other bacterial deaminases, but it shows a peculiar stabilizing effect by some monovalent cations. These include K+, Li+, NH4+ and to a lesser extent Cs+. Maximal stabilization of the deaminase is exerted by K+ at concentrations higher than 20 mM. The enzyme can be rapidly inactivated by sulphydryl reagents such as p-hydroxymercuribenzoate. Since adenosine deaminase from B. cereus, in addition to monovalent cations, is stabilized also by dithiothreitol, a possible influence of monovalent cations on the reactivity of some sulphydryl groups on the enzyme has been suggested. 相似文献

Internodes of Chara corallina were used for experiments in which cell turgor pressure was clamped by means of the pressure probe technique. Essentially, the procedure consisted of a combination of volume and turgor pressure relaxations. This technique permits the determination of the cell volume by nonoptical means. The values obtained are in agreement with the ones determined by optical means. Furthermore, the hydraulic conductivity (Lp) was determined from the initial slope of the volume relaxation; the values thus obtained are in agreement with those calculated from the half-times of pressure relaxations. The determination of Lp from volume relaxation measurements has the advantage that the cell volume, the volumetric elastic modulus of the cell wall, and the internal osmotic pressure do not have to be known. Furthermore, the half-time of volume relaxation is longer than that of pressure relaxation, as shown by theory and experiment. This may be used to enhance the resolution of the relaxation measurement and, thus, to improve the accuracy of Lp determinations for higher plant cells which exhibit a very fast pressure relaxation. 相似文献

The influence of various c oncentrations of K⁺, nitrogen sources, and inoculation with root-knot nematode Meloidogyne javanica were evaluated in tomato plants. Increased potassium concentration increased top and root fresh weights of intact plants and fresh weights of excised roots. Nitrate-fertilized plants weighed more than plants receiving ammonium independent of the K level in the medium. Nematode counts on roots were not affected by nutritional differences in intact or excised roots. In intact roots a high percentage of males was recorded at low K⁺ levels, whereas in excised roots the proportion of males in the population rose as the K⁺ levels increased. The three-dimensional structure of the sulfhydryl protease calotropin DI from the madar plant, Calotropis gigantea, has been determined at 3·2 Å resolution using the multiple isomorphous replacement method with five heavy atom derivatives. A Fourier synthesis based on protein phases with a mean figure of merit of 0·857 was used for model building. The polypeptide backbone of calotropin DI is folded to form two distinct lobes, one of which is comprised mainly of α-helices, while the other is characterized by a system of all antiparallel pleated sheets. The overall molecular architecture closely resembles those found in the sulfhydryl proteases papain and actinidin.Despite the unknown amino acid sequence of calotropin DI a number of residues around its active center could be identified. These amino acid side-chains were found in a similar arrangement as the corresponding ones in papain and actinidin. The polypeptide chain between residues 1 and 18 of calotropin DI folds in a unique manner, providing a possible explanation for the unusual inability of calotropin DI to hydrolyze those synthetic substrates that papain and actinidin act upon. 相似文献

Summary The current-voltage relationship of carrier-mediated, passive and active ion transport systems with one charge-carrying pathway can exactly be described by a simple reaction kinetic model. This model consists of two carrier states (one inside, one outside) and two pairs (forwards and backwards) of rate constants: a voltage-dependent one, describing the transport of charge and a voltage-insensitive one, summarizing all the other (voltage-independent) reactions. For the electrogenic Cl– pump inAcetabularia these four rate constants have been determined from electrical measurements of the current-voltage relationship of the pump (Gradmann, Hansen & Slayman, 1981;in: Electrogenic Ion Pumps, Academic Press, New York). The unidirectional Cl– efflux through the pump can also be calculated by the availiable reaction kinetic parameters.36Cl– efflux experiments on singleAcetabularia cells with simultaneous electrical stimulation (action potentials) and recording, demonstrate the unidirectional Cl– efflux to depend on the membrane potential. In isolated canine atrium, perfused with blood from a donor dog, the infusions of both prostaglandins (PG)I2 and E2 (0.1–1 μg/min) into the sinus node arterial cannula neither altered the sinus rate and developed tension nor the positive chronotropic and inotropic responses elicited by either electrical stimulation or by injected norepinephrine. Infusion of arachidonic acid (10–100 μg/min), a precursor of PGs, or indomethacin (15–20 μg/min), an inhibitor of PG synthesis, into the sinus node arterial cannula also failed to alter the increase in sinus rate or developed tension produced by either adrenergic stimulus in the isolated atria. When arachidonic acid, 100–300 μg/kg or PGI2, 1 μg/kg, were injected into the jugular vein of the donor dog, they produced a fall in systemic blood pressure; this effect of arachidonic acid but not of PGI2 was abolished by indomethacin, 1 mg/kg. During administration of either arachidonic acid or indomethacin to the donor dog, the positive chronotripic and inotropic responses to adrenergic stimuli in the isolated atria also remained unaltered.
