Fluorescence in situ hybridization was used to establish the order of, and to estimate genomic distances among, members of the carcinoembryonic antigen (CEA) and pregnancy-specific glycoprotein (PSG) subgroups on chromosome 19. Fluorescence in situ hybridization to metaphase chromosomes localized the PSG subgroup telomeric to the CEA subgroup. Cosmid clones containing sequences for individual genes in the CEA and PSG subgroups were also hybridized to human sperm pronuclear and somatic interphase nuclear chromatin targets. The mapping results lead to the gene order cen-CGM7-CEA-NCA-CGM1-BGP-CGM9-CGM8-PSG-te l. The genomic distances between selected pairs of gene family members were estimated from the physical distances between hybridization sites measured in pronuclei. The CEA-PSG gene family region is estimated to span 1.1 to 1.2 Mb.  相似文献

Although invertebrate drift is an important ecological process in lotic ecosystems, very little is known about it in Kenyan rivers. The primary aim of this study was to investigate the effect of driftnet mesh size and exposure duration on drift density in 2017. Drift samples were dominated by Chironomidae, Baetidae, Simuliidae, Caenidae and Culicidae. The 100 µm mesh driftnet had the highest mean invertebrate density, followed by the 250 µm and 500 µm nets. Invertebrate drift densities decreased with increased exposure time. This study demonstrates that sampler mesh size and exposure time should be taken into account when characterising invertebrate drift in streams. Future studies should consider sampling different biotopes and during different seasons.  相似文献

Rat hearts with coronary circulations perfused by the Langendorff technique were studied. Recordings were made of the electrical and mechanical activity. Once the rate and rhythm of each heart and stabilized it was perfused with Ringer-Locke solution for 90 minutes; frusemide (40 μg/ml) was added to the perfusate during the last 30 minutes of this period. Eighteen experiments were performed—six controls, six in which prolactin at a concentration of 50 ng/ml was added to the perfusate for the whole 90 minutes, and six in which a prolactin concentration of 200 ng/ml was used.With the controls heart rate and rhythm remained steady, but there was a slow decline in amplitude of the contraction. With a prolactin concentration of 50 ng/ml the heart rate rose to about 40% above control during the first hour and after an initial sharp increase the contraction amplitude declined more rapidly than in the controls. The prolactin concentration of 200 ng/ml produced a decline of about 25% in heart rate over the first hour and amplitude behaved much as in the controls. Frusemide had no clear effect on the rate of beating of the controls, but it tended to reverse both the acceleration produced by 50 ng/ml prolactin and the slowing produced by the higher dose. Both the doses of prolactin consistently caused disturbances of rhythm. These effects occur at concentration of prolactin found in human plasma in various pathophysiological conditions.  相似文献

Genetic code expansion in multicellular organisms is currently limited to the use of repurposed amber stop codons. Here, we introduce a system for the use of quadruplet codons to direct incorporation of non-canonical amino acids in vivo in an animal, the nematode worm Caenorhabditis elegans. We develop hybrid pyrrolysyl tRNA variants to incorporate non-canonical amino acids in response to the quadruplet codon UAGA. We demonstrate the efficiency of the quadruplet decoding system by incorporating photocaged amino acids into two proteins widely used as genetic tools. We use photocaged lysine to express photocaged Cre recombinase for the optical control of gene expression and photocaged cysteine to express photo-activatable caspase for light inducible cell ablation. Our approach will facilitate the routine adoption of quadruplet decoding for genetic code expansion in eukaryotic cells and multicellular organisms.  相似文献

Routine ultrasound examination of the fetus is already established in most obstetric units in Britain. A simple method was devised to evaluate one section of the fetal heart systematically. Examination of this section, the four chamber view, may readily be incorporated into routine obstetric screening. Severe cardiac abnormalities detectable in this view occur in two per 1000 pregnancies. For six years the department of paediatric cardiology at Guy''s Hospital, London, served as a referral centre for fetal echocardiography. As teaching became more widespread an increasing proportion of cases of cardiac anomaly were referred because the obstetrician suspected abnormality on examination of the four chamber view. Currently 80% of detected abnormalities are referred for this reason. Further extension and organisation of teaching might result in most severe cardiac malformations being detected in early prenatal life.  相似文献

Integrins undergo global conformational changes that specify their activation state. Current models portray the inactive receptor in a bent conformation that upon activation converts to a fully extended form in which the integrin subunit leg regions are separated to enable ligand binding and subsequent signaling. To test the applicability of this model in adherent cells, we used a fluorescent resonance energy transfer (FRET)–based approach, in combination with engineered integrin mutants and monoclonal antibody reporters, to image integrin α5β1 conformation. We find that restricting leg separation causes the integrin to adopt a bent conformation that is unable to respond to agonists and mediate cell spreading. By measuring FRET between labeled α5β1 and the cell membrane, we find extended receptors are enriched in focal adhesions compared with adjacent regions of the plasma membrane. These results demonstrate definitely that major quaternary rearrangements of β1-integrin subunits occur in adherent cells and that conversion from a bent to extended form takes place at focal adhesions.  相似文献

Summary Sand sheets near Darwin support a distinct heathland vegetation type which includes the habitat of several threatened species. Sand and gravel are extracted from shallow mines in this region. Woody vegetation recovery in 31 small, shallow former sand or gravel mine sites near Darwin that were up to 27 years old was assessed and compared to paired unmined control sites. Recovery in vegetation structure within each mine was calculated as the percentage of that in the control site. Mined sites recovered about 50% of their stem count and canopy cover, but only about 10% of basal area and mature tree count. Gravel mines showed poorer recovery than sand mines. Time since mining had no significant effect on the extent of recovery, but deeper mines had significantly poorer recovery. Only 35% of woody species in sand controls were present in mine sites, and 41% of gravel control species were present in former mine sites. It is unlikely that recovery will significantly improve in coming decades. Sand mining affects about 40 ha of land per year in this region, but is likely to increase in the future. If Darwin expands to a population of 1 million people, and mine sites are not fully rehabilitated, all of the sand‐sheet vegetation in the region could be removed in the next 100 years. Improved rehabilitation and protection is crucial for the conservation of heathland vegetation in this region.  相似文献

The ability of epidermal growth factor receptor (EGFR) to control cell fate is defined by its affinity for ligand. Current models suggest that ligand-binding heterogeneity arises from negative cooperativity in signaling receptor dimers, for which the asymmetry of the extracellular region of the Drosophila EGFR has recently provided a structural basis. However, no asymmetry is apparent in the isolated extracellular region of the human EGFR. Human EGFR also differs from the Drosophila EGFR in that negative cooperativity is found only in full-length receptors in cells. To gain structural insights into the human EGFR in situ, we developed an approach based on quantitative Förster resonance energy transfer (FRET) imaging, combined with Monte Carlo and molecular dynamics simulations, to probe receptor conformation in epithelial cells. We experimentally demonstrate a high-affinity ligand-binding human EGFR conformation consistent with the extracellular region aligned flat on the plasma membrane. We explored the relevance of this conformation to ligand-binding heterogeneity and found that the asymmetry of this structure shares key features with that of the Drosophila EGFR, suggesting that the structural basis for negative cooperativity is conserved from invertebrates to humans but that in human EGFR the extracellular region asymmetry requires interactions with the plasma membrane.  相似文献
