The cowpox virus (CPV) SPI-3 gene (open reading frame K2L in vaccinia virus) is one of three orthopoxvirus genes whose products are members of the serpin (serine proteinase inhibitor) superfamily. The CPV SPI-3 gene, when overexpressed by using the vaccinia virus/T7 expression system, synthesized two proteins of 50 and 48 kDa. Treatment with the N glycosylation inhibitor tunicamycin converted the two SPI-3 proteins to a single 40-kDa protein, close to the size of 42 kDa predicted from the DNA sequence, suggesting that the SPI-3 protein, unlike the other two orthopoxvirus serpins, is a glycoprotein. Immunoblotting with an anti-SPI-3 antibody showed that the SPI-3 protein is synthesized early in infection prior to DNA replication. SPI-3 inhibits cell-cell fusion during infections with both CPV and vaccinia virus. A transfection assay was devised to test engineered mutants of SPI-3 for the ability to inhibit fusion. Two mutants with C-terminal deletions of 156 and 70 amino acids were completely inactive in fusion inhibition. Site-directed mutations were constructed near the C terminus of SPI-3, in or near the predicted reactive-site loop which is conserved in inhibitory serpins. Substitutions within the loop at the P1 to P1' positions and P5 to P5' positions, inclusive, did not result in any loss of activity, nor did changes at the P17 to P10 residues in the stalk of the reactive loop. A unicellular cyanophycean culture contaminant had features of both Geitleribactron Kom. and Cyanophanon Geitl. The cells were elongated, sheathless, mostly similar in diameter throughout their length, and attached polarly in rosettes or groups and produced only a single elongated exocyte (=exospore). Young cells were moderately elongated and resembled Geitleribactron. As cells aged, they greatly elongated and then resembled Cyanophanon. Some cells formed Y-shaped bifurcations, features of C. mirabile Geitl. and C. minus Geitl., but they lacked the basal sheath (pseudovagina) of C. mirabile. During exocyte formation, a thick and localized L-II wall layer protuberance extended the exocyte away from the parent cell. This terminal wall thickening then appeared to move to one side from subsequent and unequal cell wall growth. Cells sovnetimes bent abruptly, occasionally opposite a thickening in the L-II wall layer. Further studies in culture of putative Geitleribactron and Cyanophanon isolates are necessary to ascertain the breadth of their structural diversity and the identity of the present taxon. 相似文献

A five year study was conducted in a 100–120 year old even-aged sugar maple stand in southern Quebec (46°07N 73° 56W; 305 m altitude) to explore the effect of different fertilization formulations aimed at 1) correcting the most common nutrient deficiencies observed in declining maple stands (K and Mg), 2) decreasing soil acidity, and 3) simulating enrichment with atmospheric N. Seven fertilizer mixtures were applied in the spring of 1987: 400 kg ha-1 of K2SO4, CaCO3, CaMg(CO3)2, (NH4)2SO4, complete fertilizer (Maplegro) and 800 kg ha-1 of an equal mixture of K2SO4+CaCO3 or K2SO4+CaMg(CO3)2. The site was divided into twenty-four 25×25 m plots and treatments including control were replicated three times. Leaves and soils (organic and mineral) were sampled in 1987, 1988 and 1991. Trees were cored at 1.2 m to measure their response in diameter growth. The application of K2SO4+CaMg(CO3)2 was the only treatment that significantly increased (+13%) the average growth rate over the five year period after fertilization. The application of (NH4)2SO4, Maplegro, CaMg(CO3)2 and K2SO4 reduced growth relative to the control for the five year period by 29, 24, 20 and 12 %, respectively. Positive and negative effects on growth can be explained mainly in terms of changes in leaf K. Both the application of Maplegro and (NH4)2SO4 increased soil P availability. Overall, the rate of growth showed a cubic pattern of change over the 5 year period with peaks in 1988 and 1991. Trees in control plots went from a limiting foliar status of Ca and Mg, and surplus N in 1987 to a surplus of Ca and Mg, and lower N concentration in 1991. Our results suggest that nutrient deficiencies observed at our site were associated with a disturbance of the biogeochemical cycle of nutrients rather than soil nutrient depletion.Abbreviations BS

The design, construction, and characterization of a prototype-regenerable glucose biosensor based on the reversible immobilization of glucose oxidase (GOx) using cellulose binding domain (CBD) technology is described. GOx, chemically linked to CBD, is immobilized by binding to a cellulose matrix on the sensor-indicating electode. Enzyme immobilization can be reversed by perfusing the cellulose matrix with a suitable eluting solution. An autocavable sensor membrane system is employed which is shown to be practical for use in real microbial fermentations. The prototype glucose biosensor was used without failure or deterioration during fed-batch fermentations of Escherichia coli reaching a maximum cell density of 85 g (dry weight)/L. Medium glucose concentration based on sensor output correlated closely with off-line glucose analysis and was controlled manually at 0.44 +/- 0.2 g/L for 2 h based on glucose sensor output. The sensor enzyme component could be eluted and replaced without interrupting the fermentation. Mycobacteria have the ability to persist within host phagocytes, and their success as intracellular pathogens is thought to be related to the ability to modify their intracellular environment. After entry into phagocytes, mycobacteria-containing phagosomes acquire markers for the endosomal pathway, but do not fuse with lysosomes. The molecular machinery that is involved in the entry and survival of mycobacteria in host cells is poorly characterized. Here we describe the use of organelle electrophoresis to study the uptake of Mycobacterium bovis bacille Calmette Guerin (BCG) into murine macrophages. We demonstrate that live, but not dead, mycobacteria occupy a phagosome that can be physically separated from endosomal/lysosomal compartments. Biochemical analysis of purified mycobacterial phagosomes revealed the absence of endosomal/lysosomal markers LAMP-1 and β-hexosaminidase. Combining subcellular fractionation with two-dimensional gel electrophoresis, we found that a set of host proteins was present in phagosomes that were absent from endosomal/lysosomal compartments. The residence of mycobacteria in compartments outside the endosomal/lysosomal system may explain their persistence inside host cells and their sequestration from immune recognition. Furthermore, the approach described here may contribute to an improved understanding of the molecular mechanisms that determine the intracellular fate of mycobacteria during infection. 相似文献

Extraction of Lactobacillus fermentum BR11 cells with 5 M LiCl yielded a preparation containing a single predominant polypeptide with an apparent molecular mass of 32 kDa. A clone encoding an immunoreactive 32-kDa polypeptide was isolated from a pUC18 library of L. fermentum BR11 DNA by screening with an antiserum raised against whole cells of L. fermentum BR11. Sequence determination of the insert in the clone revealed a complete 795-bp open reading frame (ORF) that defines a 28,625-Da polypeptide (BspA). N-terminal sequencing of the LiCl-extracted polypeptide from L. fermentum BR11 confirmed that it is the same as the cloned BspA. BspA was found to have a sequence similar to those of family III of the bacterial solute-binding proteins. The sequences of two ORFs upstream of bspA are consistent with bspA being located in an operon encoding an ATP-binding cassette-type uptake system. Unusually, BspA contains no lipoprotein cleavage and attachment motif (LXXC), despite its origin in a gram-positive bacterium. Biotin labelling and trypsin digestion of whole cells indicated that this polypeptide is exposed on the cell surface. The isoelectric point as predicted from the putative mature sequence is 10.59. It was consequently hypothesized that the positively charged BspA is anchored by electrostatic interaction with acidic groups on the cell surface. It was shown that BspA could be selectively removed from the surface by extraction with an acidic buffer, thus supporting this hypothesis. 相似文献

A computer-assisted analysis was made of 24 complete nucleotide sequences selected from the vertebrate retroviruses to represent
the ten viral groups. The conclusions of this analysis extend and strengthen the previously made hypothesis on the Moloney
murine leukemia virus: The evolution of the nucleotide sequence appears to have occurred mainly through at least three overlapping
levels of duplication: (1) The distributions of overrepresented (3–6)-mers are consistent with the universal rule of a trend
toward TG/CT excess and with the persistence of a certain degree of symmetry between the two strands of DNA. This suggests
one or several original tandemly repeated sequences and some inverted duplications. (2) The existence of two general core
consensuses at the level of these (3–6)-mers supports the hypothesis of a common evolutionary origin of vertebrate retroviruses.
Consensuses more specific to certain sequences are compatible with phylogenetic trees established independently. The consensuses
could correspond to intermediary evolutionary stages. (3) Most of the (3–6)-mers with a significantly higher than average
frequency appear to be internally repeated (with monomeric or oligomeric internal iterations) and seem to be at least partly
the cause of the bias observed by other researchers at the level of retroviral nucleotide composition. They suggest a third
evolutionary stage by slippage-like stepwise local duplications.
