Because recent patterns of permafrost collapse in boreal peatlands appear to enhance emissions of CH 4 to the atmosphere, we examined methanogenesis and methanogen diversity in peat soil from peatlands with and without permafrost in two peatland complexes situated in continental western Canada. Peat soil from the active layer of permafrost bogs had very low rates of CH 4 production (ca. 10 nmol g ?1 day ?1 ), and we were unable to PCR-amplify 16s rRNA gene sequences using Archaea-specific primers in four peat samples. Surface peat soil from continental bogs with no permafrost supported moderate rates of CH 4 production (20–600 nmol g ?1 day ?1 ), with maximum rates in soil located close to the mean water table level. Additions of ethanol stimulated CH 4 production rates, suggesting metabolic substrate limitations. Peat from internal lawns, which have experienced surface permafrost degradation in the past 150 years, had very rapid rates of CH4 production (up to 800 nmol g ?1 day ?1 ) occurring within the soil profile. Concomitant rates of anaerobic CO 2 production were greater in continental bogs (ca. 6 μmol g ?1 day ?1 ) than in internal lawns (ca. 4 μ mol g ?1 day ?1 ) or in permafrost bogs (2.8 μ mol g ?1 day ?1 ). Analysis of the 16s rRNA gene for Archaea in the continental bog indicated mostly sequences associate with Methanobacteriales and RC-I with a Methanosarcinaceae sequence in the deepest peat soil. In the internal lawn, Methanosarcinaceae were most common in peat soil with a Methanosaetaceae sequence in the deepest peat soil. This study showed that patterns of discontinuous permafrost and ongoing permafrost degradation in boreal regions create patchy soil environments for methanogens and rates of methanogenesis. 相似文献

Background and methods
Human metapneumovirus (hMPV) is a recently discovered respiratory virus associated with bronchiolitis, pneumonia, croup and exacerbations of asthma. Since respiratory viruses are frequently detected in patients with acute exacerbations of COPD (AE-COPD) it was our aim to investigate the frequency of hMPV detection in a prospective cohort of hospitalized patients with AE-COPD compared to patients with stable COPD and to smokers without by means of quantitative real-time RT-PCR.Results
We analysed nasal lavage and induced sputum of 130 patients with AE-COPD, 65 patients with stable COPD and 34 smokers without COPD. HMPV was detected in 3/130 (2.3%) AE-COPD patients with a mean of 6.5 × 105 viral copies/ml in nasal lavage and 1.88 × 105 viral copies/ml in induced sputum. It was not found in patients with stable COPD or smokers without COPD.Conclusion
HMPV is only found in a very small number of patients with AE-COPD. However it should be considered as a further possible viral trigger of AE-COPD because asymptomatic carriage is unlikely. 相似文献33.

The postnatal appearance and up-regulation of the NR2A subunit of the N-methyl-d-aspartate receptor contributes to the functional heterogeneity of the receptor during development. To elucidate the molecular mechanisms that regulate the neural and developmental specific expression of NR2A, an upstream approximately 9-kb region of the gene harboring the promoter was isolated and characterized in transgenic mice and transfected cortical neurons. Transgenic mouse lines generated with luciferase reporter constructs driven by either 9 or 1 kb of upstream sequence selectively transcribe the transgene in brain, as compared with other non-neural tissues. Reporter luciferase levels in dissociated cultures made from these mice are over 100-fold greater in neuronal/glial co-cultures than in pure glial cultures. Analysis of NR2A 5'-nested deletions in transfected cultures of cortical neurons and glia indicate that while sequences residing upstream of -1079 bp augment NR2A neuronal expression, sequences between -486 and -447 bp are sufficient to maintain neuronal preference. An RE1/NRSE element is not necessary for NR2A neuron specificity. Furthermore, comparison of the 5'-deletion constructs in cortical neurons grown for 5, 8, 11, or 14 days in vitro indicate that sequences between -1253 and -1180 bp are necessary for maturational up-regulation of NR2A. Thus, different cis-acting sequences control the regional and temporal expression of NR2A, implicating distinct regulatory pathways. 相似文献

Wildfire is the dominant disturbance in boreal forests and fire activity is increasing in these regions. Soil fungal communities are important for plant growth and nutrient cycling postfire but there is little understanding of how fires impact fungal communities across landscapes, fire severity gradients, and stand types in boreal forests. Understanding relationships between fungal community composition, particularly mycorrhizas, and understory plant composition is therefore important in predicting how future fire regimes may affect vegetation. We used an extreme wildfire event in boreal forests of Canada's Northwest Territories to test drivers of fungal communities and assess relationships with plant communities. We sampled soils from 39 plots 1 year after fire and 8 unburned plots. High‐throughput sequencing (MiSeq, ITS) revealed 2,034 fungal operational taxonomic units. We found soil pH and fire severity (proportion soil organic layer combusted), and interactions between these drivers were important for fungal community structure (composition, richness, diversity, functional groups). Where fire severity was low, samples with low pH had higher total fungal, mycorrhizal, and saprotroph richness compared to where severity was high. Increased fire severity caused declines in richness of total fungi, mycorrhizas, and saprotrophs, and declines in diversity of total fungi and mycorrhizas. The importance of stand age (a surrogate for fire return interval) for fungal composition suggests we could detect long‐term successional patterns even after fire. Mycorrhizal and plant community composition, richness, and diversity were weakly but significantly correlated. These weak relationships and the distribution of fungi across plots suggest that the underlying driver of fungal community structure is pH, which is modified by fire severity. This study shows the importance of edaphic factors in determining fungal community structure at large scales, but suggests these patterns are mediated by interactions between fire and forest stand composition. 相似文献

High‐latitude ecosystems store approximately 1700 Pg of soil carbon (C), which is twice as much C as is currently contained in the atmosphere. Permafrost thaw and subsequent microbial decomposition of permafrost organic matter could add large amounts of C to the atmosphere, thereby influencing the global C cycle. The rates at which C is being released from the permafrost zone at different soil depths and across different physiographic regions are poorly understood but crucial in understanding future changes in permafrost C storage with climate change. We assessed the inherent decomposability of C from the permafrost zone by assembling a database of long‐term (>1 year) aerobic soil incubations from 121 individual samples from 23 high‐latitude ecosystems located across the northern circumpolar permafrost zone. Using a three‐pool (i.e., fast, slow and passive) decomposition model, we estimated pool sizes for C fractions with different turnover times and their inherent decomposition rates using a reference temperature of 5 °C. Fast cycling C accounted for less than 5% of all C in both organic and mineral soils whereas the pool size of slow cycling C increased with C : N. Turnover time at 5 °C of fast cycling C typically was below 1 year, between 5 and 15 years for slow turning over C, and more than 500 years for passive C. We project that between 20 and 90% of the organic C could potentially be mineralized to CO2 within 50 incubation years at a constant temperature of 5 °C, with vulnerability to loss increasing in soils with higher C : N. These results demonstrate the variation in the vulnerability of C stored in permafrost soils based on inherent differences in organic matter decomposability, and point toward C : N as an index of decomposability that has the potential to be used to scale permafrost C loss across landscapes. 相似文献

rapid climate warming. We monitored growing season carbon dioxide (CO2) fluxes across a factorial design of in situ water table (control, drought, and flooded plots) and soil warming (control
vs. warming via open top chambers) treatments for 2 years in a rich fen located just outside the Bonanza Creek Experimental
Forest in interior Alaska. The drought (lowered water table position) treatment was a weak sink or small source of atmospheric
CO2 compared to the moderate atmospheric CO2 sink at our control. This change in net ecosystem exchange was due to lower gross primary production and light-saturated
photosynthesis rather than increased ecosystem respiration. The flooded (raised water table position) treatment was a greater
CO2 sink in 2006 due largely to increased early season gross primary production and higher light-saturated photosynthesis. Although
flooding did not have substantial effects on rates of ecosystem respiration, this water table treatment had lower maximum
respiration rates and a higher temperature sensitivity of ecosystem respiration than the control plot. Surface soil warming
increased both ecosystem respiration and gross primary production by approximately 16% compared to control (ambient temperature)
plots, with no net effect on net ecosystem exchange. Results from this rich fen manipulation suggest that fast responses to
drought will include reduced ecosystem C storage driven by plant stress, whereas inundation will increase ecosystem C storage

Climate change is altering disturbance regimes outside historical norms, which can impact biodiversity by selecting for plants with particular traits. The relative impact of disturbance characteristics on plant traits and community structure may be mediated by environmental gradients. We aimed to understand how wildfire impacted understory plant communities and plant regeneration strategies along gradients of environmental conditions and wildfire characteristics in boreal forests. We established 207 plots (60 m2) in recently burned stands and 133 plots in mature stands with no recent fire history in comparable gradients of stand type, site moisture (drainage) and soil organic layer (SOL) depth in two ecozones in Canada's Northwest Territories. At each plot, we recorded all vascular plant taxa in the understory and measured the regeneration strategy (seeder, resprouter, survivor) in burned plots, along with seedbed conditions (mineral soil and bryophyte cover). Dispersal, longevity and growth form traits were determined for each taxon. Fire characteristics measured included proportion of pre-fire SOL combusted (fire severity), date of burn (fire seasonality) and pre-fire stand age (time following fire). Results showed understory community composition was altered by fire. However, burned and mature stands had similar plant communities in wet sites with deep SOL. In the burned plots, regeneration strategies were determined by fire severity, drainage and pre- and post-fire SOL depth. Resprouters were more common in wet sites with deeper SOL and lower fire severity, while seeders were associated with drier sites with thinner SOL and greater fire severity. This led to drier burned stands being compositionally different from their mature counterparts and seedbed conditions were important. Our study highlights the importance of environment–wildfire interactions in shaping plant regeneration strategies and patterns of understory plant community structure across landscapes, and the overriding importance of SOL depth and site drainage in mediating fire severity, plant regeneration and community structure. 相似文献

Stomatin is a poorly understood integral membrane protein that is absent from the erythrocyte membranes of many patients with hereditary stomatocytosis. This report describes the cloning of the murine stomatin chromosomal gene, determination of its genomic structure, and characterization of the 5′-flanking genomic DNA sequences. The stomatin gene is encoded by seven exons spread over ∼25 kb of genomic DNA. There is no concordance between the exon structure of the stomatin gene and the locations of three domains predicted on the basis of protein structure. Inspection of the 5′-flanking DNA sequences reveals features of a TATA-less housekeeping gene promoter and consensus sequences for a number of potential DNA-binding proteins. 相似文献

Permafrost thaw can alter the soil environment through changes in soil moisture, frequently resulting in soil saturation, a shift to anaerobic decomposition, and changes in the plant community. These changes, along with thawing of previously frozen organic material, can alter the form and magnitude of greenhouse gas production from permafrost ecosystems. We synthesized existing methane (CH4) and carbon dioxide (CO2) production measurements from anaerobic incubations of boreal and tundra soils from the geographic permafrost region to evaluate large‐scale controls of anaerobic CO2 and CH4 production and compare the relative importance of landscape‐level factors (e.g., vegetation type and landscape position), soil properties (e.g., pH, depth, and soil type), and soil environmental conditions (e.g., temperature and relative water table position). We found fivefold higher maximum CH4 production per gram soil carbon from organic soils than mineral soils. Maximum CH4 production from soils in the active layer (ground that thaws and refreezes annually) was nearly four times that of permafrost per gram soil carbon, and CH4 production per gram soil carbon was two times greater from sites without permafrost than sites with permafrost. Maximum CH4 and median anaerobic CO2 production decreased with depth, while CO2:CH4 production increased with depth. Maximum CH4 production was highest in soils with herbaceous vegetation and soils that were either consistently or periodically inundated. This synthesis identifies the need to consider biome, landscape position, and vascular/moss vegetation types when modeling CH4 production in permafrost ecosystems and suggests the need for longer‐term anaerobic incubations to fully capture CH4 dynamics. Our results demonstrate that as climate warms in arctic and boreal regions, rates of anaerobic CO2 and CH4 production will increase, not only as a result of increased temperature, but also from shifts in vegetation and increased ground saturation that will accompany permafrost thaw. 相似文献
