Oryza sativa L. (domesticated rice) is a monocotyledonous plant, and its 430 Mb genome has been targeted for complete sequencing. We performed a high-resolution computer-based survey for transposable elements on 910 Kb of rice genomic DNA sequences. Both class I and II transposable elements were present, contributing 19.9% of the sequences surveyed. Class II elements greatly outnumbered class I elements (166 versus 22), although class I elements made up a greater percentage (12.2% versus 6.6%) of nucleotides surveyed. Several Mutator-like elements (MULEs) were identified, including rice elements that harbor truncated host cellular genes. MITEs (miniature inverted-repeat transposable elements) account for 71.6% of the mined transposable elements and are clearly the predominant type of transposable element in the sequences examined. Moreover, a putative Stowaway transposase has been identified based on shared sequence similarity with the mined MITEs and previously identified plant mariner-like elements (MLEs). Members of a group of novel rice elements resembling the structurally unusual members of the Basho family in Arabidopsis suggest a wide distribution of these transposons among plants. Progesterone injection in estradiol-primed, ovariectomized guinea pigs results in down-regulation of hypothalamic progestin receptors determined by in vitro binding assays. In order to determine if progesterone also decreases immunostaining of progestin receptors and if progestin receptors are down-regulated preferentially in particular neuroanatomical areas, ovariectomized guinea pigs were injected with doses of estradiol benzoate (10 micrograms at 42 h before progesterone injection) and progesterone (500 micrograms at 4, 12, or 24 h before perfusion) that reliably induce the expression of lordosis and subsequent behavioral refractoriness to progesterone. Progestin receptor-immunoreactive cells were counted in sections from discrete parts of the preoptic area and hypothalamus. As expected, estradiol dramatically increased cell nuclear, and, to a lesser extent, cytoplasmic, immunostaining in defined regions of the preoptic area and hypothalamus. By 12 h after progesterone injection, the number of progestin receptor-immunoreactive cells had decreased in some areas, but not others. The rostral and caudal aspects of the ventrolateral hypothalamus were particularly responsive showing a substantial decrease in progestin receptor-immunoreactivity by 12 h after injection. No decreases in the progestin receptor-immunoreactive cell number were observed in any of the preoptic regions examined, although obvious decreases in immunostaining intensity were seen. The results of these immunocytochemical experiments extend earlier findings from in vitro progestin binding experiments and demonstrate that as with progestin binding, progestin receptor-immunoreactivity decreases when progesterone is injected in a behavioral desensitization procedure. Molecular evolution of mitochondrial 12S RNA and cytochrome b sequences in the pantherine lineage of Felidae 总被引:4,自引:2,他引:2
DNA sequence comparisons of two mitochondrial DNA genes were used to infer
phylogenetic relationships among 17 Felidae species, notably 15 in the
previously described pantherine lineage. The polymerase chain reaction
(PCR) was used to generate sequences of 358 base pairs of the mitochondrial
12S RNA gene and 289 base pairs of the cytochrome b protein coding gene.
DNA sequences were compared within and between 17 felid and five nonfelid
carnivore species. Evolutionary trees were constructed using phenetic,
cladistic, and maximum likelihood algorithms. The combined results
suggested several phylogenetic relationships including (1) the recognition
of a recently evolved monophyletic genus Panthera consisting of Panthera
leo, P. pardus, P. onca, P. uncia, P. tigris, and Neofelis nebulosa; (2)
the recent common ancestry of Acinonyx jubatus, the African cheetah, and
Puma concolor, the American puma; and (3) two golden cat species, Profelis
temmincki and Profelis aurata, are not sister species, and the latter is
strongly associated with Caracal caracal. These data add to the growing
database of vertebrate mtDNA sequences and, given the relatively recent
divergence among the felids represented here (1-10 Myr), allow 12S and
cytochrome b sequence evolution to be addressed over a time scale different
from those addressed in most work on vertebrate mtDNA.

in highly trained endurance athletes could be related to differences in the pulmonary diffusing capacity (D

Sea urchin Hox genes: insights into the ancestral Hox cluster 总被引:3,自引:0,他引:3
We describe the Hox cluster in the radially symmetric sea urchin and
compare our findings to what is known from clusters in bilaterally
symmetric animals. Several Hox genes from the direct-developing sea urchin
Heliocidaris erythrogramma are described. CHEF gel analysis shows that the
Hox genes are clustered on a < or = 300 kilobase (kb) fragment of DNA,
and only a single cluster is present, as in lower chordates and other
nonvertebrate metazoans. Phylogenetic analyses of sea urchin, amphioxus,
Drosophila, and selected vertebrate Hox genes confirm that the H.
erythrogramma genes, and others previously cloned from other sea urchins,
belong to anterior, central, and posterior groups. Despite their radial
body plan and lack of cephalization, echinoderms retain at least one of the
anterior group Hox genes, an orthologue of Hox3. The structure of the
echinoderm Hox cluster suggests that the ancestral deuterostome had a Hox
cluster more similar to the current chordate cluster than was expected Sea
urchins have at least three Abd-B type genes, suggesting that Abd-B
expansion began before the radiation of deuterostomes.
