Metabolically stable polyamine derivatives may serve as useful surrogates for the natural polyamines in studies aimed to elucidate the functions of individual polyamines. Here we studied the metabolic stability of alpha-methylspermidine, alpha-methylspermine, and bis-alpha-methylspermine, which all have been reported to fulfill many of the putative physiological functions of the natural polyamines. In vivo studies were performed with the transgenic rats overexpressing spermidine/spermine N(1)-acetyltransferase. alpha-Methylspermidine effectively accumulated in the liver and did not appear to undergo any further metabolism. On the other hand, alpha-methylspermine was readily converted to alpha-methylspermidine and spermidine; similarly, bis-alpha-methylspermine was converted to alpha-methylspermidine to some extent, both conversions being inhibited by the polyamine oxidase inhibitor N(1), N(2)-bis(2,3-butadienyl)-1,4-butanediamine. Furthermore, we used recombinant polyamine oxidase, spermidine/spermine N(1)-acetyltransferase, and the recently discovered spermine oxidase in the kinetic studies. In vitro studies confirmed that methylation did not protect spermine analogs from degradation, whereas the spermidine analog was stable. Both alpha-methylspermidine and bis-alpha-methylspermine overcame the proliferative block of early liver regeneration in transgenic rats and reversed the cytostasis induced by an inhibition of ornithine decarboxylase in cultured fetal fibroblasts.  相似文献

Carabid beetles (Coleoptera, Carabidae) were collected by using pitfall traps in 68 sites on the Åland Islands, SW Finland The sites were divided into four environmental types 1) cultivated fields and grazed pastures. 2) recently abandoned fields, 3) abandoned, overgrown cultivations, and 4) forests A total of 4901 carabids belonging to 77 species were caught Twenty of the most abundant species were classified into four distributional types according to their occurrence among the various environmental types 1) generalists (five species), 2) field species (five species), 3) species of open habitat (seven species), and 4) forest species (three species) Associations with environmental types were strictest among field species and forest species However, most of the species were found in low numbers outside each preferred environmental type The number of species and species diversity was highest in fields and pastures and lowest in forests, which agrees with previously detected patterns on the mainland Finland Pairwise similarity of the carabid assemblages among the environmental types was highest between the two types of abandoned cultivations The spatial distribution of two carabid species, Pterostichus melanarius and P niger , were examined in detail to ascertain whether any interspecific interaction could help explain their different island-mainland distribution observed in previous studies  相似文献

Summary The correlations between certain life-history parameters (reproductive effort, reproductive life-span, age of first reproduction, general growth index, variation in juvenile survival, availability of resources) were studied in 13 populations of the mussel Anodonta piscinalis in south-western Finland in 1975 and 1976Reproductive life-span correlated positively (r s=0,823, P<0.001) with variation in juvenile survival. The average availability of resources correlated negatively both with the reproductive life-span (r s=-0.841, P<0.001 after the variation by juvenile survival had been deleted) and variation in juvenile survival (r s=-0.676, P<0.05).The reproductive effort for female mussels at each site was computed by comparing body weight of reproductive females with body weights of non-reproductive individuals. Availability of resources was much higher in 1976 than in 1975. Consequently, the reproductive effort, an index of the strain of reproduction, was higher in 1975 than in 1976. In 1975 there was a significant correlation between reproductive effort and the length of the reproductive life-span (-0.727, -0.806) and also with the reproductive effort and the variation in juvenile survival (-0.718, -0.758) in females of the length of 60 mm and 70 mm respectively. In 1976, when availability of resources was better, such correlations were not found.Spatial and temporal change in the intake of resources complicates applicability of the principle of resource allocation in the theory of life-history evolution. Studying the mere allocation is not enough if the intake of resources varies in the groups studied. The ratio ovary weight/body weight is a dubious measure of reproductive effort in comparative studies when the input of resources can vary, and this possibility can be ruled out only exceptionally.Correlation between growth and reproductive effort was positive, obviously because both are important components in creating high reproductive capacity. In 1975 reproductive effort increased with size (age). Change in reproductive effort correlated with reproductive life-span (r s=-0.633, P<0.05).The following parameters occurred together: short reproductive life-span, low age of first reproduction, high reproductive effort, rapid growth and high clutch size. They were realized at sites where the availability of resources was good and variation in juvenile survival was low-i.e. the environment was stable. The results conflict with the prediction of the theory of r and K-selection.  相似文献

The effect of inorganic nutrients on iron oxidation by Thiobacillus ferrooxidans was studied using a concentration of 2.0 g Fe2+/liter. Besides ammonium-nitrogen and phosphorus, the essential nutrients were sulphate and magnesium, with the lowest unlimiting concentration being 2.0 g SO/liter and 2.0 mg Mg2+/liter for a suspension of about 108 cells/ml. Omitting nitrate, calcium, potassium, and chloride had no influence on the iron oxidation. Nitrate and chloride were inhibitory in high concentrations although the bacteria were able to adapt to their presence. Several organic compounds tested in 0.5% (w/v) concentrations inhibited iron oxidation. Complete inhibition up to 14 days was caused by fructose, lactose, meat extract, yeast, extract, peptone, and tryptone. T. ferrooxidans-strains were able to adapt to the presence of organic material even though they were not necessarily able to use the organic compounds for energy. Some general conclusions are made concerning the role of nutrients in leaching processes.  相似文献

Profound changes to the taxonomy and classification of polypores have occurred since the advent of molecular phylogenetics in the 1990s. The last major monograph of North American polypores was published by Gilbertson and Ryvarden in 1986–1987. In the intervening 30 years, new species, new combinations, and new records of polypores were reported from North America. As a result, an updated checklist of North American polypores is needed to reflect the polypore diversity in there. We recognize 492 species of polypores from 146 genera in North America. Of these, 232 species are unchanged from Gilbertson and Ryvarden’s monograph, and 175 species required name or authority changes. In addition, 40 new species and 45 new records published since that monograph are included in the checklist. Among the 492 species of polypores, 486 species from 143 genera belong to 11 orders, while six other species from three genera have uncertain taxonomic position at the order level. Three orders, viz. Polyporales, Hymenochaetales and Russulales, accommodate most of polypore species (93.7 %) and genera (88.8 %). We hope that this updated checklist will inspire future studies in the polypore mycota of North America and contribute to the diversity and systematics of polypores worldwide.  相似文献

Objective To examine the relation of midlife raised blood pressure and serum cholesterol concentrations to Alzheimer''s disease in later life.Design Prospective, population based study.SettingPopulations of Kuopio and Joensuu, eastern Finland.ParticipantsParticipants were derived from random, population based samples previously studied in a survey carried out in 1972, 1977, 1982, or 1987. After an average of 21 years'' follow up, a total of 1449 (73%) participants aged 65-79 took part in the re-examination in 1998.Results People with raised systolic blood pressure (⩾160 mm Hg) or high serum cholesterol concentration (⩾6.5 mmol/l) in midlife had a significantly higher risk of Alzheimer''s disease in later life, even after adjustment for age, body mass index, education, vascular events, smoking status, and alcohol consumption, than those with normal systolic blood pressure (odds ratio 2.3, 95% confidence interval 1.0 to 5.5) or serum cholesterol (odds ratio 2.1, 1.0 to 4.4). Participants with both of these risk factors in midlife had a significantly higher risk of developing Alzheimer''s disease than those with either of the risk factors alone (odds ratio 3.5, 1.6 to 7.9). Diastolic blood pressure in midlife had no significant effect on the risk of Alzheimer''s disease.Conclusion Raised systolic blood pressure and high serum cholesterol concentration, and in particular the combination of these risks, in midlife increase the risk of Alzheimer''s disease in later life.

This report constitutes the seventh update of the human obesity gene map incorporating published results up to the end of October 2000. Evidence from the rodent and human obesity cases caused by single‐gene mutations, Mendelian disorders exhibiting obesity as a clinical feature, quantitative trait loci uncovered in human genome‐wide scans and in cross‐breeding experiments in various animal models, and association and linkage studies with candidate genes and other markers are reviewed. Forty‐seven human cases of obesity caused by single‐gene mutations in six different genes have been reported in the literature to date. Twenty‐four Mendelian disorders exhibiting obesity as one of their clinical manifestations have now been mapped. The number of different quantitative trait loci reported from animal models currently reaches 115. Attempts to relate DNA sequence variation in specific genes to obesity phenotypes continue to grow, with 130 studies reporting positive associations with 48 candidate genes. Finally, 59 loci have been linked to obesity indicators in genomic scans and other linkage study designs. The obesity gene map reveals that putative loci affecting obesity‐related phenotypes can be found on all chromosomes except chromosome Y. A total of 54 new loci have been added to the map in the past 12 months and the number of genes, markers, and chromosomal regions that have been associated or linked with human obesity phenotypes is now above 250. Likewise, the number of negative studies, which are only partially reviewed here, is also on the rise.  相似文献

Fas-mediated induction of apoptosis is a major factor in the selection of lymphocytes and downregulation of immunological processes. In the present study, we have assessed endothelial Fas-ligand (FasL) expression in normal human ileum, appendix, and colon, and compared the expression levels with that in inflammatory bowel disease and in acute appendicitis. In a normal appendix, endothelial FasL levels were constant in almost half of the mucosal vessels; but, in the normal ileum and colon, endothelial FasL was practically restricted to areas in close proximity to lymphatic follicles, and was expressed mainly in the submucosal aspect of the follicles in the vessels with high endothelium. In samples from subjects with either Crohn’s disease or ulcerative colitis, the extent of endothelial FasL expression was elevated in the submucosa and associated with an elevated number of lymphoid follicles. In inflammatory bowel disease, ulcers and areas with a high density of mononuclear cells expressing FasL also showed an elevated density of blood vessels with endothelial FasL expression. Although the function of endothelial FasL remains unclear, such a specific expression pattern suggests that endothelial FasL expression has a role in the regulation of lymphocyte access to the peripheral lymphoid tissues, including the intestinal mucosa.  相似文献
