Synthetic wastewater was treated in a bench scale submerged membrane bioreactor (SMBR). A long‐term experiment was conducted by varying the sludge residence time (SRT) (10–500 d) and BOD loading (1.3–0.25 kg/m3·d). The biological activity was observed in terms of the oxygen utilization rate (OUR) and adenosine triphosphate (ATP) profile; the process stability was analyzed based on the extent of organic degradation and suction pressure. The microbial population in the SMBR was dependent on the SRT and BOD loading, and its biological activity was increased with an increase in the SRT or BOD loading. At a low feed to microorganism (F/M) ratio (0.06 kg BOD/kg MLSS·d), the sludge production of the reactor was reduced to 0.04 kg MLSS/kg BOD, which is much less than in the conventional activated sludge process (0.4–0.6 kg MLSS/kg BOD). The F/M ratio influenced the biological activity (via ATP and the OUR) significantly at a short SRT (≤90 d). However, the effect of the F/M ratio ceased at a low F/M ratio (≤ 0.07 kg BOD/kg MLSS·d). The accumulation of organics in the SMBR was accompanied with an increase in the supernatant TOC, which caused a high suction pressure and an abrupt change in the operating conditions to process instability. However, the process stability of the SMBR increased with an increase in the SRT and a decrease in the BOD loading along with a concomitant decrease in the biological activity and sludge production.  相似文献

The transformation efficiency of Methylobacillus sp. strain 12S, using electroporation, was unaffected by the growth phase of the cells but competent cells grown at 21 °C had a 1.9 × 103 times higher transformation efficiency than those grown at 30 °C. The stalked crinoid, Metacrinus rotundus, is one of the most basal extant echinoderms. Here, we show the expression patterns of Six3, Pax6, and Otx in the early development of M. rotundus. All three genes are highly expressed in stages from the gastrula to the auricularia larval stage. Ectodermal expression of MrOtx appears to be correlated with development of the ciliary band. These three genes are expressed sequentially along the embryonic body axis in the anterior and middle walls of the archenteron in the order of MrPax6, MrSix3, and MrOtx. The anterior, middle, and posterior parts of the archenteron in the late gastrula differentiate into the axo-hydrocoel, the enteric sac, and somatocoels at later stages, respectively. The three genes are expressed sequentially from the tip of the axo-hydrocoel to the bottom of enteric sac in the order of MrSix3, MrPax6, and MrOtx at the later stages. This suggests that these genes are involved in patterning of the larval endo-mesoderm in stalked crinoids. The present results suggest that radical alterations have occurred in the expression and function of homeobox genes in basal echinoderms.  相似文献

Distribution of (Na+,K+)ATPase on the cell membranes of acinar and duct cells of rat parotid gland was investigated quantitatively by immunoelectron microscopy using the post-embedding protein A-gold technique. In acinar cells, ATPase was localized predominantly on the basolateral plasma membranes. A small but significant amount of (Na+,K+)ATPase was, however, detected on the luminal plasma membranes, especially on the microvillar region of the acinar cells; the surface density on the luminal membrane was approximately one third of that on the basolateral membranes. In duct cells, many gold particles were found on the basolateral membrane, especially along the basal infoldings of the plasma membranes, whereas no significant gold particles were found on the luminal plasma membranes, suggesting unilateral distribution of ATPase in duct cells. We suggest that in acinar cells sodium ion is not only transported paracellularly but is also actively transported intracellularly into the luminal space by the (Na+,K+)ATPase located on the luminal plasma membranes, and that water is passively transported to the luminal space to form a plasma-like isotonic primary saliva, while in the duct cells the same ion is selectively re-absorbed intracellularly by (Na+,K+)ATPase found in abundance along the many infoldings of the basal plasma membranes, thus producing the hypotonic saliva.  相似文献
