The effects of chilling on protein synthesis in vivo in etiolatedhypocotyls of Vigna radiata L. were investigated. After exposureof the tissues to 0?C for various periods of time, proteinswere labeled with [35S]-methionine at 26?C. The total amountof 35S incorporated into soluble and membrane proteins was reversiblyreduced by chilling for 24 h, during which time the tissuessuffered no injury. Further prolonged chilling produced an irreversibledecline both in the incorporation of radioactivity and in cellviability as assessed by the extent of leakage of electrolyte.The 35S-labeled proteins in the soluble and the total membranefractions were analyzed quantitatively. Chilling of etiolatedhypocotyls for one or two days induced the syntheses of twosoluble proteins (82 and 74 kDa) and one membrane protein (80kDa). Moreover, three heat-shock proteins (HSPs) that were inducedby heat stress (41 ?C, 4h) had the same electrophoretic mobilitiesas those of the proteins induced directly or indirectly by thechilling treatment. 1Contribution No. 3170 from the Institute of Low TemperatureScience. We have devised a method for the parallel determination of choline acetyltransferase (CAT) and muscarinic cholinergic receptor (mCh-R) in the same brain tissue. The method for CAT activity determination is more rapid, simplified, stable, and economical than the usual Fonnum's method. With our method, age-associated changes in CAT activity and mCh-R levels were examined. Although CAT activity hardly changed with age except in a few areas, mCh-R binding of aged-rats was markedly reduced in all areas. These results suggest that the change in mCh-R represents an age-associated biochemical change in the brain and that determination of CAT activity is not sufficient for the study of age-associated changes in the brain cholinergic system.  相似文献

We have cloned the tryptophanase structural gene from Escherichia coli B/1t7-A into E. coli K-12 MD55 with a vector plasmid, pBR322. The cloned cells produced a large amount of the enzyme corresponding to more than 30% of the total soluble protein. With the enzyme obtained by this overproduction system, we have prepared three different crystals of tryptophanase, apo-enzyme, holo-enzyme, and a complex of holo-enzyme and L-alanine, by using polyethylene glycol 4000 or potassium phosphate as a precipitant and the hanging drop method. These single crystals appeared to be suitable for X-ray diffraction analysis.  相似文献

1. Total cellular proteins from the livers of 4-, 16- and 52-week-old hepatitis- and hepatoma-predisposed Long-Evans Cinnamon (LEC) rats were compared to those from the livers of the corresponding control rats [Long-Evans Agouti (LEA) rats] by two-dimensional gel electrophoresis. 2. A polypeptide, p50/7.2 (molecular weight x 10(-3)/isoelectric point) was only found in the LEC rats, and the p43/6.4 component was greater and the p51/6.8 component was less in the LEC rats than in the LEA rats during aging. 3. A polypeptide, p29/6.8, was dramatically greater in 4-week-old LEC rats than in 4-week-old LEA rats. 4. By sequencing and Western blotting analysis, the marked differences in the level of the p29/6.8 component were found to be due to carbonic anhydrase III.  相似文献
