Activation of contraction by voltage-clamped stimulation in crayfish muscle fibers (author's transl)

It is known that the properties of the excitation-contraction coupling of crayfish skeletal muscle are different in some respects from those of frog muscle. In the present study, activation of contraction of the crayfish muscle induced by short depolarizing pulses was investigated and it was compared with the results of frog muscle obtained by Adrian, Chandler and Hodgkin (1969). Two glass microelectrodes were inserted into the thoracal muscle of the crayfish. The muscle was stimulated by the voltage-clamped pulses of different durations and the resulting contractions were observed under the binocular microscope with the magnification of 60 X at 20 approximately 23 degrees C. The rheobasic membrane potential was -55mV. The mechanical threshold potential was -42 mV for 10 msec, -15 mV for 2.5 msec, +18 mV for 1 msec and around +90 mV for 0.5 msec pulses. For short pulses where the threshold potential was more positive than -20 mV, the area of the depolarization above -30 mV was 51 mV-msec. Subthreshold pulses produced contraction if applied repetitively. The effect of a just suprathreshold short pulse on the activation of contraction was cancelled by the hyperpolarizing pulse.

Isolation and genetic analysis of resistant mutants to the benzimidazole fungicide benomyl inCoprinus cinereus

In total, 404 variants resistant to the antimicrotubule agent benomyl were isolated from UV-irradiated oidia of the basidiomyceteCoprinus cinereus. Part of the variants showed, in addition to benomyl resistance, heat sensitivity or heat dependence. Fifteen variants selected on the basis of different phenotypes were subjected to further analyses. All of the 15 variations were due to single-gene mutations, and the mutations comprised four groups (benA, benB, benC, andbenD) in terms of genetic linkage. Some of the 15 mutations affected nuclear migration in dikaryosis and/or fruiting processes.     

Association between cytokine removal by polymyxin B hemoperfusion and improved pulmonary oxygenation in patients with acute exacerbation of idiopathic pulmonary fibrosis

Acute exacerbation of idiopathic pulmonary fibrosis (AE-IPF) is characterized by severe worsening dyspnea of unknown etiology and high mortality without effective treatment. Recently, direct hemoperfusion with polymyxin B (PMX)-immobilized fiber cartridge (PMX-DHP) has been reported to improve pulmonary oxygenation and survival in patients with AE-IPF although its mechanism of action remains unknown. To gain insights into the pathobiology of AE-IPF through the beneficial effects of PMX-DHP, we analyzed the profile of cytokines adsorbed onto PMX-fibers used in 9 AE-IPF patients. In addition, the sera of these AE-IPF patients collected immediately before and after PMX-DHP, 9 stable IPF patients and 8 healthy individuals were also analyzed. The serum levels of cytokines including IL-9, IL-12, IL-17, PDGF and VEGF were significantly decreased immediately after PMX-DHP (P < 0.02), and VEGF and IL-12 were most prominently reduced. In addition to PDGF and VEGF, IL-1β, IL-1ra, IL-8, IL-23, FGF basic, GM-CSF, IP-10, RANTES and TGF-β were eluted from used PMX-fibers. Interestingly, improved pulmonary oxygenation after PMX-DHP was correlated well with the quantities of eluted VEGF. These results suggest that adsorption of proinflammatory, profibrotic and proangiogenic cytokines onto PMX-fibers is one of the mechanisms of action of PMX-DHP in AE-IPF. Notably, removal of VEGF by PMX-DHP may contribute to the rapid improvement in oxygenation by suppressing vascular permeability in the lung.     

Fungi associated with roots and fruits of black peppers in the Dominican Republic

Several soilborne diseases have occurred in black pepper (Piper nigrum) plantations in the Dominican Republic since 1977. To understand environments in which the disease occurs fungal floras associated with them were studied. A total of 1,119 isolates, 603 from root segments of 12 vines, and 516 from fruit samples of 30 vines were identified as belonging to a total of 39 genera, namely, 31 from roots and 26 from fruits, excluding unsporulated fungi. The most dominant fungi wereFusarium in both root and fruit samples, followed byRhizoctonia, Diplodia, andPythium from roots, andDiplodia, Colletotrichum andPestalotia from the fruits. Preliminary pathogenicity tests towards leaves or roots of black pepper were conducted.
