We recently reported that Gly573Ser substitution of the transient receptor potential cation channel, subfamily V member 3 (TRPV3) caused hair loss in DS-Nh mice. To further elucidate the effects of this mutation on the development of the spontaneous hairless phenotype, we examined the temperature-response to epidermal sheets from DS-Nh and DS mice. It was indicated that the mutation was gain-of-function. We also performed genetic and histological analyses with both strain skins. DNA microarray data revealed that the levels of keratin-associated protein 16-1, 16-3, and 16-9 genes related to the anagen phase were decreased in the skins of DS-Nh mice compared with those of three days old DS mice. Histological analysis revealed that the anagen phase persisted in DS-Nh mice, and that the telogen phase was seen in DS but not DS-Nh mice at 21 days of age. Regulation of TRPV3 appears to be important for appropriate hair development in rodents.  相似文献

Cyclization of the benzoylpiperidine in lead compound 2 generated a series of novel and highly potent spiropiperidine-based stearoyl-CoA desaturase (SCD)-1 inhibitors. Among them, 1′-{6-[5-(pyridin-3-ylmethyl)-1,3,4-oxadiazol-2-yl]pyridazin-3-yl}-5-(trifluoromethyl)-3,4-dihydrospiro[chromene-2,4′-piperidine] (19) demonstrated the most powerful inhibitory activity against SCD-1, not only in vitro but also in vivo (C57BL/6 J mice). With regard to the pharmacological evaluation, 19 showed powerful reduction of the desaturation index in the plasma of C57BL/6 J mice on a non-fat diet after a 7-day oral administration (q.d.) without causing notable abnormalities in the eyes or skin up to the highest dose (3 mg/kg) in our preliminary analysis.  相似文献

To reveal the role of enzymes involved in PGI2 synthesis for vascular PGI2 generation in experimental hypertensive models, we defined PGI2 synthase and phospholipases activities in the aortic wall of two different experimental hypertensive rats, e.g. spontaneously hypertensive rats (SHR) and desoxycorticosterone acetate (DOCA)-salt hypertensive rats. In the stage of established hypertension both of the hypertensive models had a significantly large capacity of the vascular wall to produce PGI2, as compared to respective control rats. PGI2 synthase activities in the vascular wall were significantly increased by 27% for SHR and by 80% for DOCA-salt hypertensive rats. Moreover, the enzymatic activities were closely related to the blood pressure values for both of the models. On the other hand, phospholipase C or phospholipase A2 activities were increased or unchanged in SHR, respectively, whereas both of the phospholipases were significantly decreased in DOCA-salt hypertensive rats. Thus, it is indicated that PGI2 synthase is partly responsible for the increased PGI2 generation in the vasclar wall of SHR and DOCA-salt hypertensive rats, and that vascular phospholipase C is playing a more important role in providing arachidonate for PGI2 synthesis in SHR.  相似文献

We previously demonstrated that a methionine-threonine-supplemented low (8.5%) casein diet (8.5CMT) reduced symptoms such as proteinuria in nephritic rats without severe protein malnutrition. In this study, we examined whether or not L-arginine supplementation to 8.5CMT would exacerbate proteinuria and other symptoms in nephritic rats. Male Wistar rats with glomerulonephritis induced by a single intravenous injection of nephrotoxic serum were fed either a 20% casein diet (control), 8.5% casein diet, 8.5CMT, or L-arginine-supplemented 8.5CMT (8.5CMTA) for 16 days. The 8.5CMTA, as compared with the 8.5CMT, aggravated proteinuria and glomerulonephritis. Administration of L-NG-nitroarginine methyl ester, an inhibitor of nitric oxide synthase, to 8.5CMTA-fed nephritic rats by drinking water for 14 days canceled the adverse effect of L-arginine on proteinuria and histopathological damage in glomeruli. These results suggest that the supplementation of L-arginine makes exacerbation via nitric oxide production in glomerulonephritis.  相似文献

To assess the implications of vascular eicosanoids system in the hypertension of Dahl salt-sensitive (Dahl S) strain, we investigated the production of vascular vasodepressor and vasoconstrictor eicosanoids in Dahl S rats. 14-week-old Dahl S rats on a 0.11% NaCl diet (normotension) or a 0.3% NaCl diet (borderline hypertension) had a significantly lowered generation of vascular prostacyclin (PGI2), compared with Dahl salt-resistant (Dahl R) rats. The impairment of vascular PGI2 in Dahl S rats was restored to the normal level of Dahl R rats with the elevation of blood pressure induced by a high salt diet (4% NaCl). The production of vascular PGI2 was closely related to the height of blood pressure. The deterioration of vascular PGI2 was also found in 4-week-old Dahl S rats with normotension. Conversely, vascular thromboxane A2 (TXA2) was significantly enhanced in 14-week-old Dahl S rats in all of the feeding groups. Thus, it seems possible that the proved alterations of the vasodepressor and vasoconstrictor eicosanoids partially contribute to the genesis of salt hypertension. Although the exact mechanisms remain obscure, the adaptation of vascular PGI2 on a high salt diet may be suitable to compete with the high blood pressure and to protect against the vascular damage.  相似文献

Estrogen receptors in the nuclei of rat uterus were determined 1 or 6 h after the injection of [3H]estradiol. Ratio of 0.4 M KCl-extractable to -resistant receptor-estradiol complex was constant during this time interval. Three consecutive extractions took out about 95% of receptor-estradiol complex either 1 or 6 h after injection. Extraction with KCl before the exchange assay reduced the amount of salt-resistant nuclear receptor-estradiol complex. However, when exchange incubation with [3H] estradiol was done before KCl extraction, salt-resistant receptor-estradiol complex was significantly increased, even after three consecutive extractions.  相似文献

A mouse leukemic cell line L1210 Sg with a low sensitivity to interferon-γ (IFN-γ) was described. On the nature of the antiviral action and binding of IFN, L1210 Sg cells were compared with L1210m cell line which is sensitive to IFN-γ. For a half reduction of the vesicular stomatitis virus-RNA synthesis, L1210 Sg cells required 500–fold more IFN-γ than L1210m cells did. However, both cell lines were induced to the antiviral state to the same extent with IFN-α or -β. L1210 Sg and L1210m cells were sensitive to the anti-proliferative action of IFN-α and -β, but insensitive to IFN-γ. (2′-5′)Oligoadenylate synthetase was induced in these cell lines by IFN-β, but not by IFN-γ, which suggests that the induction of this synthetase may not be responsible for the antiviral action of IFN-γ. No substantial difference between L1210 Sg and L1210m cells was found in IFN receptors for IFN-γ and IFN-β either in number per cell or in their affinity to corresponding IFN type. However, differences were noted in time course profiles of cell-associated IFN-γ at 37 C: in L1210m cells, a rise-and-decay profile of IFN-γ bound to cells was observed at 37 C, but in L1210 Sg cells, rise and slight decay was observed. On the other hand, a similar rise-and-decay curve of IFN-β bound to these cells was observed. These results indicated that the low sensitivity of L1210 Sg cells to IFN-γ may be due to this slight decay of receptor-bound IFN-γ.  相似文献

Aims:  To develop a scanning electron microscopic approach using in situ hybridization (SEM–ISH) for gaining both genetic and morphological information about target bacteria.
Methods and Results:  Target cells were hybridized with DNA-targeted polynucleotide probes, and a tyramide signal amplification system was used to increase the sensitivity. The protocol of SEM–ISH enabled to detect low copy number target DNA sequences in individual cells.
Conclusions:  SEM–ISH allowed the in situ detection of bacteria carrying a specific gene.
Significance and Impact of the Study:  Combining morphological study with SEM and ISH techniques appears to be a valuable tool to understand the spatial distribution of target cells in complex microbial communities on various materials.  相似文献
