A novel anti-infection strategy to alleviate antibiotic-resistance problem and non-specific toxicity associated with chemotherapy is explored in this study. It is based on utilizing a bacteriolytic enzyme (lysozyme) as a carrier to allow specific targeting of a potential phenolic antimicrobial drug (triclosan) to microbial cells. Lysozyme (LZ) was complexed, via electrostatic and hydrophobic condensation at alkaline pH, to various degrees with triclosan (TCS), a negatively charged phenolic antimicrobial that inhibits bacterial fatty acid synthesis. Fluorescence and absorbance spectra analysis revealed non-covalent association of TCS with the aromatic residues at the interior of LZ molecule. The conjugation greatly promoted the lytic activity of LZ as the degree of TCS derivatization increased. The complexation with LZ turned TCS into completely soluble in aqueous solution. TCS-LZ complexes showed significantly enhanced bactericidal activity against several strains of Gram-positive and Gram-negative bacteria compared to the activity of TCS or LZ alone when tested at the same molar basis. Strikingly, TCS-LZ complex, but not LZ or TCS alone, exhibited unique specificity to scavenge superoxide radicals, generated by the natural xanthine/xanthine oxidase coupling system, without affecting the catalytic function of oxidase. This finding is the first to describe that the membrane disrupting function of lysozyme can be utilized to specifically target antimicrobial drug(s) to pathogen cells and heralding a fascinating opportunity for the potential candidacy of TCS-LZ as novel antimicrobial strategy for human therapy.  相似文献

We investigated the seedling survival of five evergreen tree species over 3 years inside and outside deer-exclusion fences in a warm temperate evergreen broad-leaved forest on Yakushima Island, Japan. Seedling survival was examined in relation to topography, herbivory by sika deer, and the soil surface environment (i.e., soil surface wetness, light conditions, slope inclination, and soil disturbance). The study species included Myrsine seguinii Lév., Syzygium buxifolium Hook. et Arn. (Group A: species distributed on the upper slope of the study site), Litsea accuminata (Bl.) Kurata, Schefflera octophylla (Lour.) Harms (Group B: species distributed on the lower slope), and Cleyera japonica Thunb. p.p. emend. Sieb. et Zucc. (Group C: species distributed on both slopes). The soil surface environment on the upper slope was drier, lighter, and more easily disturbed than the lower slope. Generalized linear model analyses indicated that seedling survival in fenced and unfenced quadrats was greater on the upper slope than on the lower slope for Group A and B species but not for Group C species. A micro-spatial scale analysis revealed that seedling survival was correlated with soil wetness, ground light conditions, and soil disturbance but not slope inclination. These results indicate that seedling survival was correlated with topography, sika deer herbivory, and the micro-spatial scale environment. Topography-related differences in seedling survival appear to adequately reflect the observed adult plant distributions for Group A and C species but not for Group B species.  相似文献

By defining functional defects in a congenital myasthenic syndrome (CMS), we show that two mutant residues, located in a binding site region of the acetylcholine receptor (AChR) epsilon subunit, exert opposite effects on ACh binding and suppress channel gating. Single channel kinetic analysis reveals that the first mutation, epsilon N182Y, increases ACh affinity for receptors in the resting closed state, which promotes sequential occupancy of the binding sites and discloses rate constants for ACh occupancy of the nonmutant alphadelta site. Studies of the analogous mutation in the delta subunit, deltaN187Y, disclose rate constants for ACh occupancy of the nonmutant alpha epsilon site. The second CMS mutation, epsilon D175N, reduces ACh affinity for receptors in the resting closed state; occupancy of the mutant site still promotes gating because a large difference in affinity is maintained between closed and open states. epsilon D175N impairs overall gating, however, through an effect independent of ACh occupancy. When mapped on a structural model of the AChR binding site, epsilon N182Y localizes to the interface with the alpha subunit, and epsilon D175 to the entrance of the ACh binding cavity. Both epsilon N182Y and epsilon D175 show state specificity in affecting closed relative to desensitized state affinities, suggesting that the protein chain harboring epsilon N182 and epsilon D175 rearranges in the course of receptor desensitization. The overall results show that key residues at the ACh binding site differentially stabilize the agonist bound to closed, open and desensitized states, and provide a set point for gating of the channel.  相似文献

We have investigated tree distributions in relation to topography between different tree life history stages, from the seed-dispersal stage to the adult stage in a warm temperate evergreen broadleaved forest on Yakushima Island, Japan, to clarify the critical stages in determining adult tree distributions. We conducted a census of all living trees > or =30 cm tall and collected seed falls over three years using 25 seed traps in a 50 m x 50 m quadrat. Four life stages were defined: stage 1, dispersed seed; stage 2, individuals taller than 30 cm and diameter at breast height (DBH) < 1 cm; stage 3, trunks 1 cm < or = DBH < 10 cm; stage 4, trunks with DBH > or = 10 cm. We classified 17 common tree species into three groups; group A was distributed mainly on the upper slope, group B on the lower slope, and group C on both. Most of group A and B trees at stages 2-4 showed an aggregated distribution along the topographical gradient. The densities at stage 1 showed weaker aggregations according to slope. Topography-specific tree distribution was probably determined at the regeneration stage, and later survival was less effective as a mechanism of vegetation differentiation.  相似文献

Questions: Do the population dynamics of trees differ among topographic positions and, if so, how does topographic position affect the population dynamics of species that are distributed in a topography‐specific manner? Which is the most important life stage in determining vegetation patterns? Location: Primary and secondary warm temperate evergreen broad‐leaved forest (40 ‐ 280 m a.s.l.) on the western part of Yakushima Island, Japan. Methods: Mortality, recruitment, DBH growth and distribution of stems (= 5 cm DBH) in a 2.62‐ha plot were surveyed in 1992 and 2002 to determine the relationships between population parameters and (1) topography and (2) distribution patterns of 17 common tree species. Results: Common species (n = 17) were classified into three distribution pattern groups: group A, distributed mainly on convex slopes; group B, on concave slopes, and group C, not aggregated with respect to topographic position. Stem mortality, recruitment and DBH growth were greater in group A than in group B within each topographic class. The hierarchy of stem mortality among topographic classes for groups A and B was convex > planar > concave. Stem recruitment density was relatively high on the convex and concave slopes, respectively, for groups A and B. Conclusions The topographical positions of adult trees were not always most suited for adult survival and growth. For group A, the distribution pattern of adults was determined in the juvenile stage, while this was not the case for group B. Studies of juvenile stages are important for understanding the demographic basis of vegetation distribution patterns.  相似文献

The adult mammalian brain is composed of distinct regions with specialized roles including regulation of circadian clocks, feeding, sleep/awake, and seasonal rhythms. To find quantitative differences of expression among such various brain regions, we conducted the BrainStars (B*) project, in which we profiled the genome-wide expression of ~50 small brain regions, including sensory centers, and centers for motion, time, memory, fear, and feeding. To avoid confounds from temporal differences in gene expression, we sampled each region every 4 hours for 24 hours, and pooled the samples for DNA-microarray assays. Therefore, we focused on spatial differences in gene expression. We used informatics to identify candidate genes with expression changes showing high or low expression in specific regions. We also identified candidate genes with stable expression across brain regions that can be used as new internal control genes, and ligand-receptor interactions of neurohormones and neurotransmitters. Through these analyses, we found 8,159 multi-state genes, 2,212 regional marker gene candidates for 44 small brain regions, 915 internal control gene candidates, and 23,864 inferred ligand-receptor interactions. We also found that these sets include well-known genes as well as novel candidate genes that might be related to specific functions in brain regions. We used our findings to develop an integrated database (http://brainstars.org/) for exploring genome-wide expression in the adult mouse brain, and have made this database openly accessible. These new resources will help accelerate the functional analysis of the mammalian brain and the elucidation of its regulatory network systems.  相似文献

Background  

Innate immunity is the ancient defense system of multicellular organisms against microbial infection. The basis of this first line of defense resides in the recognition of unique motifs conserved in microorganisms, and absent in the host. Peptidoglycans, structural components of bacterial cell walls, are recognized by Peptidoglycan Recognition Proteins (PGRPs). PGRPs are present in both vertebrates and invertebrates. Although some evidence for similarities and differences in function and structure between them has been found, their evolutionary history and phylogenetic relationship have remained unclear. Such studies have been severely hampered by the great extent of sequence divergence among vertebrate and invertebrate PGRPs. Here we investigate the birth and death processes of PGRPs to elucidate their origin and diversity.  相似文献

The Alexandrium spp. resting cysts were found abundantly in faecal pellets collected from the bottom sediments at two stations in Hiroshima Bay. It is considered that these faecal pellets were excreted by the macrobenthos, such as polychaeta and mollusca, based on their size and morphology. Polychaeta was the most dominant macrobenthos, and mollusca was the second most dominant group in Hiroshima Bay. The resting cysts of Alexandrium spp. in the bottom sediments at the two stations were counted in both the faecal pellets of macrobenthos and in the surrounding sediment. As a result, the number of cysts in the faecal pellets accounted for 28.9-35.2% of total cysts. In addition, cysts isolated from faecal pellets had almost the same germination ability as those in the sediment. Thus, Alexandrium cysts are tolerant to the predation and digestive processes of macrobenthic organisms.  相似文献
