A population of Stratiomys japonica, a species belonging to the family Stratiomyidae (Diptera), common name ‘soldier flies’, occurs in a hot volcanic spring, which is apparently among the most inhospitable environments for animals because of chemical and thermal conditions. Larvae of this species, which naturally often experience temperatures more than 40 °C, have constitutively high concentrations of the normally inducible heat-shock protein Hsp70, but very low level of corresponding mRNA. Larvae of three other species of the same family, Stratiomys singularior, Nemotelus bipunctatus and Oxycera pardalina, are confined to different type semi-aquatic habitats with contrasting thermal regime. However, all of them shared the same pattern of Hsp70 expression. Interestingly, heat-shock treatment of S. japonica larvae activates heat-shock factor and significantly induces Hsp70 synthesis, whereas larvae of O. pardalina, a species from constant cold environment, produce significantly less Hsp70 in response to heat shock. Adults of the four species also exhibit lower, but detectable levels of Hsp70 without heat shock. Larvae of all species studied have very high tolerance to temperature stress in comparison with other Diptera species investigated, probably representing an inherent adaptive feature of all Stratiomyidae enabling successful colonization of highly variable and extreme habitats.  相似文献

Locusts are major intermittent threats to food security and the ecological factors determining where and when these occur remain poorly understood. For many herbivores, obtaining adequate protein from plants is a key challenge. We tested how the dietary protein : non-structural carbohydrate ratio (p : c) affects the developmental and physiological performance of 4th-5th instar nymphs of the South American locust, Schistocerca cancellata, which has recently resurged in Argentina, Bolivia and Paraguay. Field marching locusts preferred to feed on high carbohydrate foods. Field-collected juveniles transferred to the laboratory selected artificial diets or local plants with low p : c. On single artificial diets, survival rate increased as foods became more carbohydrate-biased. On single local plants, growth only occurred on the plant with the lowest p : c. Most local plants had p : c ratios substantially higher than optimal, demonstrating that field marching locusts must search for adequate carbohydrate or their survival and growth will be carbohydrate-limited. Total body lipids increased as dietary p : c decreased on both artificial and plant diets, and the low lipid contents of field-collected nymphs suggest that obtaining adequate carbohydrate may pose a strong limitation on migration for S. cancellata. Anthropogenic influences such as conversions of forests to pastures, may increase carbohydrate availability and promote outbreaks and migration of some locusts.  相似文献

The phylogeny of selected genera from four subfamilies of fungus gnats (Diptera: Mycetophilidae) – Manotinae, Leiinae, Sciophilinae and Gnoristinae (including Metanepsiini) – is reconstructed based on the combined analysis of five mitochondrial (12S, 16S, COI, COII, cytB) and two nuclear (28S, ITS2) gene markers. Results of the different analyses all support Manotinae as a monophyletic group, with Leiinae as the sister group. Allactoneura DeMeijere is nested in the monophyletic and strongly supported clade of Leiinae. The tribe Metanepsiini is revealed as paraphyletic and the genera Metanepsia Edwards and Chalastonepsia Søli do not appear to be closely related. The genera Docosia Winnertz, Ectrepesthoneura Enderlein, Novakia Strobl and Syntemna Winnertz were placed with a group of genera included traditionally in the Gnoristinae. The monophyly of Dziedzickia Johannsen and Phthinia Winnertz is not supported. The genera of Sciophilinae (excluding Paratinia Mik but including Eudicrana Loew) form a monophyletic group in the Bayesian model.  相似文献

We adapted a recently developed nonrestrictional, nonligational genome walking method, Universal Fast Walking (UFW), for detection of length polymorphism in the proximal promoter region of genes. We demonstrate its efficacy at discovering naturally occurring transposition into heat‐shock genes of wild Drosophila and show that it surmounts limitations of simple polymerase chain reaction (PCR) approaches. We further present modifications to the standard UFW protocol and provide some guidelines to improve specificity. Although the resultant banding pattern of a standard UFW can be regarded as a DNA fingerprint, many amplicons result from false priming and not real polymorphisms. We describe ways to distinguish between UFW amplicons and false priming products in a high‐throughput assay.  相似文献

Spodoptera frugiperda (J.E. Smith) is a major pest in maize fields in Argentina. However, a sampling method that accounts for spatial pattern and allows reliable pest density estimations is still lacking. This paper addresses the issue of how sampling unit size can influence the characterization of S. frugiperda spatial pattern and the performance of sampling plans. An intensive sampling programme for S. frugiperda larvae was carried out in maize fields from October until March in four growing seasons. On each sampling date, 12 to 20 sampling units were taken at random. Each unit consisted of 1, 2, 4, 6, 8, 10 or 12 consecutive plants along a row. The linearized version of the Taylor's power law (TPL) was fitted to mean and variance estimated for each sampling date and data set through least squares regression. In general, b values from TPL were significantly >1 (P<0.05), indicating an aggregated sampling distribution. Regression slopes (b) ranged from 1.28 to 1.48 in small larvae and from 1.06 to 1.24 in large larvae, indicating stronger clumping in the former. TPL parameters were used to develop constant precision sampling plans. The efficiency of these plans in terms of expected total cost (searching and handling sampling units) was very sensitive to the size of the sampling units with small larvae but not with large larvae. The influence of sampling unit size and spatial pattern on TPL parameters and sampling costs is discussed.  相似文献

The influence of weather factors and the predator Doru luteipes Scudder density on Diatraea saccharalis (F.) egg predation was studied. Mortality of D. saccharalis eggs was determined by artificially infesting maize plots with egg masses at various times within each of two maize-growing seasons. Each egg cohort was monitored every 24-48 h to determine the fate of eggs, and predation rates were calculated. Doru luteipes were sampled every 7-10 d, and the mean air temperature, the minimum percentage of relative humidity, and rainfall accumulations were recorded during the egg exposure period. To test the effects of abiotic and biotic variables on egg predation, we used a generalized linear model (GLM). Diatraea saccharalis egg predation was negatively associated with rainfall, whereas D. luteipes density and mean temperature were positively correlated with mortality. The implications of these findings for the management of D. saccharalis are discussed.  相似文献
