Changes in adult body mass during breeding can reveal how parents prepare energetically for care, the stress of care, and the need to terminate care in a state conducive for future reproduction. Interpreting changes in parent mass can be difficult, however, because temporal variation in body mass may reflect a constraint imposed by the stress of care, revealing conflict within the family, or a shift to a new body mass optimum adaptive for a different stage of the breeding cycle. Here, we examined the effect of food deprivation and parenting on variation in female body mass of Nicrophorus orbicollis, an insect in which parents and offspring share a common food resource (a prepared carrion ball). Female parents demonstrated a remarkable degree of regulation of body mass: Despite varied periods of food deprivation (0–8 d) prior to discovery of a carcass, females attained a similar body mass (108.3–109.2% of pre‐deprivation mass) at the time of larval hatching. Females attained a greater body mass in anticipation of rearing a greater number of young. Mothers lost mass during active parental care, and mass at the end of caregiving was less in mothers that reared more and heavier young. Body mass at the end of care was less than the preferred mass for females searching for a carcass, indicating that the mothers sacrificed self‐maintenance and future reproductive potential for their current brood. Contrary to prediction, pre‐breeding food deprivation had no effect on offspring size or on female condition at the end of care. We conclude that there is a limited degree of conflict over the sharing of food among N. orbicollis parents and offspring, but that this conflict is not exacerbated by food deprivation prior to breeding.  相似文献

The relationships of larval nutritional resources with adult body size, starvation resistance and reproductive decisions are not always clear. Burying beetle larvae with inadequate nutrition are hypothesized to develop into relatively large adults that are able to contest for breeding resources. The trade‐off is that the emerging adult has minimal energy reserves and is more susceptible to starvation, and must gain proportionately more weight after emerging. These hypotheses are investigated in Nicrophorus orbicollis Say. In addition, sex differences in size–mass ratios as well as starvation and recovery in reproductively mature females are examined. The larval mass to adult size ratio is similar in male and female N. Orbicollis and, contrary to prediction, small larvae do not result in adults that are relatively large in size for their mass. Emerging adults of lesser mass resist starvation less well, as expected. Emerging adults of smaller pronotal size gain more relative mass but less absolute mass than larger adults. In reproductively mature adult females, recovery from food deprivation is rapid, with most if not all the weight that is lost during a 9–10‐day starvation period being re‐gained within 1 day of engorging. The ability to gain weight rapidly and regulate body mass provides a nutritional framework for understanding the larva to adult transition and the reproductive and parenting decisions of burying beetles that otherwise would appear to be of too high risk.  相似文献

Recent work [Ecoscience (2000) vol. 7, 395-397] suggests that the burying beetle Nicrophorus pustulatus may have undergone a remarkable host shift, exploiting snake eggs rather than carrion as resources for breeding. We conducted behavioural and physiological experiments to examine the hypothesis of a host shift and to formulate hypotheses on its origin. Two congeners of N. pustulatus, Nicrophorus orbicollis and Nicrophorus defodiens did not respond to snake eggs with typical breeding behaviour. When N. pustulatus male-female pairs (n = 14) were presented with clutches of snake eggs, the number of offspring but not the mean size of offspring varied with snake egg mass, indicating effective regulation of brood size. When breeding on turtle eggs, N. pustulatus had a more variable response than when exploiting snake eggs, suggesting that turtle eggs are not a primary resource for breeding. Nicrophorus pustulatus presented with both snake eggs and a mouse carcass combined and exploited the two resources within the same nest (10 of 12 trials). Mouse carcasses and snake eggs were treated differently. Carcasses were moved, buried and stripped of hair in a manner characteristic of burying beetles, whereas snake eggs were not moved or buried. Females that discovered a mouse carcass also had a significantly greater juvenile hormone increase than did females discovering snake eggs. Some responses to the two resources, however, were similar. Female N. pustulatus oviposited rapidly in response to either a mouse carcass or snake eggs, and males elevated sex pheromone emission in response to either resource. The efficient use of snake eggs, the ability to regulate brood size and the different responses to snake eggs and carrion suggest that N. pustulatus is well adapted to exploiting snake eggs for breeding. The use of snake eggs by N. pustulatus has potential implications for conservation of oviparous reptiles.  相似文献

The challenge hypothesis, conceived for testosterone and vertebrates, has recently been applied to juvenile hormone (JH) and insects. Scott [Scott, M.P., 2006a. Resource defense and juvenile hormone: the "challenge hypothesis" extended to insects. Horm. Behav. 49, 276-281] found that JH in the burying beetle Nicrophorus orbicollis increased in response to a social challenge in the presence of a breeding resource, while there was no such JH response in the purported brood parasite Nicrophorus pustulatus. Two important implications of the challenge hypothesis that need to be tested are whether JH affects dominance and why there are interspecific differences in the JH response to a social challenge. The effect of JH on dominance in burying beetles was examined by topical application of JH III to one of two competing females at 24 h and again at 1 h prior to presentation of a breeding resource (mouse carcass). JH supplementation had no effect on dominance in intraspecific interactions in N. orbicollis, as measured by possession of the carcass on Day 1, 3 or 7. Similarly, JH did not alter carcass ownership during competition between N. orbicollis and N. pustulatus nor did it affect reproductive success. Irrespective of JH supplementation, N. pustulatus became increasingly dominant as the trials progressed, rarely occupying the carcass on Day 1, but excluding N. orbicollis in nearly half the trials by Day 7. These findings, and a brief review of the burying beetle literature, suggest that the challenge hypothesis, as applied to testosterone and vertebrates, does not yet have an analogous model for JH and insects.  相似文献

Polymerization of 1,2-anhydro-3,4,6-tri-O-benzyl-beta-D-mannopyranose under acid catalysis has led to a series of polymers varying in anomeric configuration from approximately 90% alpha to 70% beta. Optical rotations follow 13C-n.m.r. estimates of anomeric composition linearly over this range. Low-temperature polymerization with trifluoromethanesulfonic anhydride as initiator favors mainly cis-opening of the anhydro ring, presumably through the intermediary of a macroester. These results are compared with related glycosylation and polymerization reactions on 1,2-anhydro sugar derivatives, and some mechanistic conclusions are proposed.  相似文献

Single-strand breaks (ssb) in opposite strands of DNA can be sufficiently near that a double-strand break (dsb) results. A theory is presented by which the maximum number h of base pairs which cannot prevent double-strand breakage can be determined from the rates of production of ssb and dsb. The assumptions required to derive the necessary equations as well as the range of validity of the equations are discussed in detail. In the experiments ssb and dsb were produced by x-irradiation in buffers which do not eliminate indirect effects and were measured by analytical ultracentrifugation. Values of h have been determined in low and high ionic strength and in low ionic strength over a range of temperatures. The values, 2.64 and 15.8, were obtained for high and low ionic strength, respectively.  相似文献

Protease-activated receptor 4 (PAR4) is cleaved by thrombin at the R47-G48 peptide bond. Unlike PAR1, PAR4 does not contain a sequence readily predicted to interact with thrombin anion binding exosite-I. HPLC kinetic results on hydrolysis of PAR4 peptides (38-51 and 38-62) reveal that extending the sequence from the active site toward the exosite does not promote further binding interactions with thrombin. One-dimensional-proton line-broadening NMR indicates that the amino acids occupying the P(4)-P(1) positions of PAR4 (38-47), 44PAPR(47), come into direct contact with the thrombin surface. Less contact arises from the Leu43 at the P(5) position. Two-dimensional total correlation spectroscopy and two-dimensional transferred nuclear Overhauser effect spectroscropy studies on this complex reveal that Leu43 is flexible and can exhibit two conformational states. The binding mode observed for PAR4 peptides is similar to that of PAR1 peptides. PAR4 takes advantage of a distinctive sequence to optimize its interactions with the thrombin active site surface.  相似文献

In the blood coagulation cascade, thrombin cleaves fibrinopeptides A and B from fibrinogen revealing sites for fibrin polymerization that lead to insoluble clot formation. Factor XIII stabilizes this clot by catalyzing the formation of intermolecular cross-links in the fibrin network. Thrombin activates the Factor XIII a(2) dimer by cleaving the Factor XIII activation peptide segment at the Arg(37)-Gly(38) peptide bond. Using a high performance liquid chromatography assay, the kinetic constants K(m), k(cat), and k(cat)/K(m) were determined for thrombin hydrolysis of fibrinogen Aalpha-(7-20), Factor XIII activation peptide-(28-41), and Factor XIII activation peptide-(28-41) with a Val(34) to Leu substitution. This Val to Leu mutation has been correlated with protection from myocardial infarction. In the absence of fibrin, the Factor XIII activation peptide-(28-41) exhibits a 10-fold lower k(cat)/K(m) value than fibrinogen Aalpha-(7-20). With the Factor XIII V34L mutation, decreases in K(m) and increases in k(cat) produce a 6-fold increase in k(cat)/K(m) relative to the wild-type Factor XIII sequence. A review of the x-ray crystal structures of known substrates and inhibitors of thrombin leads to a hypothesis that the new Leu generates a peptide with more extensive interactions with the surface of thrombin. As a result, the Factor XIII V34L is proposed to be susceptible to wasteful conversion of zymogen to activated enzyme. Premature depletion may provide cardioprotective effects.  相似文献

Undertakers are considered to be among the most specialized of pre-foraging honey bee (Apis mellifera L.) workers. In this study we examined a possible benefit and a cost of the corpse-rem oval specialty, the improvement in performance with experience, and interference by individuals attempting to perform the same task in the same location, respectively. Experienced bees removed corpses significantly faster than less experienced bees and also were less likely to drop corpses while exiting the hive (5.5% vs. 14.3% of attempts). Superior performance by experienced undertakers might occur as a consequence of learning, or by greater ability from the outset. Because active undertakers (≥ 3 corpse removals) did not improve with experience over their own careers, learning was not demonstrated. An extreme specialist, Yellow 54, removed a total of 114 corpses (33.8% of experimentally introduced dead bees) from the hive over a 13-day period. This is the longest recorded tenure of undertaking to date and demonstrates how a few individuals can dominate this task in a honey bee colony-Yellow 54 removed corpses significantly faster than other active bees, but she demonstrated no obvious improvement in performance over her undertaking career. This suggests the possibility that active undertakers were more talented than less active undertakers, irrespective of learning. When two undertakers worked together to remove a corpse from the hive, they took longer to complete the task than did single individuals. When multiple undertakers flew together from the hive, they were less likely to clear a nearby obstruction than single undertakers and were more likely to drop the corpse within 1 m of the hive. Thus, mutual interference exacted a measurable cost as a result of the undertaking specialization while learning provided few benefits.  相似文献

Research was conducted to evaluate the ability of a broad-specificity beta-glucosidase in mammalian tissues to catalyze the hydrolytic release of free pyridoxine from pyridoxine-5'-beta-D-glucoside, a naturally occurring form of vitamin B6 in plant-derived foods. Activity was detected in liver and intestinal mucosa using tritiated pyridoxine glucoside as a substrate. In the rat and guinea pig, enzyme activity was greater in intestine than in liver or kidney while even greater activity was detected in human intestinal tissue. Reaction rates were, however, low in all tissues. Hydrolysis of the synthetic substrate 4-methylumbelliferyl-beta-D-glucoside was also greatest in intestinal tissue. The characteristics of the enzymatic hydrolysis of pyridoxine glucoside to pyridoxine included: (i) most activity in the soluble tissue fraction, (ii) a pH optimum of approximately 6.0, and (iii) inhibition caused by the addition of sodium taurocholate. These characteristics are very similar to those of the broad-specificity beta-glucosidase in mammalian tissues with respect to the hydrolysis of a variety of naturally occurring and synthetic substrates. The apparent Km was greater than 2 mM for pyridoxine glucoside hydrolysis by intestinal preparations of each species, which is much greater than expected intestinal concentrations derived from dietary sources. In vivo studies have indicated that the intestine is involved in the metabolic utilization of dietary pyridoxine glucoside. The results observed here suggest that an alternate process, possibly involving intestinal microorganisms, may also be involved in the in vivo hydrolysis of pyridoxine glucoside.  相似文献
