A new huanglongbing (HLB) "Candidatus Liberibacter" species is genetically characterized, and the bacterium is designated "Candidatus Liberibacter psyllaurous." This bacterium infects the psyllid Bactericera cockerelli and its solanaceous host plants potato and tomato, potentially resulting in "psyllid yellowing." Host plant-dependent HLB transmission and variation in psyllid infection frequencies are found.  相似文献

Among the many topics of interest to ecologists studying associations between phytophagous insects and their host plants are the influence of natal host plant on future oviposition decisions and the mechanisms of generalist versus specialist host selection behavior. In this study, we examined the oviposition preferences, behavior and larval development of the tomato/potato psyllid, Bactericera cockerelli. By rearing psyllids with two distinct geographically-linked haplotypes on different host plants, we were able to examine the role of natal host plant and potential local adaptation on host plant usage. Choice bioassays among three host species demonstrated that psyllids from California had clear preferences that were influenced by natal plant. We further found that patterns in choice bioassays corresponded to observed feeding and movement responses. No-choice bioassays demonstrated that there is little to no association between development and host-plant choice for oviposition, while also indicating that host choice varies between haplotypes. These findings support the concept that mothers do not always choose oviposition sites optimally and also add support for the controversial Hopkins'' host selection principle.  相似文献

Tamarixia triozae (Burks) (Hymenoptera: Eulophidae) is an important parasitoid of the potato or tomato psyllid, Bactericera cockerelli (Sulc) (Hemiptera: Trizoidae), a serious pest of potato (Solanum tuberosum L.), tomato (Solanum lycopersicum L.), and other solanaceous vegetables in many countries. To produce a marketable crop, insecticides are required when B. cockerelli populations reach economically damaging levels. We evaluated 11 commonly used insecticides for their effects on T. triozae. Glass-surface residues of spinetoram, imidacloprid-cyfluthrin, abamectin, and tolfenpyrad caused 100% mortality of T. triozae in 72 h, and the leaf residue of spinetoram was extremely toxic to T. triozae adults; even 15-d-old residues caused 100% mortality. Cyantraniliprole, fenpyroximate, pymetrozine, spirotetramat, spiromesifen, and chenopodium oil did not cause significant mortality in either glass surface or leaf-residue bioassays. Ingestion of spinetoram, abamectin, and imidacloprid+cyfluthrin (Leverage) by the adults resulted in 100% mortality in 12 h, and tolfenpyrad, 75.0% mortality in 12 h; whereas chenopodium oil and pymetrozine showed moderate effects on adult survival. Ingestion of abamectin, imidacloprid-cyfluthrin, and spinetoram killed all adults in the first day of treatment, whereas female adults in the treatment of pymetrozine lived 80.8 d, which was similar to those in the control. Ingestion of abamectin, imidacloprid-cyfluthrin, chenopodium oil, and spinetoram killed all male adults in the first day, whereas ingestion of other insecticides did not cause significant mortality, but reduced percent parasitism. Abamectin, imidacloprid-cyfluthrin, and spinetoram had the most deleterious effects on T. triozae, and have the least potential for use in integrated control programs using this parasitoid.  相似文献

Calmodulin-binding proteins are involved in numerous cellular signaling pathways. The biotinylated-calmodulin overlay is a nonradioactive method widely used to detect calmodulin-binding proteins in tissue and cell samples. This method has several limitations; therefore, we developed a nonradioactive calmodulin-binding protein detection overlay using an S-tag-labeled calmodulin fusion protein. An expression system was used to generate a calmodulin fusion protein with an S-tag label, a 15 amino acid sequence that binds to a 105 amino acid S-protein. The S-protein is conjugated to horseradish peroxidase for final detection with a chemiluminescent substrate. The S-tag calmodulin was compared to purified calmodulin and biotinylated calmodulin in a calmodulin-dependent phosphodiesterase assay. The results of the calmodulin-dependent phosphodiesterase assay indicate that S-tag calmodulin induces higher phosphodiesterase activity than biotinylated calmodulin and lower activity than purified calmodulin. A comparison of the biotinylated and S-tag calmodulin overlay assays indicate that S-tag calmodulin is more sensitive than biotinylated calmodulin in the detection of calcineurin, a known calmodulin-binding protein. The overlay assay results also indicate that the S-tag calmodulin and biotinylated calmodulin detect similar calmodulin-binding proteins in colon epithelial cells. In conclusion, the S-tag calmodulin overlay assay is a consistent, sensitive, and rapid nonradioactive method to detect calmodulin-binding proteins.  相似文献

Adult tomato psyllid, Bactericerca (Paratrioza) cockerelli (Sulc) (Homoptera: Psyllidae), behavioral responses were evaluated for five tomato plant lines and for the interactions of insecticides with four commercial cultivars. Plant lines tested included the commercial 'Shady Lady', 'Yellow Pear', '7718 VFN', 'QualiT 21', and the plant introduction line PI 134417. Insecticides included a kaolin particle film, pymetrozine, pyriproxyfen, spinosad, and imidacloprid. Psyllids spent significantly more time feeding on 'Yellow Pear' than all other plant lines except '7718 VFN'. In comparisons among plant lines, psyllids exposed to the wild accession PI 1.34417 showed a 98% reduction in feeding, a significant increase in jumping behavior, and a significant tendency to abandon the leaves, thereby demonstrating repellency, not just an antixenosis response. Interactions between plant lines and insecticides influenced behavioral responses. All insecticides tested significantly reduced feeding durations on all cultivars except the preferred 'Yellow Pear'. However, nonfeeding activities such as walking, probing, resting, and jumping varied substantially with chemical and cultivar combination. The behavior assay results offered insight into host resistance mechanisms, provided a useful technique for measuring effects of interaction of plant lines with insecticides, and generated information for selecting insecticides for specific cultivars used in integrated pest management program for the tomato psyllid.  相似文献

The biological impact of consumption and utilization of celery was examined for the generalist herbivore, Spodoptera exigua (Hübner). An analysis of leaves and petioles demonstrated that both plant parts contained high levels of water (>85%), but that leaves contained twice as much total organic nitrogen as petioles. Developmental and reproductive rates were significantly higher (P<0.01, t-test) for larvae fed on leaves. Penultimate and final instars consumed significantly more (P0.05, t-test) dry weight of plant material when fed petioles as opposed to leaves. Although growth rates were similar regardless of food source for penultimate instars, conversion of food to body weight was lower for larvae limited to petioles. In the final instar, both growth and conversion rates were lower for larvae fed petioles than for those given leaves. Thus, since larvae exhibit preferences for the nutritionally less suitable petioles during later larval stages, environmental factors other than nutrition must influence movement and distribution of S., exigua in celery.
Résumé L'impact biologique de la consommation et de l'utilisation du céleri a été examiné sur un herbivore généraliste Spodoptera exigua Hübner. Une analyse de feuilles et de pétioles montre que ces deux parties de la plante ont une haute teneur en eau (>85%), mais que les feuilles contiennent deux fois plus d'azote organique total que les pétioles. Les taux de développement et de reproduction on été significativement plus élevés (<0,01 test t) pour les chenilles nourries sur feuilles. Les 2 derniers stades ont consommé significativement plus (P<0,05 test t) de poids sec de matière végétale en s'alimentant sur pétioles qu'en s'alimentant sur feuilles. Bien que les taux de croissance aient été voisins quel que soit l'aliment pour les avant-derniers stades, la conversion d'aliment en poids de corps était inférieure pour les chenilles ne consommant que des pétioles. Au dernier stade, tant les taux de croissance que de conversion des chenilles élevées sur pétioles étaient inférieurs à ceux des chenilles élevées sur feuilles. Ainsi, puisque les chenilles présentent une préférence pour les pétioles moins nutritifs au dernier stade larvaire, des paramètres écologiques autres que l'alimentation doivent influencer les mouvements et la distribution de S. exigua sur céleri.

A 4.0-kb BamHI-HindIII fragment encoding the cryIIA operon from the NRD-12 isolate of Bacillus thuringiensis subsp. kurstaki was cloned into Escherichia coli. The nucleotide sequence of the 2.2-kb AccI-HindIII fragment containing the NRD-12 cryIIA gene was identical to the HD-1 and HD-263 cryIIA gene sequences. Expression of cryIIA and subsequent purification of CryIIA inclusion bodies resulted in a protein with insecticidal activity against Heliothis virescens, Trichoplusia ni, and Culex quinquefasciatus but not Spodoptera exigua. The 4.0-kb BamII-HindIII fragment encoding the cryIIA operon was inserted into the B. thuringiensis-E. coli shuttle vector pHT3101 (pMAU1). pMAU1 was used to transform an acrystalliferous HD-1 strain of B. thuringiensis subsp. kurstaki and a leaf-colonizing strain of B. cereus (BT-8) by using electroporation. Spore-crystal mixtures from both transformed strains were toxic to H. virescens and T. ni but not Helicoverpa zea or S. exigua.  相似文献

The diet of adult and juvenile Weddell seals (Leptonychotes weddellii) in McMurdo Sound, Antarctica, was determined from both scat and stable isotope analyses, to ascertain if foraging behavior varied with age, season, or diving pattern. Scats were collected over 6 years and recovered hard parts identified. Stable carbon and nitrogen isotope values were determined for seal blood samples and potential prey items and used to identify primary prey species and assess trophic interactions. Pleuragramma antarcticum remains were recovered from between 70 and 100% of the scats, and there was little evidence for inter-annual or age-specific variation in foraging behavior. However, stable isotope and dive data analyses indicated that while most seals foraged predominantly on pelagic fish and squid, some juveniles concentrated on shallow benthic Trematomus spp. Combining these three methods permitted firm conclusions about diet and foraging behavior to be drawn.
