Nitrogen metabolism and senescence-associated changes during growth of carnation flowers 总被引:1,自引:0,他引:1
Nitrogen metabolism including nitrate reductase (EC 1.6.6.1), glutamate dehydroge-nase (EC 1.4.1.2) and glutamate-oxalacetate aminotransferase (EC 2.6.1.1) activities were studied during growth of petals taken from carnation flowers ( Dianthus caryophyllus L. cv. Sir Arthur) together with senescence parameters (lipid hydroper-oxides, soluble amino acids and permeability). A slight decline in nitrogen percentage on a dry weight basis was found together with a sharp decrease in nitrate reduct-ase, glutamate-oxalacetate aminotransferase and glutamate dehydrogenase activities during the maximum growth phase, which was characterized by increase in respiration, dry weight, length, organic nitrogen and DNA per petal. Changes generally associated with senescence, like lipid hydroperoxide and soluble ammo nitrogen accumulation and increases in permeability began to appear already during early growth. The results indicate that permeability and proteolysis may be closely related. The possible significance of the decrease in nitrogen percentage and enzyme activities during growth of petals is discussed. 相似文献

Copper is a potent sulfhydryl reagent which can also catalyse the generation of active oxygen. Since nitrate reductase (EC 1.6.6.1) is an SH-enzyme sensitive to oxidative environments, the relations among copper, active oxygen species and nitrate reductase (NR) activity are of interest. Foliar segments of wheat ( Triticum aestivum cv. Oasis) were floated on CuSO4 solutions (up to 250 μ M ) for 24 h under continuous light. Copper decreased NR activity before affecting active oxygen generation as estimated by changes in oxidative parameters, including malondialdehyde, K+ leakage and chlorophyll degradation. Cysteine and Na-benzoate counteracted this decrease, suggesting an oxidative damage of the enzyme in leaves exposed to high copper levels. Copper-induced NR inactivation was further studied in the partially purified enzyme. Preincubation with CuSO4 inhibited NR. Copper inhibition was reversed by subsequent incubation with EDTA, indicating that the metal bonded to key -SH groups of the enzyme. In addition, an ˙ OH-generating system (composed of CuSO4 , ascorbate and H2 O2 ) irreversibly decreased the activity of purified NR to a greater extent than copper alone. Our results show that copper affects nitrogen metabolism by diminishing NR activity, involving a direct effect on key SH-groups and an indirect effect via attack by active oxygen species induced by the metal. 相似文献

A method for the micropropagation of Lippia junelliana (Mold.) Tronc. from shoot tips or nodal segments was developed. Proliferating microshoot cultures were obtained by placing
shoot tips or nodal segments on full strength Murashige and Skoog medium (MS) supplemented with 4.4 μM benzyladenine (BA)
or 0.04 μM indolebutyric acid- (IBA) plus 4.4 μM BA. The rooting of shoots was better on full-strength MS medium without growth
regulators. Rooted plantlets were successfully acclimatized to soil. The shoot cultures showed a lower essential oil accumulation
in comparison with parent plants. Essential oil accumulation is closely related with growth and shows a negative correlation
with shoot proliferation.

The effect of O2 concentration on energy metabolism and senescence has been studied in cut flowers of Dianthus caryophyllus L. cv. Scania. As compared to the control (21% O2 ), 5% O2 delays flower senescence as well as decay of nucleotide level and AEC (adenylate energy charge). An atmosphere of 100% O2 accelerates senescence as well as the decrease of nucleotide level and AEC. While anoxia brings about a faster decrease of ATP and AEC than of total nucleotides, hyperoxia brings about a faster decrease in adenyl nucleotides than in ATP and AEC values. Petal oxygen uptake is over 90% of the maximal value under 4% O2 and saturates at 10% O2 . The development of senescence is dicussed as a two phase process (first phase-progressive and second phase-catastrophic) triggered by the action of hyperoxia, first on the system for energy utilization and later on the system for energy production, the degradation of which seems to be linked with increase in membrane permeability and withering. 相似文献

We used immunohistochemistry to quantify and compare the expression of Toll-like receptor 2 (TLR2) and cluster of differentiation 14 (CD14) in gingival tissues of both healthy individuals and patients with chronic periodontitis. We also correlated the expression of TLR2 and CD14 with the histological grades of chronic periodontitis. We examined 30 gingival specimens from chronic periodontitis patients and 10 from healthy individuals. Tissues from both groups were immunostained with antibodies against TLR2 and CD14. TLR2 and CD14 were expressed by endothelial cells, fibroblasts, lymphocytes and plasma cells. The immunohistochemical expression of TLR2 and CD14 was significantly greater in inflammatory cells of the chronic periodontitis group than in healthy individuals. Expression of these molecules was greater in the inflammatory cells of connective tissue adjacent to pocket epithelium in both groups. The expression of TLR2 and CD14 was greatest in the periodontitis group that was classified as severe grade, followed by moderate and mild grades, which suggests a role of TLR2 and CD14 in the pathogenesis of chronic periodontitis. The positive correlation of TLR2 and CD14 expression levels with the severity grades of chronic periodontitis suggests that they are correlated also with disease severity; therefore, they may be useful for predicting disease progression. We investigated the photoperiodic regulation of aging in Anagallis arvensis L. propagated by cuttings. Short-day (SD) conditions caused the persistence of juvenile forms and vegetative growth, as well as high rooting capacity and a high RNA content, through successive vegetative multiplications. SD + 1 long day (LD) every 15th day induced reproductive growth but allowed the persistence of a high rooting capacity and high RNA content through vegetative multiplications. LD conditions caused the persistence of the reproductive stage, the progressive loss of rooting capacity, and a decrease of RNA content. The longevity of the clones seems to be connected to the photoperiodic effects which induce indeterminate growth (SD) and/or determinate and reproductive growth. The loss of propagation capacity and the decrease of RNA content can be considered as indexes of aging not only in plants but also in clones. 相似文献

Effects of Oxidative Stress Caused by Oxygen and Hydrogen Peroxide on Energy Metabolism and Senescence in Oat Leaves 总被引:2,自引:0,他引:2
In attached oat leaves the levels of adenine nucleotides decreasedduring leaf development and senescence. However, the energycharge (EC) only decreased from 0.90 in 4-cm leaves to 0.80in senescent leaves. In detached leaves the levels of adeninenucleotides increased for 48 h, in association with an increasein RNase activity and a decrease in levels of RNA. The EC remainedhigh until late senescence when levels of adenine nucleotidesfell to about 30% of initial values. A decrease in energy parametersinduced by transfer from light to darkness and from high (21%)to low (0.5% and anoxia) concentrations of oxygen resulted inan increase in membrane permeability. Oxidative stress (above 0.5% O2 induced an increase in levelsof malondialdehyde (MDA) and then in permeability, associatedwith a decrease in levels of adenine nucleotides. Oxidativestress provoked by 0.05 and 0.10 M H2O2 caused a more rapiddecrease in energy parameters than O2. Under oxidative stress(above 0.5% O2) there is, first of all, an increase in membranepermeability and then a decrease in energy parameters, whichin turn are involved with senescence via increases in oxidationof membranes and degradation of energy-producing systems.
