Toxoplasma gondii is an obligate intracellular parasite that invades host cells, creating a parasitophorous vacuole where it communicates with the host cell cytosol through the parasitophorous vacuole membrane. The lytic cycle of the parasite starts with its exit from the host cell followed by gliding motility, conoid extrusion, attachment, and invasion of another host cell. Here, we report that Ca2+ oscillations occur in the cytosol of the parasite during egress, gliding, and invasion, which are critical steps of the lytic cycle. Extracellular Ca2+ enhances each one of these processes. We used tachyzoite clonal lines expressing genetically encoded calcium indicators combined with host cells expressing transiently expressed calcium indicators of different colors, and we measured Ca2+ changes in both parasites and host simultaneously during egress. We demonstrated a link between cytosolic Ca2+ oscillations in the host and in the parasite. Our approach also allowed us to measure two new features of motile parasites, which were enhanced by Ca2+ influx. This is the first study showing, in real time, Ca2+ signals preceding egress and their direct link with motility, an essential virulence trait. 相似文献

Giant reed (Arundo donax L.) and miscanthus (Miscanthus × giganteus Greef et Deu.) are two perennial rhizomatous grasses (PRGs), considered as promising sources of lignocellulosic biomass for renewable energy production. Although the agronomic performance of these species has been addressed by several studies, the literature dedicated to the crop water use of giant reed and miscanthus is still limited. Our objective was thus to investigate giant reed and miscanthus water use by assessing crop evapotranspiration (ETc), crop coefficients (Kc) and water use efficiency (WUE). The study was carried out in central Italy and specifically designed water-balance lysimeters were used to investigate the water use of these PRGs during the 2010 and 2011 growing seasons. Giant reed showed the highest cumulative evapotranspiration, with an average consumption of approximately 1100 mm, nearly 20% higher than miscanthus (900 mm). Crop evapotranspiration rates differed significantly between the species, particularly during the midseason (from June to September), when average daily ETc was 7.4 and 6.2 mm in giant reed and miscanthus respectively. The Kc values determined in our study varied from 0.4 to 1.9 for giant reed and 0.3 to 1.6 for miscanthus. Finally, WUE was higher in miscanthus than in giant reed, with average values of 4.2 and 3.1 g L−1 respectively. Further studies concerning water use under nonoptimal water conditions should be carried out and an assessment of the response to water stress of both crops is necessary to integrate the findings from this study. 相似文献

We compared genotypes at eight (AC)n microsatellite loci in domestic sheep
(Ovis aries) and wild Rocky Mountain bighorn sheep (O. canadensis). The
domestic sheep had greater genetic variation, higher allele-size variances,
and larger allele sizes than the wild sheep. Accumulating evidence from
higher taxonomic comparisons shows that these parameters are biased if
microsatellite loci are selected in one taxon and used in another. Our
results demonstrate similar biases between congeneric species. We compared
standard measures of genetic variation, differentiation, and distance
within and between species (H, D, FST) to newer measures based on
allele-size variance (SW, SB, RST). The size-based distances better
detected species-level divergence, but standard measures better
distinguished allopatric populations. Empirical calibration of these
measures at the subspecies level is needed to establish their useful
ranges.

Weta possess typical Ensifera ears. Each ear comprises three functional parts: two equally sized tympanal membranes, an underlying system of modified tracheal chambers, and the auditory sensory organ, the crista acustica. This organ sits within an enclosed fluid-filled channel–previously presumed to be hemolymph. The role this channel plays in insect hearing is unknown. We discovered that the fluid within the channel is not actually hemolymph, but a medium composed principally of lipid from a new class. Three-dimensional imaging of this lipid channel revealed a previously undescribed tissue structure within the channel, which we refer to as the olivarius organ. Investigations into the function of the olivarius reveal de novo lipid synthesis indicating that it is producing these lipids in situ from acetate. The auditory role of this lipid channel was investigated using Laser Doppler vibrometry of the tympanal membrane, which shows that the displacement of the membrane is significantly increased when the lipid is removed from the auditory system. Neural sensitivity of the system, however, decreased upon removal of the lipid–a surprising result considering that in a typical auditory system both the mechanical and auditory sensitivity are positively correlated. These two results coupled with 3D modelling of the auditory system lead us to hypothesize a model for weta audition, relying strongly on the presence of the lipid channel. This is the first instance of lipids being associated with an auditory system outside of the Odentocete cetaceans, demonstrating convergence for the use of lipids in hearing. 相似文献

Targeted genetic manipulation using homologous recombination is the method of choice for functional genomic analysis to obtain a detailed view of gene function and phenotype(s). The development of mutant strains with targeted gene deletions, targeted mutations, complemented gene function, and/or tagged genes provides powerful strategies to address gene function, particularly if these genetic manipulations can be efficiently targeted to the gene locus of interest using integration mediated by double cross over homologous recombination.Due to very high rates of nonhomologous recombination, functional genomic analysis of Toxoplasma gondii has been previously limited by the absence of efficient methods for targeting gene deletions and gene replacements to specific genetic loci. Recently, we abolished the major pathway of nonhomologous recombination in type I and type II strains of T. gondii by deleting the gene encoding the KU80 protein1,2. The Δku80 strains behave normally during tachyzoite (acute) and bradyzoite (chronic) stages in vitro and in vivo and exhibit essentially a 100% frequency of homologous recombination. The Δku80 strains make functional genomic studies feasible on the single gene as well as on the genome scale1-4.Here, we report methods for using type I and type II Δku80Δhxgprt strains to advance gene targeting approaches in T. gondii. We outline efficient methods for generating gene deletions, gene replacements, and tagged genes by targeted insertion or deletion of the hypoxanthine-xanthine-guanine phosphoribosyltransferase (HXGPRT) selectable marker. The described gene targeting protocol can be used in a variety of ways in Δku80 strains to advance functional analysis of the parasite genome and to develop single strains that carry multiple targeted genetic manipulations. The application of this genetic method and subsequent phenotypic assays will reveal fundamental and unique aspects of the biology of T. gondii and related significant human pathogens that cause malaria (Plasmodium sp.) and cryptosporidiosis (Cryptosporidium). 相似文献

The whale shark is an endangered species that usually feeds in coastal areas of highly productive seas such as the Gulf of California, Mexico. This study aims to describe the effect of sea surface temperature, chlorophyll a, bathymetry and slope on the habitat suitability of whale sharks in three important aggregation sites of the Gulf of California. A total of 2396 records of occurrence of whale sharks were obtained from international databases and scientific literature between 1996 and 2018. These records were used for the creation of a species distribution model using MaxEnt for each of the three aggregation sites. The concentration of chlorophyll a explained 71% of the habitat suitability, followed by bathymetry and slope with a combined 17%, and sea surface temperature constituting 10% of the model. Habitat suitability was related to areas where nontargeted fisheries may impact whale sharks through bycatch, entanglement and ship strikes. The implications for the conservation of whale sharks should be considered for management decisions in terms of marine protected areas, fishing refugees or bans, and other regulations regarding fisheries activities. 相似文献
