A natural antibacterial-substance-producing gram-positive bacterium was isolated from terasi shrimp paste, a popular fermented product in Indonesia. This strain, a spore-forming and strictly aerobic bacterium, was identified as Virgibacillus salexigens by 16S rRNA gene sequence analysis. The antibacterial substance purified from the precipitated product in the culture supernatant of the strain using ammonium sulfate showed a broad inhibition spectrum against gram-positive bacteria, including a typical foodborne bacterium, namely, Listeria monocytogenes. The antibacterial activity of the substance was inactivated by treatments with various proteolytic enzymes. It was stable after heating or pH treatment, and approximately 60 % of the initial activity remained even after heating at 121 °C for 15 min. In addition, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis indicated that its monoisotopic mass weight was 5318.4 Da (M+H)+. On the basis of the results obtained by the automated Edman degradation technique and MALDI-TOF MS analysis, the substance can be classified as a member of Class IId bacteriocins, but it could not be identified as any of the previously purified substances except for the putative bacteriocin predicted from the draft genome sequence data of gram-positive bacteria such as Virgibacillus and Bacillus strains. 相似文献

Extracts of the Canadian lowbush blueberry (Vaccinium angustifolium Ait.) have recently been demonstrated to possess significant antidiabetic potential, in accordance with the traditional use of this plant as an antidiabetic natural health product. Fermentation of blueberry juice with the Serratia vaccinii bacterium is known to modify the phenolic content and increase antioxidant activity. The present study evaluated the effects of fermented blueberry juice on glucose uptake, adipogenesis, and the signaling pathways that regulate glucose transport in muscle cells and adipocytes. A 6-hour treatment with fermented juice potentiated glucose uptake by 48% in C2C12 myotubes and by 142% in 3T3-L1 adipocytes, in the presence or absence of insulin, whereas nonfermented juice had no effect on transport. Fermented juice dramatically inhibited triglyceride content during adipogenesis of 3T3-L1 cells. Chlorogenic acid and gallic acid, both major phenolic components of fermented juice, had no effect on glucose uptake. Western blot analysis of the insulin-independent AMP-activated protein kinase revealed increased phosphorylation resulting from a 6-hour treatment. This activation or the increase in glucose uptake could not be explained by increased cytosolic calcium. Fermentation with S. vaccinii is concluded to confer antidiabetic activities to blueberry juice. Although the active principles and their mechanisms of action remain to be identified, transformed blueberry juice may nevertheless represent a novel complementary therapy and a source of novel therapeutic agents against diabetes mellitus. 相似文献

"Probiotics: From Bench to Market" was a one-day conference convened by the New York Academy of Sciences on June 11, 2010, with the goal of stimulating discussion of the physiological effects of probiotics on the gastrointestinal, nervous, and immune systems. The program included speakers from academia, industry, and government to give conference participants a full understanding of the state of the field of probiotics. Streptomyces viridosporus T7A and S. badius 252 were grown in 1 to 2% (wt/vol) slurry cultures with mineral salts solution containing 0.6% yeast extract and 100/200 mesh ground and extracted corn lignocellulose at 37°C. Enzyme activities rapidly increased in the first 3 to 4 days and then declined and remained at a relatively constant level. Concentrations of endoglucanase and xylanase produced by S. badius were lower than those produced by S. viridosporus. However, the lignin-peroxidase peak concentration was threefold higher than with S. viridosporus and was obtained at 9 to 10 days of incubation. By polyacrylamide gel analysis, it was determined that peroxidases from both species consisted of four enzymes, with only one, the lignin peroxidase, having high activity. A culture pH of 8.5 was preferable for lignocellulose degradation by S. badius. 相似文献

Technology has routinely been used in zoos to enhance the visitor experience, for public education and for conservation awareness. However, studies into technology-aided exhibits in zoos have thus far been limited to zoos in western society. This article reports on an Indonesian zoo deploying new technology in its exhibits and how visitors interacted with these exhibits. A case study guided both an investigation of the visitors' technology usage of zoo exhibits and zoo implementation of technological processes. Interviews with visitors, zoo staff, and managers were conducted and analyzed and these were supplemented with on-site observations. The results indicated that the majority of zoo visitors did not use the technological applications provided by the zoo. In addition, there was found to be a significant lack of staff engagement in the consultation, design, implementation, and maintenance of the technology. Correlations were found between visitors' level of technology usage, their reading of signage, their day-to-day technology usage, and their conservation knowledge. The findings suggest that an understanding of visitor' behavior, active staff engagement in the development and implementation of new technology, in addition to emotion-inducing interpretation, and purposefully guided interactions, could increase the use of technology at a zoo and thus enhance positive visitor engagement, ultimately resulting in heightened educational as well as conservational outcomes. 相似文献

The rapidly evolving field of photoacoustic tomography utilizes endogenous chromophores to extract both functional and structural information from deep within tissues. It is this power to perform precise quantitative measurements in vivo—with endogenous or exogenous contrast—that makes photoacoustic tomography highly promising for clinical translation in functional brain imaging, early cancer detection, real-time surgical guidance, and the visualization of dynamic drug responses. Considering photoacoustic tomography has benefited from numerous engineering innovations, it is of no surprise that many of photoacoustic tomography’s current cutting-edge developments incorporate advances from the equally novel field of artificial intelligence. More specifically, alongside the growth and prevalence of graphical processing unit capabilities within recent years has emerged an offshoot of artificial intelligence known as deep learning. Rooted in the solid foundation of signal processing, deep learning typically utilizes a method of optimization known as gradient descent to minimize a loss function and update model parameters. There are already a number of innovative efforts in photoacoustic tomography utilizing deep learning techniques for a variety of purposes, including resolution enhancement, reconstruction artifact removal, undersampling correction, and improved quantification. Most of these efforts have proven to be highly promising in addressing long-standing technical obstacles where traditional solutions either completely fail or make only incremental progress. This concise review focuses on the history of applied artificial intelligence in photoacoustic tomography, presents recent advances at this multifaceted intersection of fields, and outlines the most exciting advances that will likely propagate into promising future innovations. 相似文献
