Colony growth of three Fusarium spp. on potato dextrose agar was followed by collecting near-infrared (NIR) hyperspectral images of the colonies at regular intervals after inoculation up to 55?h. After principal component analysis (PCA), two clusters were apparent in the score plot along principal component 1. Using the brushing technique, these clusters were divided into four groups of pixels with similar score values. These could be visualised as growth zones within the colonies in the corresponding score image. Three spectral bands, i.e. 1,166, 1,380 and 1,918?nm, were prominent in the multiplicative scatter corrected and Savitzky?CGolay second derivative spectra. These indicated chemical changes, associated with carbohydrates (1,166 and 1,380?nm) and protein (1,918?nm), that occurred as the mycelium grew and matured. The protein band was more prominent in the mature fungal material while the carbohydrate band was less pronounced. The younger material and the agar were characterised by the carbohydrate spectral band. Integrating whole mycelium colonies as the sum of pixels over time made it possible to construct curves that resembled growth curves; this included the lag phase, active growth phase, deceleration phase and phase of constant growth. Growth profiles constructed from individual growth zones indicated more detailed growth characteristics. The use of NIR hyperspectral imaging and multivariate image analysis (MIA) allowed one to visualise radial growth rings in the PCA score images. This would not have been possible with bulk spectroscopy. Interpreting spectral data enabled better understanding of microbial growth characteristics on agar medium. NIR hyperspectral imaging combined with MIA is a powerful tool for the evaluation of growth characteristics of fungi.  相似文献

Competition has negative effects on population size and also drives ecological character displacement, that is, evolutionary divergence to utilize different portions of the resource spectrum. Many species undergo an annual cycle composed of a lean season of intense competition for resources and a breeding season. We use a quantitative genetic model to study the effects of differential reproductive output in the summer or breeding season on character displacement in the winter or nonbreeding season. The model is developed with reference to the avian family of Old World leaf warblers (Phylloscopidae), which breed in the temperate regions of Eurasia and winter in tropical and subtropical regions. Empirical evidence implicates strong winter density-dependent regulation driven by food shortage, but paradoxically, the relative abundance of each species appears to be determined by conditions in the summer. We show how population regulation in the two seasons becomes linked, with higher reproductive output by one species in the summer resulting in its evolution to occupy a larger portion of niche space in the winter. We find short-term ecological processes and longer-term evolutionary processes to have comparable effects on a species population size. This modeling approach can also be applied to other differential effects of productivity across seasons.  相似文献

Frankliniella fusca (Hinds) naturally disperses from winter weeds to crops in spring, causing direct and indirect damage. Field preparation before planting includes use of herbicides or cultivation to kill unwanted vegetation, which adversely affects F. fusca host plants and potentially influences F. fusca dispersal. Common chickweed, Stellaria media (L.), infested with F. fusca, was used as a model to study effects of timing and type of vegetation management on adult dispersal. Infested weeds were caged and F. fusca weekly dispersal was monitored using sticky traps. Weed management treatments performed at an early (14 April-11 May) or late (2 wk after early treatment) date consisted of glyphosate, paraquat, disking, hoeing, or untreated control. Late glyphosate and hoeing treatments resulted in cumulative dispersal statistically similar to or greater than from control plots. Compared with the control, significantly more F. fusca dispersed from the glyphosate and hoeing plots during the 3 wk after treatment. More thrips dispersed from the late paraquat treatment 1 wk post-application than from the control. Dispersal from the disked treatment and early paraquat treatment was similar to that of the control 1- to 3-wk post-treatment. Early treatments resulted in significantly smaller cumulative dispersal than the control in all but one instance. Late disking and paraquat treatments resulted in cumulative F. fusca captures that were statistically similar or less than that in the control. Winter weed management type and timing affect F. fusca dispersal magnitude and duration.  相似文献

The use of enzymatically isolated cardiac myocytes is ubiquitous in modern cardiovascular research. Parallels established between cardiomyocyte shortening responses and those of intact tissue make the cardiomyocyte an invaluable experimental model of cardiac function. Much of our understanding regarding the fundamental processes underlying heart function is owed to our increasing capabilities in single-cell stimulation and direct or indirect observation, as well as quantitative analysis of such cells. Of the many important mechanisms and functions that can be readily assessed in cardiomyocytes at all stages of development, contractility is the most representative and one of the most revealing. The purpose of this review is to provide a survey of various methodological approaches in the literature used to assess adult and neonatal cardiomyocyte contractility. The various methods employed to evaluate the contractile behavior of enzymatically isolated mammalian cardiac myocytes can be conveniently divided into two general categories??those employing optical (image)-based systems and those that use transducer-based technologies. This survey is by no means complete, but we have made an effort to include the most popular methods in terms of reliability and accessibility. These techniques are in constant evolution and hold great promise for the next generation of breakthrough studies in cell biology for the prevention, treatment, and cure of cardiovascular diseases.  相似文献

There are many examples of ecological theory informing fishery management. Yet fisheries also provide tremendous opportunities to test ecological theory through large-scale, repeated, and well-documented perturbations of natural systems. Although treating fisheries as experiments presents several challenges, few comparable tests exist at the ecosystem scale. Experimental manipulations of fish populations in lakes have been widely used to develop and test ecological theory. Controlled manipulation of fish populations in open marine systems is rarely possible, but fisheries data provide a valuable substitute for such manipulations. To highlight the value of marine fisheries data, we review leading ecological theories that have been empirically tested using such data. For example, density dependence has been examined through meta-analysis of spawning stock and recruitment data to show that compensation (higher population growth) occurs commonly when populations are reduced to low levels, while depensation (the Allee effect) is rare. As populations decline, spatial changes typically involve populations contracting into high-density core habitats while abandoning less productive habitats. Fishing down predators may result in trophic cascades, possibly shifting entire ecosystems into alternate stable states, although alternate states can be maintained by both ecological processes and continued fishing pressure. Conversely, depleting low trophic level groups may affect central-place foragers, although these bottom–up effects rarely appear to impact fish—perhaps because many fish populations have been reduced to the point that they are no longer prey limited. Fisheries provide empirical tests for diversity–stability relations: catch data suggest that more diverse systems recover faster and provide more stable returns than less diverse systems. Fisheries have also provided examples of the tragedy of the commons, as well as counter-examples where common property resources have been managed successfully. We also address two barriers to use of fisheries data to answer ecological questions: differences in terminology for similar concepts and misuse of records of fishery landings (catch data) as a proxy for biomass trends.  相似文献

Concerns over fishing impacts on marine populations and ecosystems have intensified the need to improve ocean management. One increasingly popular market-based instrument for ecological stewardship is the use of certification and eco-labeling programs to highlight sustainable fisheries with low environmental impacts. The Marine Stewardship Council (MSC) is the most prominent of these programs. Despite widespread discussions about the rigor of the MSC standards, no comprehensive analysis of the performance of MSC-certified fish stocks has yet been conducted. We compared status and abundance trends of 45 certified stocks with those of 179 uncertified stocks, finding that 74% of certified fisheries were above biomass levels that would produce maximum sustainable yield, compared with only 44% of uncertified fisheries. On average, the biomass of certified stocks increased by 46% over the past 10 years, whereas uncertified fisheries increased by just 9%. As part of the MSC process, fisheries initially go through a confidential pre-assessment process. When certified fisheries are compared with those that decline to pursue full certification after pre-assessment, certified stocks had much lower mean exploitation rates (67% of the rate producing maximum sustainable yield vs. 92% for those declining to pursue certification), allowing for more sustainable harvesting and in many cases biomass rebuilding. From a consumer's point of view this means that MSC-certified seafood is 3-5 times less likely to be subject to harmful fishing than uncertified seafood. Thus, MSC-certification accurately identifies healthy fish stocks and conveys reliable information on stock status to seafood consumers.  相似文献

The expansion of HIV antiretroviral therapy into decentralized rural settings will increasingly require simple point-of-care (POC) diagnostic tests that can be used without laboratory infrastructure and technical skills. New POC test devices are becoming available but decisions around which technologies to deploy may be biased without systematic assessment of their suitability for decentralized healthcare settings. To address this, we developed a standardized, quantitative scorecard tool to objectively evaluate the operational characteristics of POC diagnostic devices. The tool scores devices on a scale of 1–5 across 30 weighted characteristics such as ease of use, quality control, electrical requirements, shelf life, portability, cost and service, and provides a cumulative score that ranks products against a set of ideal POC characteristics. The scorecard was tested on 19 devices for POC CD4 T-lymphocyte cell counting, clinical chemistry or hematology testing. Single and multi-parameter devices were assessed in each of test categories. The scores across all devices ranged from 2.78 to 4.40 out of 5. The tool effectively ranked devices within each category (p<0.01) except the CD4 and multi-parameter hematology products. The tool also enabled comparison of different characteristics between products. Agreement across the four scorers for each product was high (intra-class correlation >0.80; p<0.001). Use of this tool enables the systematic evaluation of diagnostic tests to facilitate product selection and investment in appropriate technology. It is particularly relevant for countries and testing programs considering the adoption of new POC diagnostic tests.  相似文献

The oxidative pathway for the metabolism of tryptophan along the kynurenine pathway generates quinolinic acid, an agonist at N-methyl-D-aspartate receptors, as well as kynurenic acid which is an antagonist at glutamate and nicotinic receptors. The pathway has become recognized as a key player in the mechanisms of neuronal damage and neurodegenerative disorders. As a result, manipulation of the pathway, so that the balance between the levels of components of the pathway can be modified, has become an attractive target for the development of pharmacological agents with the potential to treat those disorders. This review summarizes some of the relevant background information on the pathway itself before identifying some of the chemical strategies for its modification, with examples of their successful application in animal models of infection, stroke, traumatic brain damage, cerebral malaria and cerebral trypanosomiasis.  相似文献
