One of the main competing theories to describe the evolution of recombination is the Red Queen hypothesis (RQH). Presently, many theoretical analyses of the RQH typically examine fitness interactions in host-parasite frameworks. Less emphasis has been placed on understanding the impact of host ploidy in these systems. In this study, we look to investigate the high observed rates of recombination observed in two common haplodiploid species (Apis mellifera and Bombus terrestris). We compared haplodiploid to diploid host populations under infection with haploid asexual parasites, using a matching allele model. Results from a simulation analysis showed that the Red Queen does not run in haplodiploid hosts and is therefore, probably not responsible for the high recombination rates observed so far in haplodiploid hosts.  相似文献

Molecular understanding of actin dynamics requires a genetically traceable model system that allows live cell imaging together with high-resolution microscopy techniques. Here, we used Drosophila pupal macrophages that combine many advantages of cultured cells with a genetic in vivo model system. Using structured illumination microscopy together with advanced spinning disk confocal microscopy we show that these cells provide a powerful system for single gene analysis. It allows forward genetic screens to characterize the regulatory network controlling cell shape and directed cell migration in a physiological context. We knocked down components regulating lamellipodia formation, including WAVE, single subunits of Arp2/3 complex and CPA, one of the two capping protein subunits and demonstrate the advantages of this model system by imaging mutant macrophages ex vivo as well as in vivo upon laser-induced wounding.  相似文献

Excessive exposure to alcohol prenatally has a myriad of detrimental effects on the health and well-being of the offspring. It is unknown whether chronic low-moderate exposure of alcohol prenatally has similar and lasting effects on the adult offspring’s health. Using our recently developed Sprague-Dawley rat model of 6% chronic prenatal ethanol exposure, this study aimed to determine if this modest level of exposure adversely affects glucose homeostasis in male and female offspring aged up to eight months. Plasma glucose concentrations were measured in late fetal and postnatal life. The pancreas of 30 day old offspring was analysed for β-cell mass. Glucose handling and insulin action was measured at four months using an intraperitoneal glucose tolerance test and insulin challenge, respectively. Body composition and metabolic gene expression were measured at eight months. Despite normoglycaemia in ethanol consuming dams, ethanol-exposed fetuses were hypoglycaemic at embryonic day 20. Ethanol-exposed offspring were normoglycaemic and normoinsulinaemic under basal fasting conditions and had normal pancreatic β-cell mass at postnatal day 30. However, during a glucose tolerance test, male ethanol-exposed offspring were hyperinsulinaemic with increased first phase insulin secretion. Female ethanol-exposed offspring displayed enhanced glucose clearance during an insulin challenge. Body composition and hepatic, muscle and adipose tissue metabolic gene expression levels at eight months were not altered by prenatal ethanol exposure. Low-moderate chronic prenatal ethanol exposure has subtle, sex specific effects on glucose homeostasis in the young adult rat. As aging is associated with glucose dysregulation, further studies will clarify the long lasting effects of prenatal ethanol exposure.  相似文献

Kidney transplantation is the treatment of choice for patients suffering from end-stage renal disease. It offers better life expectancy and higher quality of life when compared to dialysis. Although the last few decades have seen major improvements in patient outcomes following kidney transplantation, the increasing shortage of available organs represents a severe problem worldwide. To expand the donor pool, marginal kidney grafts recovered from extended criteria donors (ECD) or donated after circulatory death (DCD) are now accepted for transplantation. To further improve the postoperative outcome of these marginal grafts, research must focus on new therapeutic approaches such as alternative preservation techniques, immunomodulation, gene transfer, and stem cell administration.Experimental studies in animal models are the final step before newly developed techniques can be translated into clinical practice. Porcine kidney transplantation is an excellent model of human transplantation and allows investigation of novel approaches. The major advantage of the porcine model is its anatomical and physiological similarity to the human body, which facilitates the rapid translation of new findings to clinical trials. This article offers a surgical step-by-step protocol for an autotransplantation model and highlights key factors to ensure experimental success. Adequate pre- and postoperative housing, attentive anesthesia, and consistent surgical techniques result in favorable postoperative outcomes. Resection of the contralateral native kidney provides the opportunity to assess post-transplant graft function. The placement of venous and urinary catheters and the use of metabolic cages allow further detailed evaluation. For long-term follow-up studies and investigation of alternative graft preservation techniques, autotransplantation models are superior to allotransplantation models, as they avoid the confounding bias posed by rejection and immunosuppressive medication.  相似文献

In this study, step variations in temperature, pH, and carbon substrate feeding rate were performed within five high cell density Escherichia coli fermentations to assess whether intraexperiment step changes, can principally be used to exploit the process operation space in a design of experiment manner. A dynamic process modeling approach was adopted to determine parameter interactions. A bioreactor model was integrated with an artificial neural network that describes biomass and product formation rates as function of varied fed‐batch fermentation conditions for heterologous protein production. A model reliability measure was introduced to assess in which process region the model can be expected to predict process states accurately. It was found that the model could accurately predict process states of multiple fermentations performed at fixed conditions within the determined validity domain. The results suggest that intraexperimental variations of process conditions could be used to reduce the number of experiments by a factor, which in limit would be equivalent to the number of intraexperimental variations per experiment. © 2016 American Institute of Chemical Engineers Biotechnol. Explanations for the coexistence of multiple species from the same functional group or taxonomic clade frequently include fine‐scale resource partitioning. However, despite the hypothesized importance of niche partitioning, we know relatively little about the underlying mechanisms. For example, differences in resource use may be fixed consequences of organism traits, or they may be achieved via context‐dependent behaviors. In this study we investigated mechanisms of microhabitat partitioning using eight species of marine mesograzers inhabiting seagrass and algae habitats, using laboratory trials to measure microhabitat use in the presence and absence of both predators and competitors. We found clear evidence for microhabitat partitioning between the species, which account for over 60% of the mesograzers commonly found in this system and vary in both body size and the ability to build tubes on habitat substrates. Species‐specific microhabitat use was poorly predicted by these two traits, but remained remarkably consistent across contexts. Habitat use was not affected by the presence of fish predators common in this system, even though predation pressure is thought to place strong constraints on microhabitat in communities of plant‐associated arthropods. The presence of competing species also did not affect the relative separation of microhabitat use. Behavioral responses to potential competitors did cause significant changes in microhabitat use in all of the smallest species, but these changes did not depend on competitor identity and were relatively small compared to among‐species patterns of microhabitat partitioning. The consistency of species‐specific microhabitat use, regardless of the presence of predators or competitors, should make coexistence most likely among species that differ in these choices. For these species, it appears that the benefits accrued from their selected microhabitats are not affected by species interactions, or that any benefits of alternative microhabitat use are outweighed by risks associated with movement.  相似文献

Pyroptosis is a unique, pro‐inflammatory form of lytic cell death that is initiated by the activation of inflammatory caspases. The caspase substrate gasdermin D (GSDMD) plays a critical function in pyroptosis, yet the precise mode of action of this molecule in cell death execution remained unclear. Several recent reports, including a The EMBO Journal article, show that the caspase‐matured N‐terminal fragment of GSDMD is recruited to lipid membranes to form pore‐like structures, which constitutes the key effector mechanism of pyroptotic cell death.  相似文献
