Schizophrenia is a devastating neuropsychiatric disorder with genetically complex traits. Genetic variants should explain a considerable portion of the risk for schizophrenia, and genome-wide association study (GWAS) is a potentially powerful tool for identifying the risk variants that underlie the disease. Here, we report the results of a three-stage analysis of three independent cohorts consisting of a total of 2,535 samples from Japanese and Chinese populations for searching schizophrenia susceptibility genes using a GWAS approach. Firstly, we examined 115,770 single nucleotide polymorphisms (SNPs) in 120 patient-parents trio samples from Japanese schizophrenia pedigrees. In stage II, we evaluated 1,632 SNPs (1,159 SNPs of p<0.01 and 473 SNPs of p<0.05 that located in previously reported linkage regions). The second sample consisted of 1,012 case-control samples of Japanese origin. The most significant p value was obtained for the SNP in the ELAVL2 [(embryonic lethal, abnormal vision, Drosophila)-like 2] gene located on 9p21.3 (p = 0.00087). In stage III, we scrutinized the ELAVL2 gene by genotyping gene-centric tagSNPs in the third sample set of 293 family samples (1,163 individuals) of Chinese descent and the SNP in the gene showed a nominal association with schizophrenia in Chinese population (p = 0.026). The current data in Asian population would be helpful for deciphering ethnic diversity of schizophrenia etiology.  相似文献

Thy28 protein is conserved among plants, bacteria, and mammalian cells. Nuclear Thy28 protein is substantially expressed in testis, liver, and immune cells such as lymphocytes. Lymphocyte apoptosis plays a crucial role in homeostasis and formation of a diverse lymphocyte repertoire. In this study, we examined whether Thy28 affects induction of apoptosis in WEHI-231 B lymphoma cells following engagement of membrane immunoglobulin (mIg). Once they were established, the Thy28-overexpressing WEHI-231 cells showed similar expression levels of IgM and class I major histocompatibility complex (MHC) molecule compared with controls. The Thy28-overexpressing cells were considerably resistant to loss of mitochondrial membrane potential (ΔΨm), caspase-3 activation, and increase in annexin-positive cells upon mIg engagement. These changes were concomitant with an increase in G1 phase associated with upregulation of p27Kip1. The anti-IgM-induced sustained activation of c-Jun N-terminal kinase (JNK), which was associated with late-phase hydrogen peroxide (H2O2) production, was partially reduced in the Thy28-expressing cells relative to controls. Taken together, the data suggest that in WEHI-231 B lymphoma cells, Thy28 regulates mIg-mediated apoptotic events through the JNK-H2O2 activation pathway, concomitant with an accumulation of cells in G1 phase associated with upregulation of p27Kip1 in WEHI-231 B lymphoma cells.  相似文献

Homoplasmic m.1624C>T mutation of the mitochondrial tRNAVal gene was previously demonstrated to cause fatal neonatal Leigh syndrome. Here, we report the clinical phenotypes of a Japanese male and his mother with heteroplasmic m.1624C>T mutation. The 36-year-old male presented with repeated episodes of consciousness disturbance since the age of 25, cognitive decline, and personality change. Cerebrospinal fluid levels of lactate and pyruvate were elevated. His mother showed similar symptoms and course. The mutation m.1624C>T was identified heteroplasmically in the proband's muscle and leukocytes and in the mother's leukocytes. The heteroplasmy load decreased with age.  相似文献

Recent evidence suggests that insulin induces hydrolysis of phosphatidylinositol-glycan (PI-G) and releases inositol-glycan (IG) and diacylglycerol (DAG). These two mediators are speculated to mediate different insulin actions. In this study, we examined metabolic labeling of PI-G in BC3H-1 myocytes with known precursors of PI-G. PI-G was metabolically labeled with [3H]myo-inositol, [3H]glucosamine, [3H]galactose, [3H]glycerol, and [3H]myristic acid. The treatment of 3H-labeled PI-G with phosphatidylinositol-specific phospholipase C liberated [3H]myo-inositol, [3H]glucosamine, or [3H]galactosamine-labeled IgGs, and [3H]glycerol or [3H]myristic acid-labeled DAG. In BC3H-1 myocytes, insulin induced phosphodiesteratic hydrolysis of PI-G and stimulated generation of IGs and DAG. Released IGs were labeled with [3H]myo-inositol, [3H]glucosamine, and [3H]galactose. Released DAG was labeled with [3H] glycerol and [3H]myristic acid. The IG had a dose-dependent insulin-like activity on glucose oxidation and lipogenesis without affecting glucose transport in rat adipocytes. Insulin increased 3H radioactivities of IG and insulin-mimicking activities of IG. These results provided further evidence that hydrolysis of PI-G and generation of IGs and DAG might be early steps in some insulin actions.  相似文献

Abstract Effects of organic amendments and alterations of environmental conditions on the inoculum potential of Fusarium oxysporum f. sp. raphani PEG-4 (PEG-4), estimated from its population dynamics and spore germinability, were investigated using soils suppressive (S-soil) and conducive (C-soil) to Fusarium-wilt of radish. The results on the population dynamics of PEG-4 in S- and C-soils showed that the germination-lysis mechanism, proposed by Chinn and Ledingham (Can. J. Bot. (1962) 39, 739–748), seemed to be applicable to S-soil, while not application to C-soil. Germination of PEG-4 microconidia in soils supplemented with glucose and asparagine (1 mg g−1 of soil) was higher in C-soil than in S-soil, showing that S-soil possessed a greater degree of fungistasis than C-soil. Organic amendments, especially rice straw and fresh radish residue (FRR), brought about suppressive effects on the germination of PEG-4 in both soils along with their decomposition. These results suggest that the autecology of PEG-4 in S- and C-soils was quite different depending on incubation conditions, and presence or absence of organic amendments.  相似文献

Background

Transmission of tuberculosis (TB) to health care workers (HCWs) is a global issue. Although effective infection control measures are expected to reduce nosocomial TB, HCWs'' infection has not been assessed enough in TB high burden countries. We conducted a cross-sectional study to determine the prevalence of TB infection and its risk factors among HCWs in Hanoi, Viet Nam.

Methodology/Principal Findings

A total of 300 HCWs including all staff members in a municipal TB referral hospital received an interferon-gamma release assay (IGRA), QuantiFERON-TB Gold In-TubeTM, followed by one- and two-step tuberculin skin test (TST) and a questionnaire-based interview. Agreement between the tests was evaluated by kappa statistics. Risk factors for TB infection were analyzed using a logistic regression model. Among the participants aged from 20 to 58 years (median = 40), prevalence of TB infection estimated by IGRA, one- and two-step TST was 47.3%, 61.1% and 66.3% respectively. Although the levels of overall agreement between IGRA and TST were moderate, the degree of agreement was low in the group with BCG history (kappa = 0.29). Working in TB hospital was associated with twofold increase in odds of TB infection estimated by IGRA. Increased age, low educational level and the high body mass index also demonstrated high odds ratios of IGRA positivity.

Conclusions/Significance

Prevalence of TB infection estimated by either IGRA or TST is high among HCWs in the hospital environment for TB care in Viet Nam and an infection control program should be reinforced. In communities with heterogeneous history of BCG vaccination, IGRA seems to estimate TB infection more accurately than any other criteria using TST.  相似文献
