Studying Pneumocystis has proven to be a challenge from the perspective of propagating a significant amount of the pathogen in a facile manner. The study of several fungal pathogens has been aided by the use of invertebrate model hosts. Our efforts to infect the invertebrate larvae Galleria mellonella with Pneumocystis proved futile since P. murina neither caused disease nor was able to proliferate within G. mellonella. It did, however, show that the pathogen could be rapidly cleared from the host.  相似文献

Abstract Plasmid-containing strains of Clostridium acetobutylicum produced higher levels of solvents and lower levels of acids than wild-type cells in controlled pH 4.5 batch fermentations. This effect was observed regardless of whether or not the plasmids contained C. acetobutylicum genes. The effect was less prevalent in higher pH fermentations and apparently independent of the actual DNA sequences contained on these plasmids. The plasmid-containing strains were found to have lower growth-rates and higher solventogenic enzyme activities than wild-type cells. However, similar activity levels were found for both butyrate-pathway enzymes.  相似文献

This work addresses an unmet clinical need, that of glaucoma monitoring through intraocular pressure (IOP) interrogation in patients with artificial corneas (keratoprost hesis). We demonstrate direct integration of a low‐drift, fiber‐optic Fabry‐Perot pressure sensor embedded in the keratoprosthesis via rapid, non‐contact micromagnetic fiber alignment. IOP interrogation is achieved by using white‐light interferometry, which provides sub‐mmHg IOP sensitivity. Wild boar (Sus scrofa L.) were introduced in the island of Cyprus in 1990, when five animals were imported from Greece for game farming. In 1994, wild boars were illegally released in Lemesos (Limassol) Forest and in 1996 in the Troodos National Forest Park. Soon the population increased and dispersed throughout the park. In 1997, the government of Cyprus decided to eradicate wild boar because of the danger of transmitting diseases to livestock and to prevent possible environmental destruction. To control the wild boar, hunting was permitted and game wardens were instructed to eliminate the free-ranging animals. In 2004, no animals were observed in localities where they had been seen before. Surveys in September 2004 (Troodos National Park) and January/February 2005 (Troodos Forest, Pafos Forest, and Lemesos Forest) revealed no signs of recent wild boar presence. The reasons for the possible failure of wild boar to establish in Cyprus are discussed.  相似文献

Artificial cornea is an effective treatment of corneal blindness. Yet, intraocular pressure (IOP) measurements for glaucoma monitoring remain an urgent unmet need. Here, we present the integration of a fiber‐optic Fabry‐Perot pressure sensor with an FDA‐approved keratoprosthesis for real‐time IOP measurements using a novel strategy based on optical‐path self‐alignment with micromagnets. Additionally, an alternative noncontact sensor‐interrogation approach is demonstrated using a bench‐top optical coherence tomography system. We show stable pressure readings with low baseline drift (<2.8 mm Hg) for >4.5 years in vitro and efficacy in IOP interrogation in vivo using fiber‐optic self‐alignment, with good initial agreement with the actual IOP. Subsequently, IOP drift in vivo was due to retroprosthetic membrane (RPM) formation on the sensor secondary to surgical inflammation (more severe in the current pro‐fibrotic rabbit model). This study paves the way for clinical adaptation of optical pressure sensors with ocular implants, highlighting the importance of controlling RPM in clinical adaptation.  相似文献
