Physarum polycephalum expresses a membrane-bound beta-glucosidase (BglM1) with a molecular mass of 130 kDa. The primary structure of BglM1 consists of a glycosyl hydrolase family 3 domain at an amino-terminal domain and a carboxyl-terminal region without homology to the sequence of known glycosidases. The latter region contains two calx-beta motifs known as Ca(2+)-binding sites; an RGD sequence, which is known to be a cell attachment sequence; and a transmembrane region. The molecular mass calculated from the amino acid sequence is 130 kDa, but that in the crude extract was estimated by SDS-PAGE to be 230 kDa, and decreased to 130 kDa during purification. However, when BglM1 was purified in the presence of calcium ion, the molecular mass remained 230 kDa. The biochemical characteristics of the 130- and 230-kDa BglM1 forms were analyzed: differences were found in the kinetic data for some substrates specific for both these enzymes; however, no difference was found in their intrinsic characteristics such as optimum pH and temperature. In addition, the molecular mass of native BglM1 with a calcium ion was estimated to be 1,000 kDa or larger by gel filtration. These results suggest that the calcium ion influences the conformation of BglM1. The evidence that BglM1 localizes on the plasma membrane of plasmodia was confirmed using immunofluorescence microscopy. Although Physarum BglM1 was expressed in microplasmodia and plasmodia, little expression was detected in other stages. BglM1 may have some function only in multinuclear cells.  相似文献

The mechanisms whereby the parathyroid hormone (PTH) exerts its anabolic action on bone are incompletely understood. We previously showed that inhibition of ERK1/2 enhanced Smad3-induced bone anabolic action in osteoblasts. These findings suggested the hypothesis that changes in gene expression associated with the altered Smad3-induced signaling brought about by an ERK1/2 inhibitor would identify novel bone anabolic factors in osteoblasts. We therefore performed a comparative DNA microarray analysis between empty vector-transfected mouse osteoblastic MC3T3-E1 cells and PD98059-treated stable Smad3-overexpressing MC3T3-E1 cells. Among the novel factors, Tmem119 was selected on the basis of its rapid induction by PTH independent of later increases in endogenous TGF-β. The levels of Tmem119 increased with time in cultures of MC3T3-E1 cells and mouse mesenchymal ST-2 cells committed to the osteoblast lineage by BMP-2. PTH stimulated Tmem119 levels within 1 h as determined by Western blot analysis and immunocytochemistry in MC3T3-E1 cells. MC3T3-E1 cells stably overexpressing Tmem119 exhibited elevated levels of Runx2, osteocalcin, alkaline phosphatase, and β-catenin, whereas Tmem119 augmented BMP-2-induced Runx2 levels in mesenchymal cells. Tmem119 interacted with Runx2, Smad1, and Smad5 in C2C12 cells. In conclusion, we identified a Smad3-related factor, Tmem119, that is induced by PTH and promotes differentiation in mouse osteoblastic cells. Tmem119 is an important molecule in the pathway downstream of PTH and Smad3 signaling in osteoblasts.  相似文献

NADH-cytochrome b5 reductase activities in hemolysates of young and old human erythrocytes, and in hemolysates of rabbit reticulocytes and erythrocytes were measured after the separation of the enzyme from the bulk of hemoglobin only by isoelectric focusing. In any cases, a single main peak of the enzyme activity was detected after the electrophoresis in the fraction with pH 6.8 and 8.3 for human and rabbit red cells, respectively. The rabbit enzyme showed more than 30 times higher enzyme activity than that of human erythrocytes under the standard assay conditions. Significant differences of Micahelis constants for cytochrome b5 of the enzyme were found between young and old human erythrocytes, and also between human and rabbit red cells.  相似文献

We recently reported that capsaicin, a pungent principle of hot red pepper, evokes catecholamine secretion from the rat adrenal medulla. In this study, the effects of some pungent principles of spices on adrenal catecholamine secretion were investigated as compared with that of capsaicin. An increase in catecholamine, especially epinephrine, secretion was observed not only on capsaicin infusion but also on piperine (a pungent principle of pepper) and zingerone (ginger) infusion. Even on infusion of the same amount (650 nmol/kg, i.v.), the order of potency as to catecholamine secretion was capsaicin much greater than piperine greater than or equal to zingerone. While, sulfur-containing and volatile pungent principles, allylisothiocyanate (mustard, etc.) and diallyldisulfide (garlic, etc.), did not even cause slight catecholamine secretion. Furthermore, these adrenergic secretagogues were readily transported via the gut into the body. These results indicate that some pungent principles of dietary spices can induce a warming action via adrenal catecholamine secretion.  相似文献

Biochemical aspects of b-type cytochromes in swine cerebral microsomes were different from those of cytochrome b5 in liver microsomes, as well as the difference in absorption spectra. First, the kinetic constants, Km and Vmax, in rotenone-insensitive NADH-cytochrome c reductase activity were different from those of liver microsomes, and the activity of cerebral microsomes was higher than that of liver microsomes. Second, midpoint potentials (Em) of b-type cytochromes in cerebral microsomes were measured and compared with liver microsomal cytochrome b5. In cerebral microsomes two components of b-type cytochromes were resolved, and showed Em's of ?30 and +50 mV, respectively, in the presence of 2 mm KCN. On the other hand, the Em of liver microsomal cytochrome b5 was ?6 mV. The high-potential component of cerebral microsomal b-type cytochromes was identified as brain-b′5 [S. Yoshida, T. Yubisui, and M. Takeshita (1983)Biochem. Int. 7, 291–298] and the low-potential component as brain-b5. The significance of the difference between cerebral and liver microsomal b-type cytochromes was discussed.  相似文献

The present experiment was performed in order to analyze and compare the histopathological features of Candida infection in various states of host defense capacity. The pulmonary lesions were induced by an intratracheal inoculation of 108 live cells of Candida albicans into each of the rabbits in the following 4 groups: 1) untreated controls, 2) animals sensitized non-specifically to bovine serum albumin (BSA), 3) those sensitized specifically to formalin-killed C. albicans cells, and 4) those treated with cyclophosphamide and mitomycin C. The animals were sacrificed at appropriate intervals up to 16 days after inoculation. At autopsy, the lungs were cultured and then subjected to histopathological, electron microscopic and enzyme cytochemical examinations.In the healthy control animals, the fungal lesions consisted of polymorphonuclear leukocytes (PMN) at the initial stage, gradually changing to granulomatous inflammation, which contained no peroxidase-positive macrophages. In the animals sensitized non-specifically to BS A exudative macrophages appeared in the lesions, the nature of which did not differ from that of the control animals. In the animals sensitized specifically to Candida cells, more extensive infiltration of PMN was observed at the initial stage, a fact which may suggest the participation of the Arthus phenomenon in the development of the lesions. Furthermore, an epithelioid cell transformation of the macrophages in the granulomatous lesions may also suggest that immune complexes contributed to their formation. In the drug-treated animals, the lesions consisted of necrotic or less prominent cellular foci which correspond to the features of human candidiasis in debilitated states, and the inoculated fungi grew progressively to form pseudohyphae. An asteroid structure protruding radially from the surface of the fungal cells and attaching to primary lysosomes in the phagocytes was observed occasionally. This structure seems to be formed when the function of the phagocytes in the defense mechanisms against the fungi is suppressed to some extent.From the results of the present experiment, we would emphasize that PMN play an initial role in the elimination of C. albicans cells in the lung, and that macrophages then contribute to the formation of the lesions immunologically or non-immunologically.  相似文献
