). We succeeded in obtaining a fragment of the W

chromosome designated as Ze
W, having 3 of 12 W-RAPD markers (W-Bonsai, W-Yukemuri-S, W-Yukemuri-L). Inheritance of the Ze
W fragment by males indicates that it does not include the Fem gene. On the basis of these results, we determined the relative positions of W-Yukemuri-S and W-Yukemuri-L, and we narrowed
down the region where Fem gene is located. In addition to the Ze
W fragment, the Z chromosome was also broken into a large fragment (Z1) having the +
sch
(1-21.5) and a small fragment (Z2) having the +
od
(1-49.6). Moreover, a new chromosomal fragment (Ze
WZ2) was generated by a fusion event between the Ze
W and the Z2 fragments. We analyzed the genetic behavior of the Z1 fragment and the Ze
WZ2 fragment during male (Z/Z1

WZ2) and female (Z1
Ze
WZ2/W) meiosis using phenotypic markers. It was observed that the Z1 fragment and the Z or the W chromosomes separate without fail. On the other hand, non-disjunction between the Ze
WZ2 fragment and the Z chromosome and also between the Ze
WZ2 fragment and the W chromosome occurred. Furthermore, the females (2A: Z/Ze
WZ2/W) and males (2A: Z/Z1) resulting from non-disjunction between the Ze
WZ2 fragment and the W chromosome had phenotypic defects: namely, females exhibited abnormal oogenesis and males were flapless

The differential distribution of microtubules in osteoclasts in culture was examined by using antibodies against acetylated, tyrosinated, or detyrosinated tubulins. Tyrosinated tubulin was found throughout the cytoplasmic microtubules in all cells examined. An expanding protrusion that contained tyrosinated tubulin but none of the detyrosinated or acetylated form was seen in the immature osteoclasts. Detyrosinated or acetylated tubulin was detectable in the peripheral cytoplasm of the mature osteoclasts displaying the loss of the expanding protrusion. Although most of the microtubules were derived from the centrosome, noncentrosomal microtubules were distributed in the expanding protrusion, which was predominantly positive for tyrosinated tubulin. By tracing single microtubules, the authors found that their growing ends were always rich in tyrosinated tubulin subunits. End binding protein 1 bound preferentially to the microtubule ends. Both acetylated and tyrosinated microtubules were shown to be closely associated with podosomes. Microtubules appeared to grow over or into the podosomes; in addition, the growing ends of single microtubules could be observed to target the podosomes. Moreover, a microtubule-associated histone deacetylase 6 was localized in the podosomes of the osteoclast. On the basis of these results, the authors conclude that posttranslational modifications of microtubules may correlate with characteristic changes in podosome dynamics in osteoclasts. 相似文献

Summary A prenatal diagnosis of GM1-gangliosidosis was made in a pregnancy at risk, on the basis of a deficiency of -galactosidase activity demonstrated in cultured aminiotic fluid cells. Biochemical analyses were performed in the aborted fetus. GM1-ganglioside -galactosidase activity was reduced to 1% of the control value in both the brain and liver of the affected fetus. Lamellar bodies suggestive of membranous cytoplasmic bodies were found in cells of basal ganglions, while the accumulation of GM1-ganglioside in the brain was not remarkable. 相似文献

The introduced Bombus terrestris has been blamed for the decline of native bumblebee populations in Japan. To control this invasive species, it is necessary
to understand its ecological traits in its newly established range. Here, we investigated the colony growth pattern and reproductive
ability of feral nests of B. terrestris in northern Japan from 2003 to 2006. Nests collected at various times showed initially slow growth followed by rapid development.
This pattern is consistent with findings in previous studies using laboratory-raised colonies. Our results also suggested
that protandrous colonies tend to specialize in male production, whereas protogynous colonies specialize in gyne production,
producing a split sex ratio in feral colonies of B. terrestris. The numerical population sex ratio was 1.40 gynes per male, calculated from the numbers of pre-emergence cocoons for gynes
and males, showing a female-biased sex ratio at the population level. Mature colonies produced a mean of 376.5 cocoons and
90.2 gyne cocoons (22.1% of the total). The proportion of gynes produced by B. terrestris nests exceeded both those of conspecifics observed in other countries and those of consubgeneric native Japanese species.
The propagule pressure hypothesis appears to explain the probability of establishment of this invasive species. Suitable nest
sites for B. terrestris queens appeared to be in short supply, and B. terrestris may win usurpation contests against native species due to its large size, resulting in the decline of native bumblebee species. 相似文献

Ribosomal protein S6 kinase 1 (S6K1) is a serine/threonine protein kinase that plays an important role in the PIK3/mTOR signaling pathway, and is implicated in diseases including diabetes, obesity, and cancer. The crystal structures of the S6K1 kinase domain in complexes with staurosporine and the S6K1-specific inhibitor PF-4708671 have been reported. In the present study, five compounds (F108, F109, F176, F177, and F179) were newly identified by in silico screening of a chemical library and kinase assay. The crystal structures of the five inhibitors in complexes with the S6K1 kinase domain were determined at resolutions between 1.85 and 2.10 Å. All of the inhibitors bound to the ATP binding site, lying along the P-loop, while the activation loop stayed in the inactive form. Compound F179, with a carbonyl group in the middle of the molecule, altered the αC helix conformation by interacting with the invariant Lys123. Compounds F176 and F177 bound slightly distant from the hinge region, and their sulfoamide groups formed polar interactions with the protein. The human Rad51B protein is involved in the recombinational repair of damaged DNA. Chromosomal rearrangements of the Rad51B gene have been found in uterine leiomyoma patients, suggesting that the Rad51B gene suppresses tumorigenesis. In the present study, we found that the purified Rad51B protein bound to single-stranded DNA and double-stranded DNA in the presence of ATP and either Mg(2+) or Mn(2+) and hydrolyzed ATP in a DNA-dependent manner. When the synthetic Holliday junction was present along with the half-cruciform and double-stranded oligonucleotides, the Rad51B protein only bound to the synthetic Holliday junction, which mimics a key intermediate in homologous recombination. In contrast, the human Rad51 protein bound to all three DNA substrates with no obvious preference. Therefore, the Rad51B protein may have a specific function in Holliday junction processing in the homologous recombinational repair pathway in humans. 相似文献
