Background

Lutzomyia longipalpis is the vector of Leishmania infantum in the New World, and its saliva inhibits classical and alternative human complement system pathways. This inhibition is important in protecting the insect´s midgut from damage by the complement. L. longipalpis is a promiscuous blood feeder and must be protected against its host’s complement. The objective of this study was to investigate the action of salivary complement inhibitors on the sera of different host species, such as dogs, guinea pigs, rats and chickens, at a pH of 7.4 (normal blood pH) and 8.15 (the midgut pH immediately after a blood meal). We also investigated the role of the chicken complement system in Leishmania clearance in the presence and absence of vector saliva.

Results

The saliva was capable of inhibiting classical pathways in dogs, guinea pigs and rats at both pHs. The alternative pathway was not inhibited except in dogs at a pH of 8.15. The chicken classical pathway was inhibited only by high concentrations of saliva and it was better inhibited by the midgut contents of sand flies. Neither the saliva nor the midgut contents had any effect on the avian alternative pathway. Fowl sera killed L. infantum promastigotes, even at a low concentration (2%), and the addition of L. longipalpis saliva did not protect the parasites. The high body temperature of chickens (40°C) had no effect on Leishmania viability during our assays.

Conclusion

Salivary inhibitors act in a species-specific manner. It is important to determine their effects in the natural hosts of Leishmania infantum because they act on canid and rodent complements but not on chickens (which do not harbour the parasite). Moreover, we concluded that the avian complement system is the probable mechanism through which chickens eliminate Leishmania and that their high body temperature does not influence this parasite.  相似文献

The acidic and the basic subunits were shown to be present in equimolar amounts in the 11S globulin molecule by the densitometric scanning of the SDS gel and the molecular weight consideration. The four acidic subunits (A1, A2, A3 and A4) were found to be present in the approximate molar ratio of 1:1:2:2. Four basic subunits separated and designated as B1, B2, B3 and B4 based on the relative mobilities in the acidic gel in 7 m urea were found to be present in the approximate molar ratio of 1:1:2:2. The four basic subunits were fractionated in approximately same amounts into three different peaks, peak I (B1 and B2), peak II (B3) and peak III (B4) by CM-Sephadex C–50 column chromatography in the presence of 6 m urea. Three kinds of intermediary subunits of 11S globulin were fractionated with DEAE-Sephadex A–50 in the absence of reducing agents in 6 m urea, and disulfide bonds appeared to participate in the binding between the acidic and the basic subunits in the molar ratio of 1: 1 with the following combinations; A1 and A2 combined with B3, A3 with B1 and B2, and A4 with B4. In view of the above results and molecular weight consideration, a new model of subunit structure was proposed for 11S globulin.  相似文献

In order to enhance the efficiency converting dibenzothiophene (DBT) into water-soluble compounds by DBT-utilizing bacteria, the following subjects were studied: comparison of activities of isolated strains, their mixed cultivation, DBT dosing, nitrogen sources, substrate concentration, pH, temperature, and oxygen supply. Light oil solution of DBT (5%) was used as substrate. The conversion efficiency was calculated by the amount of sulfur accumulated in the aqueous layer of culture broth.

The conversion ratio of 40% was attained by the mixed culture of Pseudomonas abikonensis and Ps. jianii under the following conditions; medium, DBT 4.6 g dissolved in 87.8 g of light oil, meat extract 4.0 g, Na2HPO4·12H2O 9.5 g, KH2PO4 1.4 g, MgCl2·6H2O 0.2 g, distilled water 1000 ml; pH, 6.9 to 7.3 (m/30 phosphate buffer); oxygen supply, 50 ml broth 500 ml flask, shake culture at 220 rpm; inoculum size, 10%; temperature, 28°C; and period of cultivation, 3 days.  相似文献

Adult T-cell leukemia (ATL) occurs in human T-lymphotropic virus type I-infected individuals and is endemic to the southwestern area of Kyushu in Japan. Here, we found that nM levels of genistein and 17β-estradiol had cytotoxic effects on ATL cells and activated caspase-3. The estrogen receptor antagonist ICI182780 negated the cytotoxic effect of genistein. In addition, G protein-coupled estrogen receptor agonist G-1 also had a cytotoxic effect on ATL cells. This is the first report suggesting that estrogen receptors are a molecular target for ATL therapy.  相似文献

An unidentified bacterial strain S107B1, isolated from soil by use of isopropylbenzene as a carbon source, was shown to bring about oxidation of α-methylstyrene and β-methylstyrene,

One of the oxidation products produced from α-methylstyrene was identified as the new compound, (—)-cis-23-dihydroxy-1-isopropenyl-6-cyclohexene.

The same strain S107B1 also oxidized β-methylstyrene and produced 3-phenylpropionaldehyde and benzoic acid.

From these results, the existence of reductive step for the aerobic degradation of these aromatic hydrocarbons by this strain was made clear. The initial attack on these aromatic hydrocarbons and a cyclohexenediol compound formed from α-methylstyrene were discussed.  相似文献

On the screening of microorganisms which accumulate ultra violet light absorbing substances, some sporogenous bacteria were selected as powerful strains for production of a crystalline substance having a maximum absorption at 267.5 mμ (pH 6.0 in water). These strains were microbiologically examined and named B. subtilis var. thermophilus. Submerged fermentation was carried out for 48 hrs at 37°C in glucose bouillon medium and the isolation of substance was performed by absorption to active carbon and elution with ammonia water.

On the basis of chemical studies of this crystal, it was identified as urocanic acid.  相似文献

2,3-Diaminopropionate ammonia-lyase (DAPAL), which catalyzes α,β-elimination of 2,3-diaminopropionate regardless of its stereochemistry, was purified from Salmonella typhimurium. We cloned the Escherichia coli ygeX gene encoding a putative DAPAL and purified the gene product to homogeneity. The protein obtained contained pyridoxal 5′-phosphate and was composed of two identical subunits with a calculated molecular weight of 43,327. It catalyzed the α,β-elimination of both D- and L-2,3-diaminopropionate. The results confirmed that ygeX encoded DAPAL. The enzyme acted on D-serine, but its catalytic efficiency was only 0.5% that with D-2,3-diaminopropionate. The enzymologic properties of E. coli DAPAL resembled those of Salmonella DAPAL, except that L-serine, D- and L-β-Cl-alanine were inert as substrates of the enzyme from E. coli. DAPAL had significant sequence similarity with the catalytic domain of L-threonine dehydratase, which is a member of the fold-type II group of pyridoxal phosphate enzymes, together with D-serine dehydratase and mammalian serine racemase.  相似文献

1-Naphthoic acid derivatives substituted with chlorine, bromine, methyl or nitro groups at various positions of the ring were assayed for their activities in the pea straight-growth and the callus formation tests. Their growth activities are discussed from two points of view, i.e. the spatial structure hypothesis of Veldstra and the ortho reaction hypothesis advanced for benzoic acid derivatives proposed by Hansch and Muir. The effects of cellulose on the production and stimulation of β-transglycosylase were studied. The β-transglycosylase of Trichoderma longibrachiatum was produced specifically in the presence of cellulose in Czapeck-Dox medium containing sucrose as a sole carbon source. The enzyme activity was stimulated by the addition of cellulose in the reaction mixture, where the transfer reaction product (a water-insoluble glucan) was apparently synthesized on the surface of the added cellulose fibers.

The hyphal wall fraction of the fungus had the same stimulatory effect on β-transglycosylase as the cellulose fibers. A cellulose-like material in this fraction was found by partial acid hydrolysis and gas chromatography. Cellotriose was the smallest substrate effective for the synthesis of a water-insoluble glucan in the presence of cellulose by the β-transglycosylase, though a significant amount of glucan could not be synthesized without the addition of cellulose.  相似文献
