The mutagenic and clastogenic effects of N-nitrosodi-n-propylamine (NDPA) in lacZ transgenic mice (MutaMouse) were investigated as a part of the second collaborative study of the transgenic mouse mutation assay by a subgroup of the Mammalian Mutagenesis Study Group, a suborganization of the Environmental Mutagen Society of Japan. Male MutaMouse mice were administered NDPA intraperitoneally at a dose of 250 mg/kg, which is half of the LD(50) of the compound. The clastogenicity of NDPA was examined by the peripheral blood micronucleus test just before and at 24, 48 and 72 h after the treatment. The mutant frequencies in the bone marrow, liver, lung, kidney and urinary bladder were examined by the positive selection method for lacZ kidney. These findings demonstrate that NDPA induces organ-specific mutagenesis with specific expression times, and that the mutagenicity of NDPA in lacZ transgenic mice is consistent with its carcinogenicity.  相似文献

The presumed ancestral karyotype of Muridae, previously reconstructed, is compared with that of a selected Cricetidae species, Akodon a arviculoides. Most of the chromosomes of the presumed ancestral chromosomes of Muridae are homeologous to chromosome arms or segments in the Akodon karyotype. This result strengthens the validity of the reconstruction of the ancestral karyotype for both Muridae and Cricetidae.  相似文献

Preference-aversion for heavy metals in ddy mice was examinedusing the two-bottle preference technique. Mice preferred MnCl2,CoCl2 at certain concentration ranges, but avoided NiCl2, CuCl2,ZnCl2 and CdCl2. The avoidance threshold for the salts becamelower in the order of Mn > Co > Ni > Cd > Cu >Zn, though the degree of rejection for CuCl2 was larger thanthat for ZnCl2. Preferences for sucrose and Na saccharin werereduced by low concentrations of CuCl2 or ZnCl2. However, preferencefor glycine was scarcely affected by addition of either saltat 1 mM. Electrophysiological experiments in mice on the inhibitionby heavy metal salts of chords tympani nerve responses to thefour basic taste stimuli revealed that CuCl2 and ZnCl2 at 1mM inhibited the response to sucrose strongly and those to guinineHCl (QHCl) and NaCl moderately, but had no effect on the HClresponse. On the other hand, MnCl2, CoCl2 and NiCl2 at 1 mMscarcely inhibited the response to sucrose but suppressed thoseto other stimuli. CoCl2 moderately inhibited responses to allthe stimuli. In general, metal salts producing a larger inhibitionof neural responses to sucrose and QHCL are avoided by miceat a lower concentration.  相似文献

Atypical fatty acid metabolism has been reported in attention deficit hyperactivity disorder (ADHD), however, its relationship with temperament in this population is unclear. The current study investigated the association between blood levels of fatty acids implicated in brain structure and function (omega-3, omega-6, omega-9) and personality traits of stability (neuroticism, conscientiousness and agreeableness) and plasticity (extraversion and openness). Twenty right-handed adolescent boys with ADHD completed a self-report NEO-FFI personality questionnaire, and had fatty acid content assessed from red blood using gas chromatography. Pearson's correlations showed no significant associations between omega-3 levels and personality. After correction for multiple comparisons, Adrenic Acid (C22:4n6) was inversely associated with stability. Oleic acid (C18:1n9) was positively associated with plasticity. Results are in line with a role of fatty acids in brain function. They suggest that those fatty acids that are involved in myelination (Adrenic, Oleic) have the strongest associations with temperament in adolescents with ADHD.  相似文献

Significant immunoglobulin (Ig) production by human peripheral blood lymphocytes was induced in vitro by stimulating the cells with pokeweed mitogen (PWM) and Staphylococcus aureus Cowan I (SpA CoI). IgG, IgM, and IgA were determined by a combination of the latex fixation test and radioimmunoassay. High levels (1,000 to 5,000 μg/ml) of IgG and IgM and a lesser amount of IgA were constantly produced during 7 to 8 days of incubation with both stimulants. Ig production induced by SpA Col stimulation was independent of the presence of T cells, while Ig production induced by PWM required T cells exclusively. Depletion of monocytes in the culture caused but a slight decrease in Ig production (particularly in the case of IgG). While the addition of a small number of monocytes enhanced IgG induction by both stimulants, coculture with an excess number of monocytes inhibited Ig induction (particularly IgG) by PWM stimulation but not by SpA CoI stimulation. Marked suppression of Ig production (IgG, IgM, and IgA) was observed in cocultures with Con A-activated T cells. The phenomena of suppression were observed in both the SpA Col-stimulated and PWM-stimulated lymphocytes. These data indicate that Ig production from B cells stimulated with a polyclonal B cell activator, SpA CoI, was independent of T cells and relatively of independent of monocytes, but could be subjected to the regulation of the Con A-induced suppressor T cells.  相似文献

Chromosomal polymorphism resulting from two pericentric inversions in Akodon arviculoides (2n=14) has been described (YONENAGA, 1972a). In this paper the banding patterns are presented and identification of the inverted segments of the autosomal pairs 2 and 3 is made. The karyotype of Akodon sp., which varies in diploid number (2n-24 and 25), is described and shown to be due to the presence of a small submetacentric chromosome in the 2n=25 individuals. The karyotypes of two 2n=19 males studied show that they are hybrids between Akodon arviculoides (2n=14) and Akodon sp. (2n=24).  相似文献

DBA/2FG-pcy/pcy (D2-pcy) mice are a hereditary murine model of slowly progressive polycystic kidney disease (PKD) and characterized by the persistent excretion of acidic urine, in association with polyuria, after weaning. In this study, the activity of carbonic anhydrase (CA) and it histological distribution in the kidney of D2-pcy mice were investigated by immunohistochemistry. Significantly higher CA activity was detected in the cytosolic, but not membrane, fraction of kidney homogenates in 5-week-old D2-pcy mice than in age-matched, control DBA/2 (D2) mice, and a more rapid rate of urine acidification was noted in 11-week-old mice when acetazolamide, an inhibitor of the enzyme, was administered orally. By immunohistochemistry for the major renal CA isoenzyme (CA II), epithelial cells in the distal straight tubules and the cortical collecting ducts were stained intensely, whereas those of the proximal convoluted tubules had only weak and diffuse staining. The glomeruli, the proximal straight tubules and the ascending thin limb of Henle's loop were almost free from staining. In the cells lining cysts and/or dilated tubules, CA II activity was well preserved, although the staining intensity was considerably reduced in fully-flattened, lining cells of cysts, but no difference was found between D2-pcy and D2 mice in any segmental localization of renal CA II activity. From these results it seems that D2-pcy mice in the early stages of the cystic disease continue to secrete excess protons through the CA-mediated reaction that is stimulated for regulation of acid-base balance in the distal portion of the nephron and the collecting duct in kidney. It also suggests that monitoring urine pH may be useful in predicting the effects of early interventions on the progression of slowly developing renal cysts.  相似文献
