The dynamic process in rat thymocyte restoration after their destruction by glucocorticoid (GC) administration was examined. Thymus weight and thymocyte count became minimal 4-5 days after the administration. Then the thymus took a course of recovery. Endogenous DNA synthesis in thymocytes, reflecting their proliferation within thymus, decreased for 4 days but began to increase 6-8 days after GC treatment. Thymocyte responsiveness to soybean lectin (SBL), a possible stimulator for T-cell-precursors, showed elevation 4-5 days after the treatment. A marked decrease of lymphocytes in the cortex and unclearness of cortico-medullary junction were observed 2-3 days after GC treatment. Clusters of small lymphoid cells, which possibly contained SBL-responding cells, were found in the subcapsular area 4 days after the treatment and successively, large lymphocytes became visible in the same area. Thereafter, small lymphocytes in the cortical mid and deep zones increased, and cortico-medullary junction was restored. These histological features are discussed from the view of correspondence with the dynamic changes of endogenous DNA synthesis and SBL responsiveness in the thymocytes after GC administration. 相似文献

Growth of chimpanzees reared at the Kumamoto Primates Park of Sanwa Kagaku Kenkyusho Co. Ltd. was studied cross-sectionally
from the viewpoints of somatic growth and reproductive maturation. Distance and velocity curves were expressed using spline
function method. Males showed adolescent growth acceleration in body weight, with a peak at 7.86 yrs of age, but not in trunk
length. Females showed continuous rapid growth from mid-juvenile to adolescent phase in both body weight and trunk length,
but no isolated adolescent spurt. The Sanwa chimpanzees matured at about 12.5 yrs of age for females and 15.0 yrs for males.
The mean adult weights and trunk lengths were 53.2 kg and 507.8 mm for males and 42.7 kg and 481.6 mm for females. The Sanwa
chimpanzees had similar growth patterns to those of the Yerkes chimpanzees, although they showed a slight delay in infancy,
and a higher growth rate from the early juvenile phase onwards. Growth patterns in these two laboratories may be regarded
as “normative” for laboratory-reared chimpanzees. They matured earlier than wild chimpanzees by more than two years. The major
reason for the retarded maturation in wild chimpanzees is the delay of growth from infant to the early juvenile phases (0–4
yrs of age), probably owing to a limited nutritional supply from the mother. Development of the testes comprised three phases:
slow growth from infant to juvenile (until 6.4 yrs); rapid growth around adolescence (until 9.2 yrs); and adult (mean testicular
volume, 187 cm3). Setting the nutritional standard at 2,000–2,600 Cal/day (= Kcal/day) per adult, calories were considered for captive chimpanzees

Many eukaryotic proteins are bound to the plasma membrane via a glycosylphosphatidylinositol (GPI) anchor. Its core backbone, which is conserved in different organisms, is synthesized in the endoplasmic reticulum by the sequential addition of glycan components to phosphatidylinositol. One of the human GPI synthesis genes,PIGF(phosphatidylinositol glycan complementation class F), which is involved late in the synthesis pathway, has been cloned. In this study, we isolated complementary and genomic clones ofPigf,a murine counterpart ofPIGF. Pigfencodes a 219 amino acid protein that complements a class F mutation. ThePigfgene consists of six exons spanning 30 kb and was mapped to chromosome 17 at 17E4–E5. These features are very similar toPIGF,thus demonstrating the interspecies conservation of structure, function, gene organization, and genetic locus between these GPI synthesis genes. Cell proliferation was examined in the back and tail epidermis of larval Xenopus laevis using bromodeoxyuridine (BrdU). The BrdU labeling index of the back epidermis increased temporally at stage 59, followed by a rapid decrease to the same level as at stage 51. The temporal increase in cell proliferation of the back epidermis produced a new epidermal layer composed of basal cells. In vitro analysis showed that tri-iodothyronine (T3) promotes cell proliferation of basal cells but suppresses that of skein cells. Immunohistochemical studies showed that the newly formed basal cell layer functions as adult precursor cells which produce the adult epidermal cells. In contrast to the back epidermis, the labeling index of the tail epidermis decreased from stage 57. However, when the tail skin was transplanted to the back area, cell proliferation in the tail epidermis increased to the same level as that of the normal back epidermis. Cell proliferation of the back epidermis was not suppressed by transplanting the skin to the tail area. These results suggest that some promoting factors are produced in the body region and regulate the number of adult precursor cells, which determine the developmental fate of the larval skin. 相似文献

The effect of shock-loading of zinc, copper and cadmium ions on the removal of total organic carbon (TOC) and phosphate in an anaerobic-aerobic activated sludge process was investigated. TOC removal was not sensitive to shock-loading of Zn2+ and Cd2+ ions, and complete removal was achieved even at 20 mg Zn2+/l and 20 mg Cd2+/l. BACKGROUND: Cell cycle progression requires the activity of protein kinases and phosphatases at critical points in the cell cycle in all eukaryotes. We have previously reported that the dis2(+) and sds2(+) genes of fission yeast encode redundant catalytic subunits of a type 1-like protein phosphatase. The sds22(+) gene was shown to be essential for cell viability and to interact genetically with dis2(+) and sds21(+). RESULTS: Here we show by immunoprecipitation that the sds22 protein physically interacts with the dis2 and sds21 proteins, and that sds22-associated phosphatase activity has altered substrate specificity, The loss of sds22 function by a temperature sensitive mutation leads to cell cycle arrest at mid-mitosis, at which point cdc2-dependent histone Hl kinase activity is high while sds22-dependent H1 phosphatase activity is low. To examine the unusual properties of sds22 protein structure, we analyzed a collection of sds22 deletion and point mutants by a variety of functional criteria. CONCLUSION: We propose that sds22 is a regulatory subunit of the dis2/sds21 phosphatase catalytic subunits and that sds22-bound phosphatase carries a key phosphatase activity essential for the progression from metaphase to anaphase. Mutational analysis indicates that dis2/sds21 interacts with the central repetitive domain of sds22, while the C-terminal and central regions of sds22 may be involved in subcellular targeting and the N-terminus is important for stability. 相似文献

The density of a Japanese serow (Capricornis crispus) population, which has been in stable habitat conditions and free from hunting and predation, was investigated by direct
observation of serows at Kusoudomari (336 ha), Wakinosawa village, Aomori Prefecture, Japan, during 1977–89. The vegetation,
composed of mature mixed forest ofThujopsis with deciduous trees (43.2%), secondary forest of deciduous trees (23.4%) and a plantation of coniferous trees (30.0%) showed
little change during the study period. The serows were counted 17 times each winter for 14 consecutive years. The serow population
maintained a stable density with a mean of 12.5 individuals km−2 (SD=1.4) ranging from 8.6 to 14.6 km−2. The ratio of the proportion of kids (<1 year old) to serows more than 1 year old decreased, but density did not decrease
during the study period. Though extremely severe winters with heavy snowfall in 1984–86 caused a decline in serow density,
the density recovered within a few years. It was considered that the stability of the serow density resulted mainly from the
stable food supply and the lack of human disturbance, such as forest cutting in the habitat. Stable population density for

The inhibitory effect of capsaicin on axoplasmic transport in cultured dorsal root ganglion cells was analyzed by video-enhanced contrast microscopy. Capsaicin inhibited particle transports in a dose-dependent manner, irrespective of the diameter of axons. The effect of capsaicin was reversible at low concentrations. Capsaicin affected both the anterograde and retrograde transport. Large organelles were more sensitive to capsaicin than small ones in the retrograde transport. An experiment using calcium-sensitive dye, Fura 2, indicated that capsaicin raised the intraneuronal free calcium concentration preceding the inhibition of the transport. Electron microscopy revealed that microtubules and neurofilaments are disorganized and disoriented by capsaicin. We reached a conclusion that capsaicin inhibits fast axoplasmic transport of both anterograde and retrograde directions in all types of somatosensory neurons in culture by disorganizing intraaxonal cytoskeletal structures, through the elevated intracellular Ca2+ concentration. © 1993 John Wiley & Sons, Inc. 相似文献
