Grandinol, an inhibitor of seed germination and photosynthesis in Eucalyptus sp., inhibits transpiration and stomatal opening. The acylphloroglucinol structure in grandinol seemed to be essential for these activities. Enhancement of activity was achieved by the introduction of a formyl group into the molecule. Therefore, structural requirements for these activities were very similar to that for the inhibition of seed germination and photosynthesis. Other grandinol-related compounds having two electron-withdrawing groups on the phloroglucinol nuclei were also active in these assays.  相似文献

Brassinolide [2, 3, 22R,23R-tetrahydroxy-24S-methyl-B-homo-7-oxa-5--cholestan-6-one] and its related compounds, brassinosteroids, applied at the early stages of conditioning shortened the conditioning period required before clover broomrape seeds would germinate after exposure to germination stimulants, such as dl-strigol and natural stimulants from red clover (Trifolium pratense L.) root exudate. Brassinosteroids applied after conditioning increased the rate of the seed germination induced by these stimulants. The inhibitory effect of light on seed germination when it was induced by dl-strigol could be overcome by brassinosteroids. Brassinosteroids also eliminated the inhibitory effects of light and dl-strigol applied at the early stages of conditioning. GA3 was also effective in causing seed conditioning and increased the rate of the germination induced by these stimulants. There was a relationship between brassinosteroids and GA3 in many of the experiments conducted. These findings may have practical implications in increasing the effectiveness of applying germination stimulants in the field to soils for suicidal germination of broomrape seeds.  相似文献

Well-nodulated soybean ( Glycine max L. Merr. cv. Akisengoku) plants were allowed to assimilate 13CO2. Plant cytosol and bacteroid fractions were isolated from nodules, and the kinetics of [13C]-labelling of soluble carbohydrates, organic acids and amino acids were investigated.
The concentrations of all metabolites, with the exception of trehalose and 3-hydroxy-butyrate, were 10- to 1000-fold higher in plant cell cytosol than in bacteroids. The major portion of trehalose was found in bacteroids and 3-hydroxybutyrate only in bacteroids. Sucrose was most highly labelled with 13C in nodules, and the levels and time-course of labelling of sucrose were in good agreement with those of respired CO2 from the nodules. The levels and time-courses of labelling of sucrose were closely similar in cytosol and bacteroids. Glucose was less labelled than sucrose and the level of labelling was consistently higher in cytosol than in bacteroids. The levels of [13C]-labelling of organic acids and amino acids in nodules were lower than those of sucrose and of respired CO2. Tricarboxylic acid cycle intermediates, particularly succinate, were considerably less labelled in bacteroids than in the cytosol. All amino acids detected were also much more rapidly labelled in the cytosol. The results are discussed in relation to the utilization and possible compartmentation of carbon substrates in nodule tissues.  相似文献

A fluorescent resonance energy transfer substrate with improved sensitivity for ADAM17, −10, and −9 (where ADAM represents a disintegrin and metalloproteinase) has been designed. The new substrate, Dabcyl-Pro-Arg-Ala-Ala-Ala-Homophe-Thr-Ser-Pro-Lys(FAM)-NH2, has specificity constants of 6.3 (±0.3) × 104 M−1 s−1 and 2.4 (±0.3) × 103 M−1 s−1 for ADAM17 and ADAM10, respectively. The substrate is more sensitive than widely used peptides based on the precursor tumor necrosis factor-alpha (TNF-alpha) cleavage site, PEPDAB010 or Dabcyl-Ser-Pro-Leu-Ala-Gln-Ala-Val-Arg-Ser-Ser-Lys(FAM)-NH2 and Mca-Pro-Leu-Ala-Gln-Ala-Val-Dpa-Arg-Ser-Ser-Arg-NH2. ADAM9 also processes the new peptide more than 18-fold better than the TNF-alpha-based substrates. The new substrate has a unique selectivity profile because it is processed less efficiently by ADAM8 and MMP1, −2, −3, −8, −9, −12, and −14. This substrate provides a unique tool in which to assess ADAM17, −10, and −9 activities.  相似文献

Dichloroacetate (DCA) was administered orally to normal (nondiabetic) and streptozotocin-diabetic rats in a dose of 1000 mg/day/kg rat wt. One group of diabetic animals received DCA both orally and intraperitoneally. DCA therapy lowered the blood glucose values of diabetic animals but did not alter values in nondiabetic rats. The hepatic activity of glucokinase and pyruvate kinase were significantly lower in both DCA-treated nondiabetic and DCA-treated diabetic animals than values observed for untreated animals. However, DCA therapy was accompanied by remarkable increases in the activities of glucose-6-phosphate dehydrogenase and malic enzyme in both nondiabetic and diabetic animals. Glucose-6-phosphate dehydrogenase was 3-fold higher in DCA-treated nondiabetic animals whereas malic enzyme activity was 10-fold higher in the treated animals than observed in the untreated animals. Similar changes, although smaller in magnitude, were observed for these enzymes in the DCA-treated diabetic animals. Although DCA therapy was accompanied by a significant increase in the wet weights of the liver for both nondiabetic and diabetic animals, no morphological changes were seen by light or electron microscopy. Our observations coupled with those of previous investigators suggest that DCA therapy may have an important role in pyruvate metabolism and may lower the blood glucose concentration by inhibiting hepatic gluconeogenesis.  相似文献

Sixteen legumes were grown in N-free media so that N was suppliedentirely by symbiotic N2 fixation. The plant tissues were analyzedfor natural 15N abundance (expressed as 15N per mil relativeto air N2) with a ratio mass spectrometer. The nodules of desmodium,centro, siratro, soybean and winged bean showed high enrichmentin 15N (+9), while red clover showed slight enrichment (+2).The nodules of 9 other forage legumes (Townsville stylo, whiteclover, alsike clover, common vetch, Chinese milk vetch, senna,alfalfa, ladino clover, and hairy vetch) showed little enrichmentin 15N. In all the legumes investigated, particularly in the ureide-transportingplants such as desmodium, centro, siratro, soybean, winged beanand field bean, the 15N value of the shoots was negative (–3.2).The 15N value of the shoots in winged bean and field bean variedby about 1 depending on the Rhizobium strains used. The isotopicmass balance of 13 legumes indicated that isotopic fractionationoccurs during N2 fixation by the legume-rhizobia symbiosis witha preference for 14N over 15N, resulting in a 15N value of –0.2to –2 in the whole plant. The effect of alterations to the insulin receptor on the insulin sensitivity of isolated adipocytes was studied. Receptor changes were induced by treatment of adipocytes with either phospholipase C or trypsin. After enzyme treatment, binding of insulin to insulin receptors and insulin-mediated glucose metabolism were examined. Exposure of adipocytes to phospholipase C (2 units/ml) significantly increased insulin binding to the cells, but destroyed the ability of the cells to oxidize glucose. After treatment with trypsin (500 micrograms/ml) for 5 min, insulin binding to the adipocytes was significantly increased. This was shown to be due to an increase in insulin-receptor affinity. Metabolic studies showed that trypsin treatment led to an increase in basal glucose transport but markedly decreased the response to insulin at all concentrations tested. Adipocytes treated with trypsin showed no significant difference in basal glucose oxidation rates when compared with controls, but were less sensitive to insulin at low insulin concentrations, and showed a decreased maximum response at high insulin concentrations. In conclusion, these findings indicate a dissociation between induced changes in binding of insulin to insulin receptors and subsequent hormone action. The importance of post-receptor events in the biological action of insulin is highlighted.  相似文献

The reduction of nitrate and nitrite in leaf disks from sevendi- and two monocotyledonous species under in vivo nitrate reductaseassay conditions was studied using 15N-labeled substrates. Significantreduction of both nitrate and nitrite into ammonia and aminoacids was detected under aerobic conditions (in an atmosphereof air): in some cases, the amount of nitrate-N reduced to ammoniaand amino acids was more than that remaining as nitrite. Anaerobicincubation (under an atmosphere of N2 gas) enhanced the accumulationof nitrite, but the subsequent reduction to the basic nitrogencompounds was 40 to 180% of the aerobic rates. The present examinationindicates that in vivo assays of nitrate reductase under aerobicconditions may give greatly underestimated results due to nitritereduction and that exclusion of oxygen from the in vivo assaymixture is desirable in terms of the quantity of nitrite formed.n-Propanal treatment increased nitrite accumulation under aerobicbut not under anaerobic conditions, and depressed the incorporationof nitrate-N into basic fractions under both conditions. Therefore,addition of n-propanol may be desirable for assays under aerobicconditions. No significant difference in the reduction of nitratesupplied as sodium and potassium salts was observed on the nitriteformation and on the incorporation of nitrate-N into basic fractions. 15N experiments on dark assimilation of nitrate, nitrite andammonia into amino acids in wheat leaves showed that these threenitrogen sources were assimilated through the same route andthat the glutamine synthetase/glutamate synthase pathway wasthe major route. With anaerobic treatment, the incorporationof nitrogen into alanine and serine remained at relatively high,but the incorporation into aspartate and asparagine was muchlower than in the cases of aerobic treatment.
