? Efforts are currently underway to establish a standard DNA barcode region for fungi; we tested the utility of the internal transcribed spacer (ITS) of nuclear ribosomal DNA for DNA barcoding in lichen-forming fungi by sampling diverse species across eight orders. ? Amplification of the ITS region (ITS1-5.8S-ITS2) was conducted for 351 samples, encompassing 107, 55 and 28 species, genera and families, respectively, of lichenized fungi. We assessed the ability of the entire ITS vs the ITS2 alone to discriminate between species in a taxonomic dataset (members of the genus Usnea) and a floristic dataset. ? In the floristic dataset, 96.3% of sequenced samples could be assigned to the correct species using ITS or ITS2; a barcode gap for ITS is present in 92.1% of species. Although fewer species have a barcode gap in the taxonomic dataset (73.3% with ITS and 68.8% with ITS2), up to 94.1% of samples were assigned to the correct species using BLAST. ? While discrimination between the most closely related species will remain challenging, our results demonstrate the potential to identify a high percentage of specimens to the correct species, and the remainder to the correct genus, when using DNA barcoding in a floristic context.  相似文献

One theory to explain the high incidence of niche specialization in many animals is that it reduces attentional load during resource-seeking behaviour and thus leads to more accurate resource selection. A recent neural network model refined the predictions of this theory, indicating that a cognitive advantage in specialists is likely to occur under realistic ecological conditions, namely when 'mistakes' (i.e. selection of non-host resources) contribute moderately but positively to fitness. Here, we present a formal empirical test of the predictions of this model. Using a human-computer interactive, we demonstrate that the central prediction of the model is supported: specialist humans are more accurate decision-makers than generalists when their mistakes are rewarded, but not when mistakes are punished. The idea that increased decision accuracy drives the evolution of niche width in animals has been supported in almost all empirical systems in which it has been investigated. Theoretical work supports the idea, and now the predictions of a key theoretical model have been demonstrated in a real biological information-processing system. Considering these interlocking pieces of evidence, we argue that specialization through increased decision accuracy may contribute significantly, along with other mechanisms, to promote niche specialization in animals.  相似文献

One idea to explain the high incidence of specialisation in phytophagous insects is that their nervous systems are simple and unable to efficiently recognise multiple host plants. Here this 'neural limitations' hypothesis is tested using a generalist (the summer winged virginopara) and specialist (the autumn gynopara) morph of a single clone of the black bean aphid, Aphis fabae, that are almost identical morphologically and have the same genotype. Electrical monitoring of stylet behaviour and non-invasive behavioural observations are used to compare initial plant recognition, first reproduction, 'first registered phloem contact' and 'phloem acceptance' of the generalist and specialist on their specific host plants (generalist--herbaceous hosts, Beta vulgaris, Papaver dubium, Rumex obtusifolius, Vicia faba; specialist--woody host, Euonymus europaeus) and on the same host (E. europaeus). Additional analyses of behaviour over the first few minutes of plant contact are carried out for three mutual non-host plants: Euonymus alatus, Prunus padus and Brassica pekinensis. Few examples are found where the host-selection behaviour of the specialist on its host plant is more efficient than the generalist across its hosts and there is little difference in the behaviour of the different forms on the same host. Indeed there are several instances where the generalist is behaviourally more efficient than the specialist. These include absolute time to first reproduction by the generalist on various herbaceous hosts and the same host, E. europaeus, absolute time to first registered phloem contact by the generalist on P. dubium, and incidence of phloem acceptance by the generalist on several of its herbaceous hosts. Rejection of non-hosts is carried out more efficiently by the specialist gynopara on P. padus and B. pekinensis but not on E. alatus. For generalist and specialist morphs of the same A. fabae genotype, therefore, broad host range does not appear to be associated with a reduced ability to make plant-use decisions.  相似文献

Hepatocytes isolated from the periportal or perivenous zones of livers of fed rats were used to study the long-term (14 h) and short-term (2 h) effects of glucagon on gluconeogenesis and ketogenesis. Long-term culture with glucagon (100 nM) resulted in a greater increase (P less than 0.01) in gluconeogenesis in periportal than in perivenous cells (93 +/- 16 versus 30 +/- 14 nmol/h per mg of protein; 72% versus 30% increase), but short-term incubation (2 h) with glucagon resulted in similar stimulation in the two cell populations. Rates of ketogenesis (acetoacetate and D-3-hydroxybutyrate production) were not significantly higher in periportal cells cultured without glucagon, compared with perivenous cells. However, after long-term culture with glucagon, the periportal cells had a significantly higher rate of ketogenesis (from either palmitate or octanoate as substrate), but a lower 3-hydroxybutyrate/acetoacetate production ratio, suggesting a more oxidized mitochondrial NADH/NAD+ redox state despite the higher rate of beta-oxidation. Periportal hepatocytes had a higher activity of carnitine palmitoyltransferase but a lower activity of citrate synthase than did perivenous cells. These findings suggest that: (i) glucagon elicits greater long-term stimulation of gluconeogenesis in periportal than in perivenous hepatocytes maintained in culture; (ii) after culture with glucagon, the rates of ketogenesis and the mitochondrial redox state differ in periportal and perivenous hepatocytes.  相似文献

The kinetics of phase separation and microstructure of oat β-glucan/whey protein binary mixtures varying in concentration (4–16% w/v protein, 0.3–1.2% w/v β-glucan) and β-glucan molecular weight (1.3 × 106, 640 × 103, 180 × 103, and 120 × 103 g/mol) was investigated by turbidimetry and fluorescent microscopy. The phase separation of the mixed systems was followed at pH 7.0 and at room temperature under quiescent conditions. Application of first principles revealed that phase separation of the systems follows first-order kinetics. Acceleration of the phase-separation process was observed with increase of β-glucan concentration for the three lowest-MW samples but the highest molecular weight (1.3 × 106 g/mol) exhibited the opposite trend. Changes in the polysaccharide molecular weight resulted in considerable differences in β-glucan aggregate morphology in the mixed systems. The change in the continuity of the mixed system from polysaccharide-, to bi-, to protein-continuous was confirmed for a wide range of mixed systems differing in biopolymer concentration, and β-glucan molecular weight.  相似文献

Genetic variances and selection efficiencies for growth traits of white spruce (Picea glauca (Moench) Voss) were estimated from clonally replicated full-sib progeny tests established both in nursery and field environments in New Brunswick, Canada. The available data included heights at 4, 5, and 6 years in the nursery test; height at 9 years, height, DBH, and volume at 14 years in the field test. Estimated variance components were interpreted according to an additive-dominance-epistasis model. For heights in the nursery test, while both non-additive and additive variances were important sources of genetic variation, the former decreased but the latter increased with age; among the non-additive genetic variance, the epistatic variance was much more important than the dominance variance. Different from the nursery traits, for traits in the field test, additive variance accounted for an average of 81% of the total genetic variance, whereas dominance variance explained most of the remaining genetic variance. Genetic parameters and selection efficiencies for three vegetative deployment strategies: deploying half-sib families (VD_FAMHS), full-sib families (VD_FAMFS), and multi-varietal forestry (MVF), were compared. Heritability estimates were moderate for VD_FAMHS and VD_FAMFS (0.61–0.72), high for MVF (>0.82) for the nursery heights, and high (>0.79) for the field traits for all strategies. Genetic correlations of volume at age 14 in the field test, the target trait for improvement, were strong (>0.85) with other field traits. Genetic correlations of VOL14 with the nursery heights were also strong (>0.71) at the half-sib and full-sib family levels, but were only moderate (>0.59) for MVF. Overall, practicing MVF is the most effective deployment strategy, yielding the highest genetic gains, followed by VD_FAMFS and VD_FAMHS, regardless of traits and selection methods. Furthermore, early selections for HT9 or for HT4–HT6 were very encouraging, resulting in higher gain in volume at age 14 on a per year basis.  相似文献

Background

Tuberculosis causes significant morbidity and mortality worldwide, especially in sub-Saharan Africa. DC-SIGN, encoded by CD209, is a receptor capable of binding and internalizing Mycobacterium tuberculosis. Previous studies have reported that the CD209 promoter single nucleotide polymorphism (SNP)-336A/G exerts an effect on CD209 expression and is associated with human susceptibility to dengue, HIV-1 and tuberculosis in humans. The present study investigates the role of the CD209 -336A/G variant in susceptibility to tuberculosis in a large sample of individuals from sub-Saharan Africa.

Methods and Findings

A total of 2,176 individuals enrolled in tuberculosis case-control studies from four sub-Saharan Africa countries were genotyped for the CD209 -336A/G SNP (rs4804803). Significant overall protection against pulmonary tuberculosis was observed with the -336G allele when the study groups were combined (n = 914 controls vs. 1262 cases, Mantel-Haenszel 2x2 χ2 = 7.47, P = 0.006, odds ratio = 0.86, 95%CI 0.77–0.96). In addition, the patients with -336GG were associated with a decreased risk of cavitory tuberculosis, a severe form of tuberculosis disease (n = 557, Pearson''s 2×2 χ2 = 17.34, P = 0.00003, odds ratio = 0.42, 95%CI 0.27–0.65). This direction of association is opposite to a previously observed result in a smaller study of susceptibility to tuberculosis in a South African Coloured population, but entirely in keeping with the previously observed protective effect of the -336G allele.

Conclusion

This study finds that the CD209 -336G variant allele is associated with significant protection against tuberculosis in individuals from sub-Saharan Africa and, furthermore, cases with -336GG were significantly less likely to develop tuberculosis-induced lung cavitation. Previous in vitro work demonstrated that the promoter variant -336G allele causes down-regulation of CD209 mRNA expression. Our present work suggests that decreased levels of the DC-SIGN receptor may therefore be protective against both clinical tuberculosis in general and cavitory tuberculosis disease in particular. This is consistent with evidence that Mycobacteria can utilize DC-SIGN binding to suppress the protective pro-inflammatory immune response.  相似文献

This paper investigates the historiographical utility of psychoanalysis, focussing in particular on retrospective explanations of demonic possession and exorcism. It is argued that while ‘full-blown’ psychoanalytic explanations—those that impose Oedipus complexes, anal eroticism or other sophisticated theoretical structures on the historical actors—may be vulnerable to the charge of anachronism, a weaker form of retrospective psychoanalysis can be defended as a legitimate historical lens. The paper concludes, however, by urging historians to look at psychoanalysis as well as trying to look through it. Full-blown psychoanalysis is a bad historical lens but it may, in some cases, be an excellent explanatory template: a thorough understanding of how psychoanalytic therapy functions in the modern world is, for example, a good theoretical preparation for historians seeking to understand the sixteenth-century’s demonological universe.  相似文献
