The nisin resistance determinant and an origin of replication on pNP40, a plasmid of about 60 kilobases that is present in Streptococcus lactis subsp. diacetylactis DRC3, was cloned on a 7.6-kilobase EcoRI fragment. When self-ligated, this fragment existed as an independent replicon (pFM011) and contained a 2.6-kilobase EcoRI-XbaI fragment encoding nisin resistance.  相似文献

Scope and Background

The environmental effectiveness of the Norwegian beverage sector has been studied in a Factor 10 perspective. The objective of the study was to identify strategies that could make the beverage sector radically more effective from an environmental and resource perspective, leading to a Factor 10 improvement. Another main purpose of the work was to test the potential for using Life Cycle Assessment (LCA) methodology on an economic sector with a network of product chains, rather than for a single product.

Methods

Life Cycle Assessment data from STØ’s own studies and literature studies have been used as a basis for analysis of the environmental status of the beverage sector in Norway. The functional unit was defined as the amount of beverage products consumed per capita in Norway in the year 2000. The study includes raw material production, production of the beverage product, packaging manufacture, distribution, use and waste management of the products. The study has, for practical reasons, been limited to the environmental impact indicators total energy consumption and global warming potential. This was done as other types of data have been difficult to obtain for all of the products that were studied (tap water, coffee, milk, soft drinks, beer, squash, juice and bottled water).

Results and Discussion

The study shows differences between the drinking products with respect to energy consumption and emissions that can contribute to global warming. Due to large uncertainties in the data, general conclusions regarding the differentiation of products based on environmental performance should be made with care. Production and distribution of tap water is, however, significantly less energy intensive than the other products. For the impact categories studied, production of raw materials was the most important part of the life cycle for most drinking products.

Conclusions and Perspective

The most significant contributions to achieving a Factor 10 development can be made by consuming more water, especially tap water, and through improving raw material production in the agricultural sector. Packaging and distribution is responsible for only a small part of the energy consumption and emissions leading to global warming. Optimal packaging sizes might however reduce loss of products in the user phase, which is important in order to improve the system. A Factor 10 level seems achievable only if the consumption of tap water is increased to a high level.

The study reports the role of the isozyme forms (cA-PKI and cA-PKII) and subunits (R and C) of cAMP-dependent protein kinase in mediating the acute depression of hepatocyte DNA replication by elevated cAMP. Combinations of cAMP analogs preferentially activating cA-PKI or II showed that either isozyme could inhibit DNA replication. The effects of glucagon and cAMP analogs were counteracted by the cAMP antagonist RpcAMPS, implicating the necessity for cA-PK dissociation in cAMP action. The effect of elevated cAMP was mimicked by microinjected C subunit, but not by the RI subunit of cA-PK. Hepatocytes under continuous cAMP challenge more than regained their replicative activity. This tardive stimulatory effect of cAMP was enhanced by insulin and blocked by dexamethasone, and was preceded by downregulation of cA-PK. In conclusion, a burst of cAMP acutely inhibits hepatocyte G1/S transition in late G1 regardless of hormonal state. In the presence of high glucocorticoid/low insulin the inhibition persists. At high insulin/low glucocorticoid the inhibitory phase is followed by a prolonged stimulation of DNA replication. Downregulation of endogenous cA-PK is a mechanism for escape from the inhibitory action of highly elevated cAMP. © 1993 Wiley-Liss, Inc.  相似文献

Background

Although enteroparasites are common causes of diarrheal illness, few studies have been performed among children in Tanzania. This study aimed to investigate the prevalence of Cryptosporidium parvum/hominis, Entamoeba histolytica and Giardia lamblia among young children in Dar es Salaam, Tanzania, and identify risk factors for infection.

Methodology/Principal Findings

We performed an unmatched case-control study among children < 2 years of age in Dar es Salaam, recruited from August 2010 to July 2011. Detection and identification of protozoans were done by PCR techniques on DNA from stool specimens from 701 cases of children admitted due to diarrhea at the three study hospitals, and 558 controls of children with no history of diarrhea during the last month prior to enrollment. The prevalence of C. parvum/hominis was 10.4% (84.7% C. hominis), and that of G. lamblia 4.6%. E. histolytica was not detected. The prevalence of Cryptosporidium was significantly higher in cases (16.3%) than in controls (3.1%; P < 0.001; OR = 6.2; 95% CI: 3.7–10.4). G. lamblia was significantly more prevalent in controls (6.1%) than in cases (3.4%; P = 0.027; OR = 1.8; 95% CI: 1.1–3.1). Cryptosporidium infection was found more often in HIV-positive (24.2%) than in HIV-negative children (3.9%; P < 0.001; OR = 7.9; 95% CI: 3.1–20.5), and was also associated with rainfall (P < 0.001; OR = 2.41; 95% CI: 1.5–3.8). Among cases, stunted children had significantly higher risk of being infected with Cryptosporidium (P = 0.011; OR = 2.12; 95% CI: 1.2–3.8). G. lamblia infection was more prevalent in the cool season (P = 0.004; OR = 2.2; 95% CI: 1.3–3.8), and more frequent among cases aged > 12 months (P = 0.003; OR = 3.5; 95% CI: 1.5–7.8). Among children aged 7–12 months, those who were breastfed had lower prevalence of G. lamblia infection than those who had been weaned (P = 0.012).

Conclusions

Cryptosporidium infection is common among young Tanzanian children with diarrhea, particularly those living with HIV, and infection is more frequent during the rainy season. G. lamblia is frequently implicated in asymptomatic infections, but rarely causes overt diarrheal illness, and its prevalence increases with age.  相似文献

A total of 48 fungi were characterised for their antagonistic potential against Botrytis cinerea causing Botrytis Gray Mold (BGM) disease in chickpea by dual culture and metabolite production assays. The culture filtrate of the most promising isolate, VFI-51, was purified by various chromatographic techniques and identified as ‘citrinin’ by nuclear magnetic resonance and mass spectrometry studies. The efficacy of citrinin was demonstrated to control BGM in chickpea under greenhouse conditions. The sequences of 18S rDNA gene of the VFI-51 matched with Penicillium citrinum in BLAST analysis. The VFI-51 produced siderophore, hydrocyanic acid, indole-3-acetic acid, lipase, protease and β-1,3-glucanase; grew well in NaCl (up to 15%), at pH between 7 and 11 and temperatures between 20°C and 40°C; and was compatible with fungicides bavistin and thiram. Under greenhouse and field conditions, VFI-51 significantly enhanced the nodule number, nodule weight, root and shoot weight and stover and grain yield over the un-inoculated control. In the rhizosphere, VFI-51 also significantly enhanced total N, available P and OC over the un-inoculated control. Scanning electron microscopy analysis revealed that VFI-51 colonised on the roots of chickpea. This study concluded that VFI-51 has the potential for biocontrol of BGM and plant growth promotion in chickpea.  相似文献

BACKGROUND: Injection of DNA encoding exogenic proteins into muscle tissue combined with electroporation often results in a transient increase of the encoded protein concentration in the muscle and the blood. The reduction is normally due to an immune response against the exogenic protein but other factors may also be involved. How various electroporation parameters affect the concentration kinetics of syngenic and exogenic proteins is studied in relation to immune response and muscle damage after electroporation-mediated DNA transfer to muscle. METHODS: Electroporation was applied to mouse quadriceps and rat tibialis anterior muscles after injection of DNA encoding either secreted alkaline phosphatase (SEAP), beta-galactosidase (beta-gal), luciferase or a mouse IgG molecule. Protein concentrations in blood or muscle and antibody responses were measured for a period up to 3 months. Tissue inflammation and muscle cell damage were studied on muscle cross-sections and assessed by measuring the concentrations of creatine phosphokinase (CPK) in blood. RESULTS: Mice with the highest SEAP concentration in blood at day 7 also had the highest rate of decrease afterwards, the strongest antibody responses against SEAP and the highest acute levels of CPK in blood. DNA-transfected muscle fibers were significantly reduced in number from days 7 to 14. Mononuclear cells surrounded the reporter gene expressing muscle fibers, thus indicating a cellular immune response. When using DNA encoding a syngenic protein the protein concentration in blood was relatively stabile over a 3-month period, but showed different kinetics for various electroporation parameters. CONCLUSIONS: Our findings suggest that the optimal electroporation parameters for DNA vaccination may be different from the optimal parameters for long-term expression of genes encoding syngenic proteins.  相似文献
