Aspergillus oryzae hydrophobin RolA adheres to the biodegradable polyester polybutylene succinate-co-adipate (PBSA) and promotes PBSA degradation by interacting with A. oryzae polyesterase CutL1 and recruiting it to the PBSA surface. In our previous studies, we found that positively charged amino acid residues (H32, K34) of RolA and negatively charged residues (E31, D142, D171) of CutL1 are important for the cooperative ionic interaction between RolA and CutL1, but some other charged residues in the triple mutant CutL1-E31S/D142S/D171S are also involved. In the present study, on the basis of the 3D-structure of CutL1, we hypothesized that D30 is also involved in the CutL1–RolA interaction. We substituted D30 with serine and performed kinetic analysis of the interaction between wild-type RolA and the single mutant CutL1-D30S or quadruple mutant CutL1-D30S/E31S/D142S/D171S by using quartz crystal microbalance. Our results indicate that D30 is a novel residue involved in the ionic interaction between RolA and CutL1.  相似文献

To understand the developmental process of aphid soldier differentiation, we investigated the morphological characters of normal nymphs, soldier nymphs and developing embryos of Pseudoregma bambucicola. Results of morphometric analyses showed that normal and soldier nymphs formed discrete clusters on the basis of several morphological characters, although a small number of intermediate individuals, termed ‘intercaste nymphs’, were present. In late embryonic stages, normal and soldier embryos were morphologically distinguishable. The earlier the embryonic stage, the smaller the morphological differences between them. In early embryos less than 1000 µm in length, normal and soldier embryos were not morphologically distinguishable, suggesting that the onset of soldier differentiation occurs at an early embryonic stage. Throughout embryonic development, morphological differentiation of the soldier caste proceeded gradually. Notably, several morphological characters of soldiers grew remarkably upon larviposition. Observation of embryonic leg cuticle revealed a characteristic folding structure, indicating that some morphological traits of the soldier are exaggerated upon larviposition through expansion of the folded cuticle. We suggest that morphological differentiation of the soldier caste in P. bambucicola comprises two phases: gradual growth during embryogenesis and rapid growth upon larviposition.  相似文献

We established a novel and convenient method to construct a ura3 strain (ura3/ura3) of the asporogenous and diploid yeast, Candida tropicalis, that produces dicarboxylic acid. One copy of the URA3 gene was disrupted using a mutated hygromycin B resistance gene (HYG#). The obtained hygromycin-resistant strain was further transformed with a URA3 disruption cassette and selected on a plate containing 5-fluoroorotic acid. The obtained strains were analyzed and the disruption of the gene was confirmed by PCR and Southern blot analysis. The results showed that the strains were obtained in which allelic URA3 genes were simultaneously disrupted. Furthermore, we established a cotransformation method for this gene disruption, using HYG# in C. tropicalis. In order to disrupt the allelic POX4 genes (encoding acyl-CoA oxidase) of dicarboxylic acid-producing strains, the ARS plasmid (which contained HYG#) and a POX4 disruption cassette (which carried the LAC4 gene encoding beta-galactosidase of Kluyveromyces lactis) were simultaneously introduced by transformation. As a result, the allelic POX4 gene was successfully disrupted.  相似文献

Summary Efficient methods for the preparation of phosphopeptide thioesters were examined, using Fmoc-based solid-phase method. Phosphopeptide thioesters were obtained in good yields by the use of 1-methylpyrrolidine hexamethyl-eneimine and 1-hydroxybenzotriazole in a DMSO-DMF (1∶1, v/v) solution for deblocking the Fmoc groups. Epimerization, which is often observed at the C-terminal amino acid, was effectively suppressed by shortening the time of deblocking process via the use of highly base sensitive Fmoc(2-F) groups for α-amino protection.  相似文献

Although family studies and genome-wide association studies have shown that genetic factors play a role in glaucoma, it has been difficult to identify the specific genetic variants involved. We tested 669 single nucleotide polymorphisms (SNPs) from the region of chromosome 2 that includes the GLC1B glaucoma locus for association with primary open-angle glaucoma (POAG) and normal tension glaucoma (NTG) in the Japanese population. We performed a two-stage case-control study. The first cohort consisted of 123 POAG cases, 121 NTG cases and 120 controls: the second cohort consisted of 187 POAG cases, 286 NTG cases, and 271 controls. Out of six SNPs showing significant association with POAG in the first round screening, seven SNPs were tested in the second round. Rs678350 in the HK2 gene coding sequence showed significant allelic (p = 0.0027 in Stage Two, 2.7XE-4 in meta-analysis) association with POAG, and significant allelic (p = 4.7XE-4 in Stage Two, 1.0XE-5 in meta-analysis) association with NTG. Although alleles in the TMEM182 gene did not show significant association with glaucoma in the second round, subjects with the A/A allele in TMEM182 rs869833 showed worse visual field mean deviation (p = 0.01). Even though rs2033008 in the NCK2 gene coding sequence did not show significant association in the first round, it had previously shown association with NTG so it was tested for association with NTG in round 2 (p = 0.0053 in Stage Two). Immunohistochemistry showed that both HK2 and NCK2 are expressed in the retinal ganglion cell layer. Once multi-testing was taken into account, only HK2 showed significant association with POAG and NTG in Stage Two. Our data also support previous reports of NCK2 association with NTG, and raise questions about what role TMEM182 might play in phenotypic variability. Our data suggest that HK2 may play an important role in NTG in the Japanese population.  相似文献

Among 16 essential elements of higher plants, Ca2+ and B have been termed as apoplastic elements. This is mainly because of their localization in cell walls, however, it has turned to be highly likely that these two elements significantly contribute to maintain the integrity of cell walls through binding to pectic polysaccharides. Boron in cell walls exclusively forms a complex with rhamnogalacturonan II (RG-II), and the B-RG-II complex is ubiquitous in higher plants. Analysis of the structure of the B-RG-II complex revealed that the complex contains two molecules boric acid, two molecules Ca2+ and two chains of monomeric RG-II. This result indicates that pectic chains are cross-linked covalently with boric acid at their RG-II regions. The complex was reconstitutedin vitro only by mixing monomeric RG-II and boric acid, however, the complex decomposed spontaneously unless Ca2+ was supplemented. Furthermore, the native complex decomposed when it was incubated withtrans-1,2-diaminocyclohexane-N, N, N′, N′-tetraacetic acid (CDTA) which chelates Ca2+. When radish root cell walls were washed with a buffered 1.5% (w/v) sodium dodesyl sulfate (SDS) solution (pH 6.5), 96%, 13% and 6% of Ca2+, B and pectic polysaccharides of the cell walls, respectively, were released and the cell wall swelled twice. Subsequent extraction with 50 mM CDTA (pH 6.5) of the SDS-washed cell walls further released 4%, 80% and 61% of Ca2+, B and pectic polysaccharides, respectively. Pectinase hydrolysis of the SDS-treated cell walls yielded a B-RG-II complex and almost all the remaining Ca2+ was recovered in the complex. This result suggests that cell-wall bound Ca2+ is divided into at least two fractions, one anchors the CDTA-soluble pectic polysaccharides into cell walls together with B, and the other may control the properties of the pectic gel. These studies demonstrate that B functions to retain CDTA-soluble pectic polysaccharides in cell walls through its binding to the RG-II regions in collaboration with Ca2+.  相似文献
