Abstract The gene coding for a collagen binding protein from Lactobacillus reuteri NCIB 11951 was cloned and sequenced. A genomic lambda library was constructed and recombinant plaques were screened using antisera raised against purified collagen binding proteins from the same L. reuteri strain. The positive plaques were subjected to sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Western blot analysis, which revealed the expression of a 29 kDa protein, which reacted with the antisera and bound 125 I-labelled type I collagen. The sequence of the corresponding gene, cnb showed that the collagen binding protein has sequence similarities to the solute binding component of bacterial ABC transporters. 相似文献

An OH-functional polyester has been acrylated via transesterification of ethyl acrylate, catalyzed by Candida antarctica lipase B (CalB) in two different preparations: Novozym 435 and immobilized on Accurel MP1000. The batch process resulted in incomplete acrylation as well as severe degradation of the polyester. A high degree of acrylation was achieved by optimization through the application of low pressure (15 kPa), continuous inflow of ethyl acrylate and continuous distillation to evaporate the by-product, ethanol. The enzyme preparations displayed good stability with half-lives of 180 and 324 h for Novozym 435 and CalB/MP1000, respectively. This translates into product yields of 3600 and 6200 times the weight of the catalyst, indicating that the enzyme will have a marginal impact on the total process cost. 相似文献

The role of sialic acid for the adhesion of Helicobacter pylori to gastric mucosa cells and/or to the mucin layer is still under debate. Several but not all H. pylori strains express a sialic acid-binding adhesin, specific for terminal α-2,3-sialic acid residues. Recently, the production of sialidase by H. pylori was reported [Dwarakanath, A.D. et al. (1995) FEMS Immunol. Med. Microbiol. 12, 213–216]. We analysed several strains isolated from gastric biopsies cultivated both in liquid media and on agar plates for sialidase. Activity of this enzyme was first assayed using the fluorigenic substrate 4-methylumbelliferyl-α-d-N-acetylneuraminic acid. Since the fluorimetric assay can give false-positive results caused by non-specific interactions with umbelliferyl-tagged substances, we used also the more sensitive and specific assay with sialyl-[3H]lactitol as a substrate. No evidence for sialidase activity of H. pylori strains, cultivated under both inducible and non-inducible conditions, was obtained. 相似文献

The population ecology of newts was investigated between 1969 and 1974 in two localities near Gothenburg, SW Sweden: the Gunnebo park pond (about 300 m2 ) where Triturus cristatus Laurenti and T. vulgaris L. are sympatric, and a rock-pool (6 m2 ) in Billdal with only T. vulgaris. At Gunnebo, a population of about 350 breeding T. cristatus was found with about as many males as females but males being more aquatic than females. An annual survival of 0.7–0.8 was estimated for the adults. The T. vulgaris population at the same locality was estimated to about 250–300 breeding adults with a sex ratio possibly in favour of males. The survival of adults was found to be extremely low and a very small percent of the adults seems to breed more than once. The Billdal T. vulgaris population was estimated to contain nearly 70 breeding adults with a sex ratio very near to 1:1. The annual survival of adults was found to be 0.5. The different survival of the two T. vulgaris populations are discussed with particular reference to predation. The age at which newts breed for the first time seems to vary; for T. vulgaris five years may well be the normal age. Factors that may limit the sizes of the populations, possible reasons for differences in frequency of occurrence in water, and possible predators are discussed. 相似文献

European eels (Anguilla anguilla) undertake spawning migrations of more than 5000 km from continental Europe and North Africa to frontal zones in the Sargasso Sea. Subsequently, the larval offspring are advected by large-scale eastward ocean currents towards continental waters. However, the Sargasso Sea is oligotrophic, with generally low plankton biomass, and the feeding biology of eel larvae has so far remained a mystery, hampering understanding of this peculiar life history. DNA barcoding of gut contents of 61 genetically identified A. anguilla larvae caught in the Sargasso Sea showed that even the smallest larvae feed on a striking variety of plankton organisms, and that gelatinous zooplankton is of fundamental dietary importance. Hence, the specific plankton composition seems essential for eel larval feeding and growth, suggesting a linkage between eel survival and regional plankton productivity. These novel insights into the prey of Atlantic eels may furthermore facilitate eel larval rearing in aquaculture, which ultimately may replace the unsustainable use of wild-caught glass eels. 相似文献

The purpose of the presentation is to interconnect and illuminate certain parts of metabolism regarding stress signalling and defensive functions, including secondary metabolism in intact plants and plant tissue cultures. Increased cell/tissue levels of reactive oxygen species like H2O2, O2
- and ·OH and the metabolism of glutathione, are linked to defensive/secondary metabolism and tissue differentiation. Special attention is paid to nicotinamide. A hypothetical role of nicotinamide and its metabolites as stress signals is also put forward especially in connection with hypomethylation of DNA. A role of DNA hypomethylation, as a link between various types of stressors and the induction of plant devensive metabolism, is discussed. The process of dog domestication is still somewhat unresolved. Earlier studies indicate that domestic dogs from all over the world have a common origin in Asia. So far, major histocompatibility complex (MHC) diversity has not been studied in detail in Asian dogs, although high levels of genetic diversity are expected at the domestication locality. We sequenced the second exon of the canine MHC gene DLA–DRB1 from 128 Asian dogs and compared our data with a previously published large data set of MHC alleles, mostly from European dogs. Our results show that Asian dogs have a higher MHC diversity than European dogs. We also estimated that there is only a small probability that new alleles have arisen by mutation since domestication. Based on the assumption that all of the currently known 102 DLA–DRB1 alleles come from the founding wolf population, we simulated the number of founding wolf individuals. Our simulations indicate an effective population size of at least 500 founding wolves, suggesting that the founding wolf population was large or that backcrossing has taken place. 相似文献
