Antibodies to fibronectin-binding proteins (FnBPs) of Staphylococcus aureus, including binding domain of FnBPA, the D region, or the A-C regions of FnBPB were produced in rabbits and mice. These antibodies were used to characterize cell-associated FnBPs of S. aureus strain Cowan I, S. aureus strain U320 and a coagulase-negative Staphylococcus capitis strain LK499 as well as extracellular FnBPs in culture supernatants of the strain U320. FnBPs of S. aureus were predominantly FnBPA, while FnBPB was hardly detected on the cells or in culture supernatant of these S. aureus strains. Moreover, S. capitis strain LK499 possessed different FnBP(s) compared to S. aureus because the antibodies to S. aureus FnBPs did not recognize FnBP(s) on S. capitis.  相似文献

BACKGROUND: Infection with Helicobacter pylori is associated with an increased risk of gastric cancer. Several studies have indicated that the association differs with strain type. We aimed to find out if infection with strains lacking the virulence factor CagA is linked to gastric cancer risk. MATERIALS AND METHODS: In a hospital-based case-control study, we collected sera from 100 case patients with a newly diagnosed gastric adenocarcinoma and 96 control patients with diseases unrelated to H. pylori status. Antibodies to H. pylori were analyzed by enzyme-linked immunosorbent assay (ELISA), and antibodies to CagA were detected by immunoblot. Logistic regression was used to obtain odds ratios (ORs) as estimates of relative risk, adjusted for potential confounding. RESULTS: Among the case patients, 81% were ELISA positive and 86% had antibodies to CagA. The corresponding numbers among the controls were 58% and 55%, respectively. ELISA positivity was associated with an increased risk of gastric adenocarcinoma compared to ELISA negativity (OR for gastric cancer regardless of site 3.9, 95% CI 1.9-8.2). The OR was 7.4 (95% CI 3.3-16.6) for CagA-positive relative to CagA-negative subjects. Among ELISA-positive subjects the presence of CagA antibodies increased the risk 3.6 times (95% CI 1.2-11.1). ELISA-positive CagA-negative infections were associated with a fourfold increased risk (OR = 4.2, 95% CI 1.0-17.0) compared to no infection (ELISA-negative and CagA-negative). CONCLUSIONS: Although patients with antibodies to CagA have the greatest risk of developing gastric cancer, those with CagA-negative infections run a significantly greater risk than uninfected persons.  相似文献

The population ecology of newts was investigated between 1969 and 1974 in two localities near Gothenburg, SW Sweden: the Gunnebo park pond (about 300 m2) where Triturus cristatus Laurenti and T. vulgaris L. are sympatric, and a rock-pool (6 m2) in Billdal with only T. vulgaris. At Gunnebo, a population of about 350 breeding T. cristatus was found with about as many males as females but males being more aquatic than females. An annual survival of 0.7–0.8 was estimated for the adults. The T. vulgaris population at the same locality was estimated to about 250–300 breeding adults with a sex ratio possibly in favour of males. The survival of adults was found to be extremely low and a very small percent of the adults seems to breed more than once. The Billdal T. vulgaris population was estimated to contain nearly 70 breeding adults with a sex ratio very near to 1:1. The annual survival of adults was found to be 0.5. The different survival of the two T. vulgaris populations are discussed with particular reference to predation. The age at which newts breed for the first time seems to vary; for T. vulgaris five years may well be the normal age. Factors that may limit the sizes of the populations, possible reasons for differences in frequency of occurrence in water, and possible predators are discussed.  相似文献

European eels (Anguilla anguilla) undertake spawning migrations of more than 5000 km from continental Europe and North Africa to frontal zones in the Sargasso Sea. Subsequently, the larval offspring are advected by large-scale eastward ocean currents towards continental waters. However, the Sargasso Sea is oligotrophic, with generally low plankton biomass, and the feeding biology of eel larvae has so far remained a mystery, hampering understanding of this peculiar life history. DNA barcoding of gut contents of 61 genetically identified A. anguilla larvae caught in the Sargasso Sea showed that even the smallest larvae feed on a striking variety of plankton organisms, and that gelatinous zooplankton is of fundamental dietary importance. Hence, the specific plankton composition seems essential for eel larval feeding and growth, suggesting a linkage between eel survival and regional plankton productivity. These novel insights into the prey of Atlantic eels may furthermore facilitate eel larval rearing in aquaculture, which ultimately may replace the unsustainable use of wild-caught glass eels.  相似文献

The purpose of the presentation is to interconnect and illuminate certain parts of metabolism regarding stress signalling and defensive functions, including secondary metabolism in intact plants and plant tissue cultures. Increased cell/tissue levels of reactive oxygen species like H2O2, O2 - and ·OH and the metabolism of glutathione, are linked to defensive/secondary metabolism and tissue differentiation. Special attention is paid to nicotinamide. A hypothetical role of nicotinamide and its metabolites as stress signals is also put forward especially in connection with hypomethylation of DNA. A role of DNA hypomethylation, as a link between various types of stressors and the induction of plant devensive metabolism, is discussed. Microscale distributions of plankton around the pycnocline werestudied over a 24 h period in the southern Kattegat, using agradient sampler which collects 20 samples over a 3 m depthinterval. Moderately elevated concentrations of phyto- and zooplanktonwere observed at the pycnocline. Microscale variance was highestfor adult copepods. Nauplii and copepodites were equally wellrepresented by sampling with ordinary vertical 5 1 water bottlesas with the horizontal 1.5 1 bottles of the gradient sampler.Adult copepods were underestimated by the vertical bottles.No vertical migration of dinoflagellates was observed over the3 m interval covered by the gradient sampler. Microscale correlationsbetween copepods and phytoplankton within the gradient samplerwere weak. Copepodites (mainly Oithona sp.) and the dinoflagellateProrocentrum micans showed the best correlation.  相似文献

Abstract Immunocompetent and immunodeficient BALB/cA mice were fed orally with 108 colony forming units of 2-day-old spiral or coccoid (12 days old) Helicobacter pylori strain NCTC 11637. Immunocompetent BALB/cA mice were also fed orally with decreasing numbers of spiral or coccoid forms of H. pylori . The gastrointestinal colonisation process was monitored for 34 days post-infection by heparin magnetic separation and subsequent enzyme immunoassay (EIA) for the detection of the H. pylori cells. Both mice types were colonised with H. pylori . The coccoid form of H. pylori gave higher EIA absorbance values and more efficient colonisation in the mice than the spiral form. Immunocompetent BALB/cA mice fed with the coccoid form of H. pylori exhibited an acute inflammation process in histopathological samples from the stomachs. In conclusion, H. pylori can infect both immunocompetent as well as immunodeficient BALB/cA mice and coccoids (viable but non-culturable) obtained after 12 days of culturing can infect BALB/cA mice.  相似文献

The elucidation of the entire complement of genes encoding protein tyrosine phosphatases (PTPs) in human genome, the human 'PTPome', has made it possible to experimentally address the entire family in an unbiased manner. Here we describe a functional RNA interference-based assay, in which we evaluate 87 of the known 107 PTPs for effects on cell survival in a high throughput manner. The details of assay rationale and design, instrumentation, pitfalls, data analysis, and further validation steps are described. We also discuss the suitability of this technology for further assay development and application to other functional read-outs and signaling pathways.  相似文献
