The entomopathogenic fungus Pandora neoaphidis is a recognized pathogen of aphids, causes natural epizootics in aphid populations, and interacts and competes with aphid predators and parasitoids. Survival of entomophthoralean fungi in periods of unsuitable weather conditions or lack of appropriate host insects is accomplished mainly by thick-walled resting spores (zygospores or azygospores). However, resting spores are not known for some entomophthoralean species such as P. neoaphidis. Several hypotheses of P. neoaphidis winter survival can be found in the literature but so far these hypotheses do not include the presence of resting spores. Resting spores were found in an aphid population where P. neoaphidis was the only entomophthoralean fungus observed during surveys conducted in organic horticultural crops in greenhouses and open fields in Buenos Aires province, Argentina. This study sought to use molecular methods to confirm that these resting spores were, in fact, those of P. neoaphidis while further documenting and characterizing these resting spores that were produced in vivo in aphid hosts. The double-walled resting spores were characterized using light and transmission electron microscopy. The Argentinean resting spores clustered together with P. neoaphidis isolates with bootstrap values above 98 % in the small subunit ribosomal RNA (SSU rRNA) sequence analysis and with bootstrap values above 99 % the Internal Transcribed Spacer (ITS) II region sequence analysis. siRNA depletion of BRCA1, but not BRCA2, causes increased genome instability in Fanconi anemia cells
BRCA1 and BRCA2 proteins act in repair of interstrand crosslinks (ICLs) and maintenance of genome stability and are known to be part of the Fanconi anemia (FA) pathway. We have investigated the role of the BRCA1 and BRCA2 genes in genome stability following ICL damage in normal and FA cells. To circumvent cell lethality of complete disruptions in BRCA1 or BRCA2, small inhibitory RNA (siRNA) was used to transiently deplete the expression of the proteins. Using chromosomal stability after ICL damage as the end point, we find that BRCA1 functions in more than just the FA pathway for genome maintenance, whereas BRCA2 appears to act predominantly in the FA pathway. Depletion of BRCA1 causes a marked decrease, although not a complete absence of, ubiquitination of FANCD2. In contrast to BRCA1, BRCA2 is not needed for normal ubiquitination of FANCD2 after DNA damage, a requirement for the FA pathway to function. Thus, BRCA2 is epistatic to FA genes for ICL repair, but not for damage-induced modification of FANCD2 and may act downstream form FANCD2. 相似文献

Physicochemical gut conditions and the composition and topology of the intestinal microbiota in the major gut compartments of the root-feeding larva of the European cockchafer (Melolontha melolontha) were studied. Axial and radial profiles of pH, O2, H2, and redox potential were measured with microsensors. Terminal restriction fragment length polymorphism (T-RFLP) analysis of bacterial 16S rRNA genes in midgut samples of individual larvae revealed a simple but variable and probably nonspecific community structure. In contrast, the T-RFLP profiles of the hindgut samples were more diverse but highly similar, especially in the wall fraction, indicating the presence of a gut-specific community involved in digestion. While high acetate concentrations in the midgut and hindgut (34 and 15 mM) corroborated the presence of microbial fermentation in both compartments, methanogenesis was confined to the hindgut. Methanobrevibacter spp. were the only methanogens detected and were restricted to this compartment. Bacterial 16S rRNA gene clone libraries of the hindgut were dominated by clones related to the Clostridiales. Clones related to the Actinobacteria, Bacillales, Lactobacillales, and γ-Proteobacteria were restricted to the lumen, whereas clones related to the β- and δ-Proteobacteria were found only on the hindgut wall. Results of PCR-based analyses and fluorescence in situ hybridization of whole cells with group-specific oligonucleotide probes documented that Desulfovibrio-related bacteria comprise 10 to 15% of the bacterial community at the hindgut wall. The restriction of the sulfate-reducer-specific adenosine-5′-phosphosulfate reductase gene apsA to DNA extracts of the hindgut wall in larvae from four other populations in Europe suggested that sulfate reducers generally colonize this habitat. 相似文献

The arctic fox Alopex lagopus excavates its dens in gravely ridges and hillocks, and creates a local environment quite distinct from the surrounding tundra or heath landscape. In northern Sweden, the vegetation of 18 dens of the arctic fox was investigated, as well as reference areas off the dens but in geologically and topographically similar locations. The species composition showed considerable differences between den and reference areas, with grasses and forbs occurring more abundantly on the dens, and evergreen dwarf-shrubs occurring more in reference areas. The effect of the foxes' activities is thought to be either through mechanical soil disturbance, or through nutrient enrichment via scats, urine, and carcasses. This was expected to result in differences in plant traits with key functional roles in resource acquisition and regeneration, when comparing dens with reference areas. We hypothesised that the community mean of specific leaf area (SLA) would differ if nutrient enrichment was the more important effect, and that seed weight, inversely proportional to seed number per ramet and hence dispersal ability, would differ if soil disturbance was the more important effect. Specific leaf area showed a significant difference, indicating nutrient enrichment to be the most important effect of the arctic fox on the vegetation on its dens. Arctic foxes act as ecosystems engineers on a small scale, maintaining niches for relatively short-lived nutrient demanding species on their dens in spite of the dominance of long-lived ericaceous dwarf-shrubs in the landscape matrix. Obesity is associated with an increased risk of developing insulin resistance, Type 2 diabetes, and cardiovascular disease. Reports have suggested that adipose tissue-derived cytokines such as tumor necrosis factor-alpha, interleukin-6 and interleukin-8 could be involved in the development of these health complications. Since estrogen has been suggested to attenuate the development of atherosclerosis and cardiovascular disease, we investigated whether ovariectomy affected the production and release of these three adipose tissue-derived cytokines with and without estrogen replacement in vivo and in vitro. Female Wistar rats were submitted to either a) ovariectomy, b) ovariectomy and estrogen replacement, or c) sham operation. After five months, animals were sacrificed and parametrial adipose tissue was removed and incubated for up to 24 hours with either interleukin-1beta (IL-1beta) (5 micro g/l), dexamethasone (50 nM) or estrogen (50 nM). Ovariectomy significantly increased interleukin-6 gene expression (p < 0.05) as well as interleukin-8 protein levels (p < 0.05) and gene expression (p < 0.05) in the adipose tissue, and estrogen replacement significantly reversed this increase (p < 0.05). However, no direct effects of estrogen were found in in vitro adipose tissue incubations. Neither ovariectomy nor estrogen replacement had any effects on tumor necrosis factor-alpha protein levels or gene expression. In conclusion, estrogen-deficient rats were found to have increased production of interleukin-6 and interleukin-8, which could be attenuated by estrogen-replacement. Since estrogen is suggested to be anti-atherosclerotic, this effect might be caused by a reduction in cytokine production from the adipose tissue. 相似文献

Plant species richness in grasslands: the relative importance of contemporary environment and land-use history since the Iron Age 总被引:3,自引:0,他引:3
The richness of vascular plants in all patches of dry semi-natural grassland within a landscape was investigated. The patch level richness was scaled up to the level of parish to match data on land-use intensity from Iron Age through historical time. Three measures of diversity were obtained: the local species pool, an area-independent diversity measure (the slope of the species number vs log area regression line), and the β-diversity, A gradient through the study area in the density of grassland and a corresponding clinal variation in grassland plant species diversity were found. Explanatory models were built by partial least squares regression and conventional stepwise multiple regression. Data on contemporary environmental conditions in the grassland patches and in the parish as a whole were added first, and then data on continuously older times in sequence. In the stepwise regression analyses the variables were also added in the opposite sequence. The results show that contemporary conditions are able to explain the major part of the variation in all diversity measures. Variables concerning former land-use do, however, add significantly to the explanation of variation in local species pool and in the area-independent diversity, but not in β-diversity. It is concluded that patterns of land-use intensity from the Iron Age and onwards have contributed significantly to the shaping of local species pools, and thereby the richness of grassland communities. 相似文献
