Bovine papillomavirus type 1 (BPV1), Epstein-Barr virus (EBV), and human herpesvirus 8 genomes are stably maintained as episomes in dividing host cells during latent infection. The mitotic segregation/partitioning function of these episomes is dependent on single viral protein with specific DNA-binding activity and its multimeric binding sites in the viral genome. In this study we show that, in the presence of all essential viral trans factors, the segregation/partitioning elements from both BPV1 and EBV can provide the stable maintenance function to the mouse polyomavirus (PyV) core origin plasmids but fail to do so in the case of complete PyV origin. Our study is the first which follows BPV1 E2- and minichromosome maintenance element (MME)-dependent stable maintenance function with heterologous replication origins. In mouse fibroblast cell lines expressing PyV large T antigen (LT) and either BPV1 E2 or EBV EBNA1, the long-term episomal replication of plasmids carrying the PyV minimal origin together with the MME or family of repeats (FR) element can be monitored easily for 1 month under nonselective conditions. Our data demonstrate clearly that the PyV LT-dependent replication function and the segregation/partitioning function of the BPV1 or EBV are compatible in certain, but not all, configurations. The quantitative analysis indicates a loss rate of 6% per cell, doubling in the case of MME-dependent plasmids, and 13% in the case of FR-dependent plasmids in nonselective conditions. Our data clearly indicate that maintenance functions from different viruses are principally interexchangeable and can provide a segregation/partitioning function to different heterologous origins in a variety of cells. 相似文献

To characterize the mitochondrial DNA (mtDNA) variation in Han Chinese from several provinces of China, we have sequenced the two hypervariable segments of the control region and the segment spanning nucleotide positions 10171-10659 of the coding region, and we have identified a number of specific coding-region mutations by direct sequencing or restriction-fragment-length-polymorphism tests. This allows us to define new haplogroups (clades of the mtDNA phylogeny) and to dissect the Han mtDNA pool on a phylogenetic basis, which is a prerequisite for any fine-grained phylogeographic analysis, the interpretation of ancient mtDNA, or future complete mtDNA sequencing efforts. Some of the haplogroups under study differ considerably in frequencies across different provinces. The southernmost provinces show more pronounced contrasts in their regional Han mtDNA pools than the central and northern provinces. These and other features of the geographical distribution of the mtDNA haplogroups observed in the Han Chinese make an initial Paleolithic colonization from south to north plausible but would suggest subsequent migration events in China that mainly proceeded from north to south and east to west. Lumping together all regional Han mtDNA pools into one fictive general mtDNA pool or choosing one or two regional Han populations to represent all Han Chinese is inappropriate for prehistoric considerations as well as for forensic purposes or medical disease studies. 相似文献

Optimal behavioural decisions are expected to depend on various state variables, such as physiological condition or age. In insects, empirical evidence of the effect of adult age on oviposition selectivity is mixed. Consistently, optimality models – which primarily incorporate the effects of egg load and senescence – fail to provide universal predictions. Here we propose that spatial variation in mortality rates creates an additional mechanism able to select for an increase in selectivity with age. Females can be selected to use the fact of having reached an advanced age as a cue of low mortality rates in their environment. Older females may thus be less time-limited, and can afford for more careful host selection. This is because variation in mortality rates can cause a positive correlation between individual age, and expected residual life span. We present a simulation model that formalises the verbal argument presented above, and discuss the findings in the more general context of the dependence of reproductive output on age. 相似文献

Five complete bacterial genome sequences have been released to the scientific community. These include four (eu)Bacteria,
Haemophilus influenzae, Mycoplasma genitalium, M. pneumoniae, and Synechocystis PCC 6803, as well as one Archaeon, Methanococcus jannaschii. Features of organization shared by these genomes are likely to have arisen very early in the history of the bacteria and
thus can be expected to provide further insight into the nature of early ancestors. Results of a genome comparison of these
five organisms confirm earlier observations that gene order is remarkably unpreserved. There are, nevertheless, at least 16
clusters of two or more genes whose order remains the same among the four (eu)Bacteria and these are presumed to reflect conserved
elements of coordinated gene expression that require gene proximity. Eight of these gene orders are essentially conserved
in the Archaea as well. Many of these clusters are known to be regulated by RNA-level mechanisms in Escherichia coli, which supports the earlier suggestion that this type of regulation of gene expression may have arisen very early. We conclude
that although the last common ancestor may have had a DNA genome, it likely was preceded by progenotes with an RNA genome.

We identified the lactic acid bacteria within rye sourdoughs and starters from four bakeries with different propagation parameters and tracked their dynamics for between 5–28 months after renewal. Evaluation of bacterial communities was performed using plating, denaturing gradient gel electrophoresis, and pyrosequencing of 16S rRNA gene amplicons. Lactobacillus amylovorus and Lactobacillus frumenti or Lactobacillus helveticus, Lactobacillus pontis and Lactobacillus panis prevailed in sourdoughs propagated at higher temperature, while ambient temperature combined with a short fermentation cycle selected for Lactobacillus sanfranciscensis, Lactobacillus pontis, and Lactobacillus zymae or Lactobacillus helveticus, Lactobacillus pontis and Lactobacillus zymae. The ratio of species in bakeries employing room-temperature propagation displayed a seasonal dependence. Introduction of different and controlled propagation parameters at one bakery (higher fermentation temperature, reduced inoculum size, and extended fermentation time) resulted in stabilization of the microbial community with an increased proportion of L. helveticus and L. pontis. Female oviposition decisions in insects may strongly affect offspring growth and survival, and thus determine population performance. In this study, we examined oviposition site selection in the xerophilous ecotype of the endangered myrmecophilous butterfly Phengaris (=Maculinea) alcon (Lepidoptera, Lycaenidae) in Estonia, at the northern distribution margin of both the butterfly and its host plant, Gentiana cruciata L. Egg distribution on individual host plants appeared to be highly uneven: plants carrying high egg loads contrasted to a high proportion of host plants without any or with a few eggs. Host plant use for oviposition was strongly dependent on plant characteristics and environmental context. Host plant height relative to the surrounding vegetation rather than the absolute height of host plants was a key factor determining the use of particular host plant individuals for oviposition. In particular, plants protruding above surrounding vegetation had a higher probability of being used for oviposition, and carried more eggs. Additionally, the number of eggs laid on individual host plants was positively associated with the presence of flowers and the number of shoots. More aggregated host plants received fewer eggs than those with less conspecifics around. Feeding damage by wild herbivores, found in a substantial proportion of the butterfly’s host plants, strongly reduced the number of eggs on individual plants. Our results underline the need to assure that butterfly’s host plants do not become overgrown by surrounding vegetation. Best practices for opening vegetation around host plants may need further studies that explicitly account for butterfly’s host ants—their abundance in relation to vegetation height and their response to opening vegetation. 相似文献

Understanding ecological requirements of endangered species is a primary precondition of successful conservation practice. Regrettably, we know surprisingly little about the life history of numerous threatened insects, and about their use of larval host plants in particular. The brown butterflies (Nymphalidae: Satyrinae) have traditionally been considered polyphagous on grasses and indiscriminatory in their oviposition behavior. However, detailed studies on several species have revealed local specialization in host plant use as well as the decisive role of microlimatic conditions as determinants of habitat quality. The present study addresses host plant relationships in the endangered brown butterfly Coenonympha hero (L.) at the northern limit of its European distribution. We combine laboratory-based host preference and performance tests with an analysis of microhabitat use by adult butterflies in the field. Both lines of evidence suggest that C. hero is polyphagous enough not to be associated with one particular host species. Oviposition choices of C. hero are not driven by host plant species but rather by structural characteristics of the substrate. The preferred rigid needle-like structures may serve as cues of ‘transparent’ vegetation which allows the larvae to benefit from sunlight reaching the lower strata of the tuft. Our results suggest that conservation efforts should prioritize microclimatic parameters, rather than the presence of any particular host plant species, as decisive determinants of habitat quality in C. hero. 相似文献

Specificity (= the degree of ecological specialisation) is one of the fundamental concepts of the science of ecology. Ambiguities on how to define and measure specificity have however complicated respective research efforts. Here we propose that, in insects, a behavioural trait – adult oviposition latency in captivity without a favourable host plant – correlates with a species’ specificity in larval host use. In the absence of a suitable host, monophagous insects are expected to wait for a long time before commencing oviposition, with the long waiting time corresponding to careful host location behaviour in nature. Polyphagous insects, in contrast, should be selected for an increased oviposition rate at the expense of the quality of oviposition substrate encountered and will on average have a short latency time. Using experimentally derived data on oviposition latency, we performed a phylogenetically informed analysis based on Bayesian inference to show that this variable correlates with host specificity (larval diet breadth) in a sample of north European species of geometrid moths. A closely related index – the probability to lay any eggs on an unfavourable substrate – shows an analogous pattern. To provide an example of how these indices can be applied, we compare our sample of geometrid moths from northern Europe with a sample from equatorial Africa. A comparative analysis based on an original phylogenetic reconstruction found no differences between the two study sites in parameters of oviposition behaviour. We conclude that behavioural tests can provide information about ecological interactions when the latter can not be directly recorded. Our example study also hints at the possibility that host specificity of herbivores is not necessarily higher in a tropical region compared to a temperate one. 相似文献

Intraspecific color polymorphism is widespread in insects, and various mechanisms have been proposed to explain its maintenance. Some explanations rely on the effect of body color on the organism's thermal physiology. Darker individuals accumulate solar energy more efficiently, and therefore, dark body coloration in insects is frequently presumed to be an adaptation to low temperature conditions. However, it is largely unclear what is the importance of the thermal biology in comparison to other potential selective forces on body coloration. In this study, we evaluated the role of temperature as a potential selective factor maintaining color polymorphism in aposematic larvae of the moth Orgyia antiqua L. It was found that darker, and thus less aposematic, larvae accumulated solar energy more efficiently. However, in a set of laboratory and outdoor experiments, we found no evidence of temperature-dependent performance of different color morphs or in development of different morphs induced by rearing temperature. We conclude that the effects related to thermal physiology are not likely important determinants of optimal coloration in O. antiqua. The reasons may lie in high mobility of the larvae, which allows for effective behavioral thermoregulation, which is also shown in this study. Our results caution against an uncritical extrapolation of results obtained for model organisms and indicate the need for giving more attention to the species-specific ecological background in ecophysiological studies. 相似文献
