The effect of long-term mercury pollution on the soil microbial community   总被引:1,自引:0,他引:1  
The effect of long-term exposure to mercury on the soil microbial community was investigated in soil from three different sites along a pollution gradient. The amount of total and bioavailable mercury was negatively correlated to the distance from the center of contamination. The size of the bacterial and protozoan populations was reduced in the most contaminated soil, whereas there was no significant difference in fungal biomass measured as chitinase activity. Based on the number of colony morphotypes, moreover, the culturable bacterial population was structurally less diverse and contained a higher proportion of resistant and fast-growing forms. The profiles of amplified 16S rDNA sequences obtained from community DNA by denaturating gradient gel electrophoresis (DGGE) also reflected the altered community structure and decreased diversity along the mercury gradient as expressed in terms of the number and abundance of bands. The functional potential of the microbial population measured as sole carbon source utilization by Ecoplates((R)) differed between the soils, but there was no change in the number of substrates utilized. The observed changes in the different soil microbial populations are probably a combination of both direct and indirect effects of the mercury contamination.  相似文献

There exist many methods to detect recombination or mosaic structure in a sample of DNA sequences. But how reliable are they? Four methods were investigated with respect to their power to detect recombination in simulated samples with different amounts of recombination and mutation. In addition, we investigated the impact of the shape of the underlying genealogy on their performances. We found that the methods detected far fewer recombinations than were theoretically possible and that methods based on the principle of incompatibility in general had more power than methods that did not make use of this principle explicitly. This seemed, in particular, to be the case for phylogenies generated under population expansion scenarios which result in long branches at the tips and small deep branches. In addition to the results obtained through simulations, a series of new theoretical results on recombination is presented.  相似文献

In this paper we review various strategies for gene transfer into the skin, their applications and show some of our own examples. The skin is the most accessible somatic tissue. Inherited skin diseases, such as epidermolysis bullosa and ichthyosis, and various systemic metabolic disorders are under investigation as potential candidate diseases for cutaneous gene transfer. Research is directed against genetic therapy of wounds and malignancies as well. So far, cutaneous gene transfer only has been used experimentally, but several clinical trials are under preparation.  相似文献

Despite the common occurrence of ontogenetic niche shifts, their consequences for morphological adaptations have been little studied. To address this question, we studied morphological adaptations related to ontogenetic niche shifts in Eurasian perch (Perca fluviatilis) in eight lakes that varied in density of benthic resources and planktivorous fish biomass. Perch start to feed on pelagic zooplankton, then shift to benthic resources at intermediate sizes, and finally, when large enough, mainly feed on fish. These three functional niches over ontogeny are expected to set constraints on the morphology and size-specific growth of perch. The growth of perch was negatively related to planktivorous fish biomass in the zooplanktivorous niche, but positively related to planktivorous fish biomass in the piscivorous niche. The number of gill rakers of perch was negatively related to the biomass of planktivorous fish, providing evidence for the occurrence of character displacement as a consequence of competition in the zooplanktivorous niche. Perch in lakes with low densities of predator-sensitive macroinvertebrates had greater body height measurements and a larger mouth early during ontogeny. This pattern is suggested to be a result of a selection for increased efficiency in the benthic niche when the availability of benthic resources is low. Perch in lakes with a high biomass of planktivorous fishes had fusiform body morphology, a thicker tail and a larger mouth then the average piscivorous perch. The different responses of perch morphology to variation in the availability of benthic resources compared to variation in planktivore biomass are suggested to be partly because the availability of the former resource to a larger extent is set by abiotic conditions (humic content). We suggest that the key factors affecting size-specific growth and body morphology of perch in the system studied are the availability of resources in the benthivorous and piscivorous niches. We also provide evidence for morphological trade-offs, especially between the benthivorous and the piscivorous ontogenetic niches. Received: 6 July 1999 / Accepted: 8 September 1999  相似文献

1200 measurements of eelgrass ( Zostera marina ) biomass, shoot density and cover along 19 depth gradients in Øresund, located between Denmark and Sweden, were analysed to characterise growth of eelgrass in relation to depth. The large data set allowed analyses of boundaries of distribution as well as of average trends. Natural variability is large in shallow water where populations are disturbed by wave action and other physical parameters. Models based on average values, therefore, did not adequately describe growth regulation by resources, and only explained a minor part (up to 30%) of the overall variation in data. In contrast, boundary functions, which describe the upper bounds of distributions, focus on the variation produced by the ultimately growth-regulating resource, and therefore provide models with high predictive power. An exponential model explained up to 90% of the variation in upper bounds of eelgrass shoot density as a function of depth and indicated that shoot density was ultimately regulated by light availability. The boundary functions demonstrated that eelgrass shoot density, biomass and cover followed markedly different patterns as functions of depth and were affected differently by the factors governing their distribution. In addition, boundary functions revealed informative spatial structures in data and illustrated whether a given general trend was caused by changes in maximum values, minimum values or both. For example, upper and lower boundaries of biomass-shoot density relations changed markedly with depth, demonstrating depth-related changes in intraspecific succession and competition patterns. Boundary functions are therefore suggested as a promising tool for analysing ultimate regulating factors of distribution and growth of organisms when large data sets are available.  相似文献

The effects of treating Neepawa wheat straw with anhydrous ammonia (35 kg t?1), sodium hydroxide (50 kg t?1) and pelleting were evaluated in two experiments. The diets consisted of 51% rolled barley, 40% straw, 4% rapeseed meal, 2% tallow, plus minerals and a vitamin supplement. Straw was treated as follows: (1) shredded into 2.0-cm lengths; (2) shredded into 0.64-cm lengths and pelleted; (3) ammoniated and shredded; (4) ammoniated and pelleted; (5) sodium hydroxide and pelleted; (6) ammoniated, sodium hydroxide and pelleted.Apparent digestibility was measured with six steers per treatment. Pelleting had no effect on organic matter (OM) digestibility, but decreased neutral detergent fibre (NDF) and cellulose digestion. Respective increases for OM and NDF digestibility following chemical treatment of the straw were as follows: ammoniation — 15 and 17%; sodium hydroxide — 4 and 13%.The six diets were given to 144 Hereford steers (240 kg) for 92 days in the second experiment. Feed consumption ranged from 8.51 to 10.39 kg day?1 for diets 1 and 4, and body weight gains ranged from 0.83 to 1.26 kg day?1 for diets 1 and 6, respectively. Respective increases for intake, gain and feed efficiency compared to untreated shredded straw were as follows: pelleting — 11, 34 and 17%; ammoniation — 12, 36 and 17%. Sodium hydroxide treatment prior to pelleting improved intake, gain and feed efficiency by 5, 10 and 5%, respectively. There was no evidence of additive effects between ammoniation and either pelleting or alkali treatment, though treatment with sodium hydroxide prior to pelleting did result in the highest gains and feed efficiency.  相似文献

The glycoprotein mucin was isolated from the sputum of patients with chronic obstructive bronchitis. The fractionation procedure included treatment with 6M urea at pH 12.5 followed by gel filtration in 6M urea at neutral pH. (1) Using a low-shear Cartesian diver viscometer, we found that the mucin intrinsic viscosity equals (0.32 ± 0.03) L/g in 1000 mM NaCl solution increasing to (12 ± 3) L/g in 0.1 mM NaCl (pH 7 and 20°C). (2) The relaxation of electrically induced birefringence in mucin solutions was measured and the relaxation spectrum calculated using a Fourier-transform deconvoltion method. We found that the dominant relaxation time increased from 1 to 150 μs when the exitation pulse duration used was increased from 2 to 300 μs. (3) Mucin was vacuum-dried from glycerol-containing solutions followed by low-angle rotary shadowing and electron microscopy. Mucin was found to be unbranched, with contour lengths ranging from 300 to 2500 nm and with an average of 900 nm. Our result indicate that mucin is an extended and flexible molecule with Kuhn length 0.3–0.5% of the contour length.  相似文献
