Introduction
Recent evidence suggests that uric acid (UA), regardless of crystal deposition, may play a direct pathogenic role in renal disease. We have shown that UA is an independent predictor of hypertension and cardiovascular disease (CVD), and that CVD risk factors associate with renal dysfunction, in patients with rheumatoid arthritis (RA). Five patients at risk for primary central nervous system lymphoma (PCNSL) recurrence were treated with high-dose methylprednisolone,
(HDMP) to prevent ‘trafficking’ of malignant lymphocytes into the central nervous system (CNS). HDMP was chosen because of
its ability to stabilize the ‘blood brain barrier (BBB)’. Three men with newly diagnosed PCNSL, ages 62, 76 and 78 y, whose
survival was projected to be 6.6 months, began treatment after achieving complete response (CR) to initial radiation therapy
alone and survived 27, 37 and 59 months after treatment. In none was death from recurrent disease in CNS but one patient did
die of systemic non-Hodgkin’s lymphoma (NHL) five years after PCNSL diagnosis. A 20 y old man was treated with HDMP after
successful combined modality therapy and is alive 75+months after initial diagnosis without evidence of disease recurrence.
A 34 y old man relapsed after combined modality initial treatment and failed to respond to HDMP when treatment was begun after
unsuccessful salvage therapy; he died of disease 12 months after initial diagnosis. There were no treatment complications.

Many tumors present with increased activation of the phosphatidylinositol 3-kinase (PI3K)-PtdIns(3,4,5)P(3)-protein kinase B (PKB/Akt) signaling pathway. It has long been thought that the lipid phosphatases SH2 domain-containing inositol-5'-phosphatase 1 (SHIP1) and SHIP2 act as tumor suppressors by counteracting with the survival signal induced by this pathway through hydrolysis or PtdIns(3,4,5)P(3) to PtdIns(3,4)P(2). However, a growing body of evidence suggests that PtdInd(3,4)P(2) is capable of, and essential for, Akt activation, thus suggesting a potential role for SHIP1/2 enzymes as proto-oncogenes. We recently described a novel SHIP1-selective chemical inhibitor (3α-aminocholestane [3AC]) that is capable of killing malignant hematologic cells. In this study, we further investigate the biochemical consequences of 3AC treatment in multiple myeloma (MM) and demonstrate that SHIP1 inhibition arrests MM cell lines in either G0/G1 or G2/M stages of the cell cycle, leading to caspase activation and apoptosis. In addition, we show that in vivo growth of MM cells is blocked by treatment of mice with the SHIP1 inhibitor 3AC. Furthermore, we identify three novel pan-SHIP1/2 inhibitors that efficiently kill MM cells through G2/M arrest, caspase activation and apoptosis induction. Interestingly, in SHIP2-expressing breast cancer cells that lack SHIP1 expression, pan-SHIP1/2 inhibition also reduces viable cell numbers, which can be rescued by addition of exogenous PtdIns(3,4)P(2). In conclusion, this study shows that inhibition of SHIP1 and SHIP2 may have broad clinical application in the treatment of multiple tumor types. 相似文献

Human activities are causing rapid environmental change at a global scale. Urbanization is responsible for some of the most extreme human‐altered habitats and is a known driver of evolutionary change, but evidence and understanding of these processes is limited. Here, we investigate the potential underlying mechanisms contributing to the contemporary evolution of migration behaviour in the Eurasian blackcap (Sylvia atricapilla). Blackcaps from central Europe have been wintering in urban areas of Britain with increasing frequency over the past 60 years, rather than migrating south to the Mediterranean. It has been hypothesized that the popularization of providing supplementary foods for wild birds within Britain may have influenced this marked migratory change, but quantifying the selective forces shaping evolutionary changes remains challenging. Using a long‐term national scale data set, we examine both the spatial distribution and interannual variation in blackcap wintering behaviour in Britain in relation to supplementary food availability and local climate. Over a 12‐year period, we show that blackcaps are becoming increasingly associated with the provision of supplementary foods in British gardens, and that the reliability of bird food supplies is influencing their winter distribution at a national scale. In addition, local climatic temperatures and broader scale weather variation are also important determinants of blackcap wintering patterns once they arrive in Britain. Based on our findings, we conclude that a synergistic effect of increased availability of feeding resources, in the form of garden bird food, coupled with climatic amelioration, has enabled a successful new wintering population to become established in Britain. As global biodiversity is threatened by human‐induced environmental change, this study presents new and timely evidence of the role human activities can play in shaping evolutionary trajectories. 相似文献

Cleavage of reduced, carboxymethylated, delipidated CA2+-transporting ATPase protein from rabbit sarcoplasmic reticulum with dimethyl sulphoxide/HBr yielded two long peptides (38 and 73 residues), distinct from the known major sequences of the ATPase. The longer peptide contained at least two cysteine residues, which were disulphide-linked in the native protein. It was therefore derived from the B-fragment of the ATPase in which the disulphides had previously been located. It probably formed a loop on the luminal side of the membrane, spanning two membrane-buried tryptophan residues. The N-terminal sequence of this peptide, (Trp)-Phe-Met-Tyr-Ala, forms the basis for an oligodeoxynucleotide probe, the use of which to identify cDNA corresponding to the ATPase is described elsewhere [MacLennan, Brandl, Korczak & Green (1985) Nature (London) 316, 696-700]. 相似文献
