When white light irradiation inhibits shoot growth in derooted pea ( Pisum sativum L. cv. Alaska) cuttings, it decreases tissue tension, a driving force for shoot growth, by making the cell wall of the inner tissues mechanically rigid. To elucidate the mechanism by which light affects the mechanical properties of the cell wall in the inner tissues, its effect on the chemical properties of the cell walls was studied by analyzing qualitatively and quantitatively cell wall polysaccharides in the epdidermis and inner tissue of pea epicotyls grown in both dark and light. The amount of polysaccharides per subhook in the cell walls of both tissues increased during a 4-h dark incubation. Light suppressed the increase in hemicellulosic (HC)-II and cellulosic polysaccharides in the inner tissues, while it did not affect the increase in other wall fractions in either the epidermal or subepidermal tissues. No light effect was observed on the neutral sugar compositions of pectin, HC-I or HC-II fractions in either of the tissues. Light increased the mass-average molecular mass of xyloglucan and rhamnoarabinogalactan in HC-II fractions in the inner tissues, while such an effect was not observed in the epidermis. These facts suggest that the light-induced decrease in the tissue tension in pea epicotyls is caused by an increase in the molecular size of xyloglucan, rhamnoarabinogalactan in the HC-II fraction and/or the suppression of the synthesis of HC-II and cellulosic polysaccharides in the inner tissues.  相似文献

Eker rat known as a model of hereditary renal carcinoma (RC) is an example of Mendelian dominantly inherited predisposition to a specific cancer in experimental animals. We investigate the effects of simulated space radiation on carcinogenesis using HIMAC. We estimated RBE from the Eker rats exposed to the heavy-ions, C (290 MeV/u) and Fe (500 MeV/u) ions, comparing to the effects of X-ray irradiation. Pregnant rats were exposed to C and Fe ions and X-rays with a single dose of 1 Gy, 2 Gy, 3 Gy on day 19 of gestation. The offspring were sacrificed at 8 weeks of age. We evaluated organ weights and tumor genesis. The weights of thymus, lung, liver, spleen were found to be no difference from the control at 1 Gy irradiation but 50% decrease at 3 Gy irradiation. We found in the irradiated animal that kidney, brain and testis were very sensitive organs of which the weight decreased to approximately 80% at 1 Gy and to 40% at 3 Gy irradiations. Based on the dose-response relationship of the radiation-induced carcinoma, averaged RBE ware calculated to be 1.1 for C-ion, 1.6 for Fe-ion.  相似文献

I investigated the responses of gibbons (Hylobates muelleri) to male songs played back from different locations but similar distances from the subjects. The gibbons showed spatial variations in response to songs. Mated males led silent group approaches toward songs emanating from the center of the range. Songs delivered from positions along the range periphery elicited either group approaches or duets. Songs played back from within the ranges of neighbors rarely resulted in duetting activity. Location, a contextual factor, is important in determination of the response evoked by song. These results are consistent with the hypothesis that male song plays a role in mediating intergroup spacing among gibbons.  相似文献

It was generally believed that autosomal CpG islands (CGIs) escape methylation. However, our comprehensive analysis of allelic methylation status of 149 CGIs on human chromosome 21q revealed that a sizable fraction of them are methylated on both alleles even in normal blood cells. Here, we performed a similar analysis of 656 CGIs on chromosome 11q, which is gene-rich in contrast with 21q. The results indicate that 11q contains less methylated CGIs, especially those with tandem repeats and those in the coding or 3'-untranslated regions (UTRs), than 21q. Thus, methylation status of CGIs may substantially differ from one chromosome to another.  相似文献

TGF-beta1 is a well-known immunosuppressive cytokine that inhibits inducible nitric oxide synthase (iNOS) gene expression in various cells including macrophages. In this study, we investigated the suppressive mechanisms of TGF-beta1 on IFN-gamma-induced iNOS gene expression using the murine macrophage-like cell line RAW 264.7. TGF-beta1 decreased iNOS protein amount through enhanced degradation, although TGF-beta1 did not affect IFN-gamma-induced iNOS mRNA level or stability. In addition, the enhancement of iNOS protein degradation by TGF-beta1 treatment was almost completely blocked by MG132, a proteasome inhibitor. Furthermore, TGF-beta1 enhanced the trypsin-like activity of proteasomes in the presence of IFN-gamma, although did not enhance the peptidylglutamyl-peptide hydrolyzing and chymotrypsin-like activities of proteasomes. The level of ubiquitinated iNOS protein was not significantly altered by IFN-gamma or IFN-gamma plus TGF-beta1 treatment. Because MG132 inhibited iNOS protein degradation and IFN-gamma plus TGF-beta1 treatment increased the trypsin-like activity of proteasomes, we hypothesized that TGF-beta1 might enhance iNOS protein degradation via the ubiquitin-proteasome pathway in the presence of IFN-gamma. We propose that these mechanisms of TGF-beta1 in the posttranslational regulation of iNOS gene expression may contribute to suppression of excess nitric oxide during inflammatory processes.  相似文献

We previously reported l ‐α‐aminooxy‐phenylpropionic acid (AOPP) to be an inhibitor of auxin biosynthesis, but its precise molecular target was not identified. In this study we found that AOPP targets TRYPTOPHAN AMINOTRANSFERASE of ARABIDOPSIS 1 (TAA1). We then synthesized 14 novel compounds derived from AOPP to study the structure–activity relationships of TAA1 inhibitors in vitro. The aminooxy and carboxy groups of the compounds were essential for inhibition of TAA1 in vitro. Docking simulation analysis revealed that the inhibitory activity of the compounds was correlated with their binding energy with TAA1. These active compounds reduced the endogenous indole‐3‐acetic acid (IAA) content upon application to Arabidopsis seedlings. Among the compounds, we selected 2‐(aminooxy)‐3‐(naphthalen‐2‐yl)propanoic acid (KOK1169/AONP) and analyzed its activities in vitro and in vivo. Arabidopsis seedlings treated with KOK1169 showed typical auxin‐deficient phenotypes, which were reversed by exogenous IAA. In vitro and in vivo experiments indicated that KOK1169 is more specific for TAA1 than other enzymes, such as phenylalanine ammonia‐lyase. We further tested 41 novel compounds with aminooxy and carboxy groups to which we added protection groups to increase their calculated hydrophobicity. Most of these compounds decreased the endogenous auxin level to a greater degree than the original compounds, and resulted in a maximum reduction of about 90% in the endogenous IAA level in Arabidopsis seedlings. We conclude that the newly developed compounds constitute a class of inhibitors of TAA1. We designated them ‘pyruvamine’.  相似文献

A novel pseudo cyclic penta‐β‐peptide composed of a β‐naphthylalanine, two β‐alanines, and a sequence of ethylenediamine‐succinic acid (CP5ES) is synthesized and investigated on peptide nanotube (PNT) formation. When the PNT is formed with the maximum number of intermolecular hydrogen bonds between the cyclic peptides, the sequence enables the alignment of the side chains, naphthyl groups, on one side of the PNT. Microscopic and spectroscopic observations of CP5ES crystals reveal that CP5ES forms rod‐ or needle‐shaped molecular assemblies showing exciton coupling of the Cotton effect and predominant monomer emission, which are different from a reference cyclic tri‐β‐peptide composed of a β‐naphthylalanine and two β‐alanines. Insertion of a sequence of ethylenediamine‐succinic acid into β‐amino acids in the cyclic skeleton is therefore suggested to be effective to make the side chains aligning on one side of the PNT. Wild chimpanzees form temporary parties that vary in size and composition. Previous studies have revealed considerable intraspecific variation in party compositions. We examined patterns of association among age, sex, and reproductive classes of chimpanzees at Ngogo in the Kibale National Park, Uganda. We employed a class-based association index and a randomization procedure to control for confounding factors and to test for differences between classes. Results indicate that males associated with other males significantly more than expected if all classes behaved equivalently, while females generally associated with individuals of the same sex less than expected. To interpret these patterns we used two additional indices that separate associations into two components: general gregariousness and preference for particular classes of associates. Males and estrous females were more gregarious than other classes, while anestrous females were less so. After controlling for general gregariousness, adult males as a class showed no specific preference for associating with each other. Anestrous females preferred each other as party members, and estrous females avoided each other. These results are consistent with previous findings that adult males are more gregarious than females. They diverge from the standard picture of chimpanzee society, however, by suggesting a mutual affinity among anestrous females, but not among adult males as a class.  相似文献

Fruit syndrome found in zoochorous plants is regarded as a result of hypothetical coevolution with a seed disperser/predator. Fruiting phenology was compared among two representative syndromes, such as bird–monkey syndrome (BM) and ruminant–rodent–elephant syndrome (RRE), plus the gravity dispersal species for comparison, in south-western Cameroon in order to examine which biotic or abiotic factors educed syndrome variation. The individual size of selected plants (> 1.6 m in height) was recorded in a 16.95-ha area for recruitment estimation and their fruiting behaviour was checked for 9 months from June 1985 to February 1986. The BM species, a suggested successful group, fruited in a less synchronous manner within species and had fruiting peaks just before and during the rainy season. The RRE species, a suggested failing group, showed greater variation within syndrome in fruiting timing, duration and synchronization. Results obtained supported no clear phenological response to corresponding vertebrates except for the case of Sacoglottis gabonensis in the RRE. The reason for concealing potential responses is probably a result of conflicting requirements for seed dispersal and offspring survival. Periodical fruiting of the BM is likely to reflect ecological constraint, such as water stress on seedlings, caused from syndrome-specific morphological limitation. In this case, the animals have indirect effects on fruiting phenology through selecting syndrome-specific fruit morphology. The greater variation of the RRE suggests a broad spectrum of dispersal tactics from faithful zoochory to alternatives, with a trade off between agent restriction and seed size and another trade off between parental investment and seed-predation risk.  相似文献
