Chemical conjugation of poly(styrene-co-maleic acid) to an antitumor protein (neocarzinostatin) yielded an entirely new derivative designated as smancs (polystyrene-maleic acid conjugated neocarzinostatin). The purpose of the modification was to improve its pharmacological properties; the resulting conjugate exhibited much higher tumoritropism and lymphotropism, enhancedin vivo stability (about ten times), higher chemotherapeutic index (lower toxicity), and decreased antigenicity. Another advantage associated with this molecular engineering was an increased hydrophobicity. By this character it was solubilized in lipid contrast medium LipiodolR, which facilitated highly sensitive detection under x-ray-accompanying selective anticancer effectin situ. Three factors were responsible for such improvements: molecular size, hydrophobicity, and polyanionic nature. Most of the known drugs so far used as therapeutic agents are less than 1000 daltons, and those with larger molecular weights have been explored much less extensively. By polymer conjugation, the size of the drug can be extended to about 15,000 daltons (macromolecular therapeutics). The most outstanding characteristic of smancs (16,000 daltons) was the tumoritropicity, which may be a result of the highly developed neovasculature of solid tumors. Smancs as a prototype drug thus appears to lead the way in cancer drug targeting.  相似文献

The guidepost neurons for the lateral olfactory tract, which are called lot cells, are the earliest‐generated neurons in the neocortex. They migrate tangentially and ventrally further down this tract, and provide scaffolding for the olfactory bulb axons projecting into this pathway. The molecular profiles of the lot cells are largely uncharacterized. We found that lot cells specifically express metabotropic glutamate receptor subtype‐1 at a very early stage of development. This receptor is functionally competent and responds to a metabotropic glutamate receptor agonist with a transient increase in the intracellular calcium ion concentration. When the glutamatergic olfactory bulb axons were electrically stimulated, lot cells responded to the stimulation with a calcium increase mainly via ionotropic glutamate receptors, suggesting potential neurotransmission between the axons and lot cells during early development. Together with the finding that lot cells themselves are glutamatergic excitatory neurons, our results provide another notable example of precocious interactions between the projecting axons and their intermediate targets. © 2012 Wiley Periodicals, Inc. HeLa/MV cells which have previously been found to contain measles antigen in more than 90% of cells were very similar to normal HeLa cells in their morphology and growth. Although almost all of HeLa/MV cells may be infected, only 10% of them released infectious virus particles. The hemadsorption test, however, showed that most of the infected cells produced by hemagglutinin. Two kinds of clones were obtained by cloning HeLa/MV cells in the presence of anti-measles serum. One was the virus-releasing clone and the other the uninfected clone which did not contain any measles antigen. The proportions of virus-releasing clones to all clones varied between 20 to 54%, and did not show any increase even after recloning of the virus-releasing clones. The susceptibility of the uninfected clones to the standard measles virus was not different from that of normal HeLa cells.  相似文献

We formulated a species distribution model (SDM) for the amphidromous fish Sicyopterus japonicus on the basis of presence/absence data and confirmed its performance at various population densities. The best-fit SDM selected several environmental factors, including water depth and velocity, and performed validated prediction of presence/absence of various densities. The predicted probability of occurrence was positively correlated with the observed density. The density was positively related to habitat occupancy, suggesting that this species occupied a wide range of habitats under high densities, but only optimal habitat under low densities. Therefore, the threshold value for predicting presence/absence increased with decreasing densities.  相似文献

We had previously cloned and characterized a gene for a novel asparagine-rich protein from P. falciparum (PfARP), a target of natural human immune response. The antibodies to PfARP were localized to the surface of parasitized red blood cells and reacted with intracellular components in all erythrocytic asexual and sexual stages of the parasite. We here describe reactivity of antibodies against this novel PfARP on the surface of mosquito stage sporozoite of P. falciparum by indirect immunofluorescence assay and immunoelectron microscopy, the latter revealing a highly periodic punctate pattern of distribution of PfARP on the surface of sporozoite. These results suggest a possibility that PfARP might represent yet another sporozoite surface protein.  相似文献

Summary Cross-linking chemical modification of pyrroloquinoline quinone (PQQ) glucose dehydrogenase (GDH) by glutaraldehyde was carried out and its stability was analyzed. Although native PQQGDH was inactivated within 30 min at a higher temperature than 50 °C, cross-linked PQQGDH retained more than 40% of initial activity even after 30 min of incubation at 54 °C. In addition to the increase in thermal stability, cross-linked PQQGDH gained high EDTA tolerance. The stabilization may be achieved by increased the rigidity of PQQGDH holo enzyme conformation.  相似文献
