Treatment of the chromosomes of 25 rodent species with a 50 degrees C hypotonic solution and Giemsa staining permitted identification of the heterochromatic X chromosome in 24 species. With this technique, the facultative of the heterochromatic X chromosome or the facultative portion of large, composite-type X chromosoms is stained darker than the other chromosomes, allowing it to be distinguished from the homologous euchromatic X chromosome in female metaphase cells. Intense staining of the single X chromosome was not observed in male metaphase cells. It is suggested that this differential staining of one of the two X chromosomes might be due to qualitative differences in chromosomal proteins rather than to differences in the degree of chromosomal condensation or in DNA base sequence. 相似文献

A practical calculation procedure to correct the underestimatecaused by isotope dilution in 15N uptake experiments is described.The estimate is based on the experimental observation of 15Nincorporation into particuUtc organic matter but not on directmeasurement of isotope enrichment in the substrate. Conventionaldata sets of 15N uptake and an estimate of the ratio of theflux of regeneration and uptake can provide the informationneeded to correct for isotope dilution. Lipid peroxide formation and plasma membrane damage in mouse liver following the administration of Salmonella endotoxin were examined. The liver lipoperoxide level was markedly elevated in animals given endotoxin compared with that in the controls, and returned to its normal range after 2 days. On the other hand, superoxide dismutase activity was decreased by 18–48 hr after endotoxin injection, thereafter tending to increase. Glutathione reductase and glutathione peroxidase activities declined in the liver 18 hr after the injection. The endotoxin resulted in much lower lipoperoxide formation in the livers of tolerant mice than in those of the poisoned mice. The lipoperoxide level in endotoxin-poisoned mice after the administration of α-tocopherol was lower than that in the controls, and α-tocopherol administration prevented completely the membrane protein damage that arose from endotoxin challenge. After glutathione administration the membranes of the poisoned mice also returned to almost the normal disk electrophoretic profile. These results suggest that lipid peroxide formation in the liver plasma membrane caused by free radicals might occur in a tissue ischemic state in endotoxicosis. 相似文献

We have established 13 esophageal cancer cell lines capable of growing in a protein-free environment. The growth of these cells was not affected by conditioned medium, but the growth of NIH3T3 cells and human fibroblasts was stimulated by conditioned medium. On the other hand, conditioned medium inhibited the growth of human endothelial cells. Amplified int-2 oncogene correlated well with the growth of cells in a protein-free environment but the number of EGF receptors and growth effect of EGF did not relate to such growth. Esophageal cancer cells grow automatically, possibly involving mesenchymal cells via the paracrine system. This results in a poor prognosis in patients. 相似文献

The reproductive ecology of the gobiid fish Bathygobius fuscus was studied at Nobeoka, Miyazaki, Japan. Males of this species maintain small rock holes as a nest and females spawn an egg
mass on the wall of the nest. The males employed two forms of mating tactic: nest holding and sneaking. A nest holder stayed
in the nest and waited for a female to visit, whereas a sneaker intruded into a nest while a pair was engaged in reproduction.
Males larger than 55 mm standard length were always nest holders; those of smaller size employed both tactics. As the larger
males excluded the smaller males, the latter did not occupy a nest hole. With a decrease in the number of larger males, smaller
males changed their mating tactic from sneaking to nest holding. The results suggest that male Bathygobius fuscus adopt a conditional strategy whereby they change their tactic depending on their social status.

Inhibition of normal human natural killer cell activity by human immunodeficiency virus synthetic transmembrane peptides 总被引:7,自引:0,他引:7
The inhibitory effect on normal natural killer (NK) cell activity of two synthetic peptides corresponding to amino acid sequences 735-752 and 846-860, respectively, as deduced from the amino acid sequences of HTLV-IIIB gp160, was assessed. Sequences 735-752 and 846-860 correspond to regions located within the HIV transmembrane gp41, the carboxy terminus of HIV gp160. These two synthetic peptides have been shown previously to suppress the mitogen- and alloantigen-induced normal human lymphocyte blastogenic responses. Peptides 735-752 and 846-860 conjugated to protein carriers exerted a significant inhibition on the normal NK cell activity assayed against K562 tumor target cells in an in vitro 51Cr-release cytoltoxicity assay. At variance, control peptides similarly conjugated had no effect on NK activity. Addition of exogenous recombinant human interleukin-2 (IL-2) resulted in a partial restoration of the suppression of NK cell activity exerted by both peptides. Binding experiments indicated that peptides 735-752 and 846-860 did not affect the formation of effector cell-target cell conjugates, suggesting inhibitory effect(s) subsequent to the formation of the lytic complex as one potential mechanism of the observed NK suppression. These results suggest that peptides 735-752 and 846-860 homologous to sequences within the HIV transmembrane gp41 may play an important role in the pathogenesis of the defective NK cell activity observed in patients with acquired immunodeficiency syndrome (AIDS). 相似文献

Twenty mutants of Bacillus brevis which were deficient in gramicidin S formation were isolated by N-methyl-N′-nitrosoguanidine treatment. In addition to three groups which have been previously classified, further two groups were established according to their characteristics of amino acid activating enzymes concerned with gramicidin S formation. The fourth group mutants had a phenylalanine activating enzyme, but they had an enzyme complex from which one specific enzyme among proline, valine and leucine activating enzymes was deleted. Some of them also the ability to form d-phenylalanyl-l-prolyl diketpiperazine (DKP) even though they had phenylalanine and proline activating enzymes. The fifth group mutants contained both a phenylalanine activating enzyme and a complex of prodine, valine, ornithine and leucine activating enzymes like as a wild strain, but did not synthesize gramicidin S, and also one of them could not form even DKP.Combination of enzymes from DKP (+) mutants of the fourth or fifth groups with the first group mutant which had an intact proline, valine, ornithine and leucine activating enzyme complex showed gramicidin S formation, but the combination of enzymes from DKP (−) mutants except a proline activating enzyme minus mutant with the first group mutant could not synthesize gramicidin S. 相似文献

We have demonstrated that gramicidin S synthetase 1 (GS 1), phenylalanine racemase [EC 5.1.1.11], of Bacillus brevis catalyzes the exchange between a proton in the medium and alpha-hydrogen of phenylalanine in the course of the racemase reaction by using tritiated water or L-phenyl[2,3-3H]alanine. GS 1 from some gramicidin S non-producing mutants of B. brevis lacking phenylalanine racemase activity did not catalyze the tritium exchange reaction. The proton exchange between phenylalanine bound as thioester on the GS 1-phenylalanine complex and water in the medium was detected, but 5,5'-dithiobis(2-nitrobenzoic acid)-modified complex lacked both the proton exchange and phenylalanine racemase activity. It is suggested that a base group, probably a sulfhydryl group, on the enzyme functions as proton donor and acceptor during the phenylalanine racemase reaction. 相似文献
