Some properties of the circadian rhythm in potassium uptakeof flow medium culture of the long-day duckweed Lemna gibbaG3 were examined.
  1. In total darkness, the rhythm faded out in ca. 48 hr; it restartedon transfer to continuous light. Under low-intensity light (below700 lux), the rhythm was damped rapidly
  2. The rhythm appearedregardless of the potassium concentrationin the culture medium(from 10/m to 2 HIM). The amplitude, butnot the period, ofthe rhythm was influenced by the ambientpotassium concentration.

A genetic transformation procedure for Cryptomeria japonica was developed after co-cultivation of embryogenic tissues with the disarmed Agrobacterium tumefaciens strain C58/pMP90, which harbours the visual reporter gene sgfp and two selectable marker genes, hpt and nptII. We were able to generate eight and three independent transgenic lines per gram of embryogenic tissue after selection on hygromycin and kanamycin medium, respectively. Transgenic plants were regenerated through somatic embryogenesis in 4 lines out of these 11 lines. Green fluorescent protein fluorescence was observed under fluorescent microscopy. Integration of the genes into the genome was confirmed by polymerase chain reaction analysis of embryogenic tissues and Southern blot analysis of regenerated plantlets.  相似文献

Recent studies reveal that the intracellular localization of pyruvate,Pi dikinase (PPDK, EC 2.7.9.1) in mesophyll cells of malic enzyme (ME)-dependent Crassulacean acid metabolism (CAM) plants varies among species, occurring not only in the chloroplasts but also in the cytosol in some species. The facultative CAM plant Kalanchoë blossfeldiana accumulates PPDK in both compartments of the mesophyll cells. In this study, the patterns of accumulation of the chloroplastic and cytosolic PPDKs were investigated for K. blossfeldiana plants with different CAM activities by immunogold labeling and electron microscopy. Greater CAM activity was found in plants grown under drought conditions with short days than under well-watered conditions with long days, and in lower leaves than in higher leaves. There was a trend that plants and leaves with greater CAM activity show denser labeling for PPDK in both the cytosol and chloroplasts. However, the ratio of the density of PPDK labeling in the cytosol to that in the chloroplasts was almost constant (2.4–3.0). Higher labeling for phosphoenolpyruvate carboxylase (EC 4.1.1.31) in the cytosol was also correlated with higher CAM activity but there was almost no difference in the density of labeling for ribulose-1,5-bisphosphate carboxylase/oxygenase (EC 4.1.1.39) in the chloroplasts. These results indicate that the increase in accumulation of cytosolic PPDK is closely associated with the increase of chloroplastic PPDK during enhanced CAM expression. This suggests that both PPDKs are involved in CAM function.  相似文献

In human cells, hMLH1, hMLH3, hPMS1 and hPMS2 are four recognised and distinctive homologues of MutL, an essential component of the bacterial DNA mismatch repair (MMR) system. The hMLH1 protein forms three different heterodimers with one of the other MutL homologues. As a first step towards functional analysis of these molecules, we determined the interacting domains of each heterodimer and tried to understand their common features. Using a yeast two-hybrid assay, we show that these MutL homologues can form heterodimers by interacting with the same amino acid residues of hMLH1, residues 492–742. In contrast, three hMLH1 partners, hMLH3, hPMS1 and hPMS2 contain the 36 homologous amino acid residues that interact strongly with hMLH1. Contrary to the previous studies, these homologous residues reside at the N-terminal regions of three subdomains conserved in MutL homologues in many species. Interestingly, these residues in hPMS2 and hMLH3 may form coiled-coil structures as predicted by the MULTICOIL program. Furthermore, we show that there is competition for the interacting domain in hMLH1 among the three other MutL homologues. Therefore, the quantitative balance of these three MutL heterodimers may be important in their functions.  相似文献

Protein kinase C (PKC) plays a prominent role in immune signaling. To elucidate the signal transduction in a respiratory burst and isoform-specific function of PKC during FcgammaR-mediated phagocytosis, we used live, digital fluorescence imaging of mouse microglial cells expressing GFP-tagged molecules. betaI PKC, epsilonPKC, and diacylglycerol kinase (DGK) beta dynamically and transiently accumulated around IgG-opsonized beads (BIgG). Moreover, the accumulation of p47(phox), an essential cytosolic component of NADPH oxidase and a substrate for betaI PKC, at the phagosomal cup/phagosome was apparent during BIgG ingestion. Superoxide (O(2)(-)) production was profoundly inhibited by G?6976, a cPKC inhibitor, and dramatically increased by the DGK inhibitor, R59949. Ultrastructural analysis revealed that BIgG induced O(2)(-) production at the phagosome but not at the intracellular granules. We conclude that activation/accumulation of betaI PKC is involved in O(2)(-) production, and that O(2)(-) production is primarily initiated at the phagosomal cup/phagosome. This study also suggests that DGKbeta plays a prominent role in regulation of O(2)(-) production during FcgammaR-mediated phagocytosis.  相似文献
