 Degradation of tetrachloroguaiacol is catalyzed by an extracellular enzyme, the laccase of the white-rot fungus Coriolus versicolor. This enzyme catalyzes the dechlorination of tetrachloroguaiacol and release of chloride ions. The pathway for the degradation was deduced from the intermediates produced by purified laccase and 18O-labeling experiments. The first step is demethylation. The resulting tetrachlorocatechol is dechlorinated to give 2,3,5-trichloro-6-hydroxy-p-benzoquinone, 2,5-dichloro-3,6-dihydroxy-p-benzoquinone, and dichloro-6-hydroxy-p-benzoquinone. Isotopic experiments established the mechanism of dechlorination of tetrachloroguaiacol by laccase. The laccase-catalyzed dechlorination is not caused by oxidative coupling but by nucleophilic substitution in which Cl- is released by water from cation radicals generated by laccase. An engineering tool for controlling flux distribution on metabolic pathways to an appropriate state is highly desirable in bioproduction. An optogenetic switch, which regulates gene expression by light illumination is an attractive on/off switchable system, and is a promising way for flux control with an external stimulus. We demonstrated a light-inducible flux control between glycolysis and the methylglyoxal (MGO) pathway in Escherichia coli using a CcaS/CcaR system. CcaR is phosphorylated by green light and is dephosphorylated by red light. Phosphorylated CcaR induces gene expression under the cpcG2 promoter. The tpiA gene was expressed under the cpcG2 promoter in a genomic tpiA deletion strain. The strain was then cultured with glucose minimum medium under green or red light. We found that tpiA messenger RNA level under green light was four times higher than that under red light. The repression of tpiA expression led to a decrease in glycolytic flux, resulting in slower growth under red light (0.25 hr −1) when compared to green light (0.37 hr −1). The maximum extracellular MGO concentration under red light (0.2 mM) was higher than that under green light (0.05 mM). These phenotypes confirm that the MGO pathway flux was enhanced under red light.  相似文献

Specialized trophic interactions in plant–herbivore–parasitoid food webs can spur “bottom–up” diversification if speciation in plants leads to host‐shift driven divergence in insect herbivores, and if the effect then cascades up to the third trophic level. Conversely, parasitoids that search for victims on certain plant taxa may trigger “top–down” diversification by pushing herbivores into “enemy‐free space” on novel hosts. We used phylogenetic regression methods to compare the relative importance of ecology versus phylogeny on associations between Heterarthrinae leafmining sawflies and their parasitoids. We found that: (1) the origin of leafmining led to escape from most parasitoids attacking external‐feeding sawflies; (2) the current enemies mainly consist of generalists that are shared with other leafmining taxa, and of more specialized lineages that may have diversified by shifting among heterarthrines; and (3) parasitoid–leafminer associations are influenced more by the phylogeny of the miners’ host plants than by relationships among miner species. Our results suggest that vertical diversifying forces have a significant—but not ubiquitous—role in speciation: many of the parasitoids have remained polyphagous despite niche diversification in the miners, and heterarthrine host shifts also seem to be strongly affected by host availability.  相似文献

Introduced predators can have harmful top-down effects on their newly colonized system through competition with and direct predation on native species. Following an initial introduction of muskellunge in Lac Frontière, Québec in the 1970s at the headwaters of the Wolastoq/Saint John River, the species rapidly migrated downstream, expanding its range by ~500 km over ~20 years. Despite this expansive colonization and concern over possible threats to native species, little is known about the basic ecology of muskellunge in this system. The last downstream barrier is the hydroelectric facility, Mactaquac Generating Station (MGS), 150 km upstream of the sea. While there are no downstream fish passage facilities at MGS, adult muskellunge have been recorded downstream. In this study, muskellunge (n = 23) were surgically tagged with very-high-frequency (VHF) radio or combined acoustic radio telemetry (CART) tags and tracked over two spawning seasons. We sought to determine if there was a reproducing population downstream of MGS and tracked Tagged muskellunge over two spawning seasons. We tracked fish to locate and confirm spawning sites, and followed up with egg and/or juvenile sampling surveys. Tagged muskellunge (90%) moved upstream towards the MGS during the spawning period in each year (2016 and 2017), where they remained throughout the entire spawning period. No spawning or nursery sites were confirmed near MGS, but in 2016 three distinct spawning locations and six distinct nursery sites were confirmed 10–12 km downstream amongst a chain of flooded islands. In 2016, eggs, sac-fry and juveniles were collected and confirmed as muskellunge by genetic sequencing, providing the first empirical observation of successful spawning downstream of MGS.  相似文献

Food limitation has been suggested as one of the most important factors affecting life history evolution in terrestrial vertebrates. However, this inference is based mainly on evidence from birds, and reproductive trade-offs may differ among groups with different forms of parental care. To study whether the costs of enlarged litters (decreased mass of offspring) would appear when food is not limiting, we performed outdoor enclosure experiments in which we manipulated simultaneously the litter size (control versus litter plus two pups) and food availability (control versus food-supplemented) of female bank voles, Clethrionomys glareolus. The weaning success of females increased significantly in response to supplementary food. When females were provided with extra food, no differences were observed in body masses of weanlings of control and enlarged litters. Further, food-supplemented females grew to larger sizes during nursing than unsupplemented females. Our experiment suggests that energetic requirements during nursing constrain the number of offspring that can be raised successfully in a particular breeding attempt. The results also indicate that unlimited food resources may increase future reproductive potential of females because they can use more energy for somatic growth.  相似文献

Dilute formaldehyde was the most suitable treatment to inhibit sediment bacteria, since bacterial activity remained low during long-term incubations and the chemical changes in the sediment were minimal. The inhibiting effects of HgCl2, autoclaving, and gamma radiation were diminished during longer incubations; these treatments also caused increases in dissolved nutrients.  相似文献

Biochemical reactions powered by ATP hydrolysis are fundamental for the movement of molecules and cellular structures. One such reaction is the encapsidation of the double-stranded DNA (dsDNA) genome of an icosahedrally symmetric virus into a preformed procapsid with the help of a genome-translocating NTPase. Such NTPases have been characterized in detail from both RNA and tailed DNA viruses. We present four crystal structures and the biochemical activity of a thermophilic NTPase, B204, from the nontailed, membrane-containing, hyperthermoacidophilic archaeal dsDNA virus Sulfolobus turreted icosahedral virus 2. These are the first structures of a genome-packaging NTPase from a nontailed, dsDNA virus with an archaeal host. The four structures highlight the catalytic cycle of B204, pinpointing the molecular movement between substrate-bound (open) and empty (closed) active sites. The protein is shown to bind both single-stranded and double-stranded nucleic acids and to have an optimum activity at 80°C and pH 4.5. The overall fold of B204 places it in the FtsK-HerA superfamily of P-loop ATPases, whose cellular and viral members have been suggested to share a DNA-translocating mechanism.  相似文献
