Potential porcine circovirus type 2 (PCV2) capsid protein epitopes, suitable for expression on the surface of cucumber mosaic virus (CMV) particles were determined by a thorough analysis of the predicted PCV capsid protein structure. The ab initio protein structure prediction was carried out with fold recognition and threading methods. The putative PCV epitopes were selected on the basis of PCV virion models and integrated into the plant virus coat protein, after amino acid position 131. The recombinants were tested for infectivity and stability on different Nicotiana species and stable recombinant virus particles were purified. The particles were tested for their ability to bind to PCV induced porcine antibodies and used for specific antibody induction in mice and pigs. The results showed that PCV epitopes expressed on the CMV surface were recognized by the porcine antibodies and they were also able to induce PCV specific antibody response. Challenge experiment with PCV2 carried out in immunized pigs showed partial protection against the infection. Based on these results it was concluded that specific antiviral vaccine production for the given pathogen was feasible, offering an inexpensive way for the mass production of such vaccines. 相似文献

Seed shape has traditionally been used as a tribal character in the capsular-fruited Melastomataceae. The seed morphology of the five neotropical tribes—Microlicieae, Tibouchineae, Rhexieae, Merianieae, and Bertolonieae—was studied with scanning electron and light microscopes. On the basis of seed morphology, five seed types are recognized: microlicioid, tibouchinoid, rhexioid, merianioid, and bertolonioid. Each of these types is illustrated and discussed, with some observations on the range of variation found. The genera of these five neotropical tribes were surveyed for seed morphology, and the results are presented here, together with a discussion of their systematic significance. On the whole, seed morphology confirms the existence of five tribes among the neotropical capsular-fruited Melastomataceae and provides interesting evidence for generic relationships; however, it also calls into doubt the taxonomic disposition of certain genera and the delimitation of these five tribes. The seeds of most genera fit into one of these five basic types, but there are some which do not; these exceptions are discussed in terms of their seed morphology and possible relationships. 相似文献

Foraminifera have one of the best known fossil records among the
unicellular eukaryotes. However, the origin and phylogenetic relationships
of the extant foraminiferal lineages are poorly understood. To test the
current paleontological hypotheses on evolution of foraminifera, we
sequenced about 1,000 base pairs from the 3' end of the small subunit rRNA
gene (SSU rDNA) in 22 species representing all major taxonomic groups.
Phylogenies were derived using neighbor- joining, maximum-parsimony, and
maximum-likelihood methods. All analyses confirm the monophyletic origin of
foraminifera. Evolutionary relationships within foraminifera inferred from
rDNA sequences, however, depend on the method of tree building and on the
choice of analyzed sites. In particular, the position of planktonic
foraminifera shows important variations. We have shown that these changes
result from the extremely high rate of rDNA evolution in this group. By
comparing the number of substitutions with the divergence times inferred
from the fossil record, we have estimated that the rate of rDNA evolution
in planktonic foraminifera is 50 to 100 times faster than in some benthic
foraminifera. The use of the maximum-likelihood method and limitation of
analyzed sites to the most conserved parts of the SSU rRNA molecule render
molecular and paleontological data generally congruent.

A survey of pollen morphology in 20 species representing the 11 genera of the North American subtribe Stephanomeriinae by light, scanning electron, and transmission electron microscopy revealed 10 of the 11 genera to have echinate, tricolporate pollen grains, Lygodesmia being the only genus with echinolophate pollen. Sectioned exines of most of the species examined are similar, being composed of ektexine and endexine. The ektexine surface is composed of spines which typically have globose perforate bases. A cavus occurs as a separation between the basis (foot layer) and the columellae in all of the genera examined except Chaetadelpha. Pollen of the two species of Glyptopleura were found to be strikingly different in exomorphology. Pollen of the putatively self-fertile G. marginata has much shorter spines than the closely related G. setulosa. Atrichoseris, Anisocoma, Calycoseris, Glyptopleura, Pinaropappus, Prenanthella, and most species of Malacothrix have pollen which lack paraporal ridges. The remaining genera, Chaetadelpha, Lygodesmia, Rafinesquia, and Stephanomeria have well-developed ridges of fused spine bases around the apertures. Pollen characters, particularly those of the aperture region, have been found to be systematically useful in the subtribe, therefore acetolyzed material gives more useful information than untreated pollen. 相似文献

The past several years have seen a flurry of papers seeking to clarify the utility and limits of DNA barcoding, particularly in areas such as species discovery and paralogy due to nuclear pseudogenes. Platelet activation is tightly regulated by products of the endothelium and platelets including nitric oxide (NO). Excess vascular oxidative stress has been associated with impaired NO release, and antioxidant status has been shown to alter endothelium-derived NO bioactivity. Although physiological levels of a-tocopherol are known to inhibit platelet function, the effect of a-tocopherol on platelet NO release is unknown. Loading platelets with physiologic levels of a-tocopherol increased platelet NO production approximately 1.5-fold (Pa-tocopherol, platelet NO release increased 50% (Pa-Tocopherol-loaded platelets also produced 74% less superoxide as compared with control (Pa-tocopherol inhibited PKC-dependent eNOS phosphorylation as determined by immunoprecipitation. Lastly, platelets isolated from NOS3-deficient mice released 80% less superoxide as compared with control animals (P=0.011), and incubation of NOS III-deficient platelets with 500 mM a-tocopherol only caused a modest additional decrease in platelet superoxide release (NS). Thus, a-tocopherol appears to enhance platelet NO release both in vitro and in vivo through antioxidant- and PKC-dependent mechanisms. 相似文献
