Bacterial proteases are considered virulence factors and it is presumed that by abrogating their activity, host endogenous protease inhibitors play a role in host defense against invading pathogens. Here we present data showing that Staphylococcus aureus cysteine proteases (staphopains) are efficiently inhibited by Squamous Cell Carcinoma Antigen 1 (SCCA1), an epithelial-derived serpin. The high association rate constant (k(ass)) for inhibitory complex formation (1.9×10(4) m/s and 5.8×10(4) m/s for staphopain A and staphopain B interaction with SCCA1, respectively), strongly suggests that SCCA1 can regulate staphopain activity in vivo at epithelial surfaces infected/colonized by S. aureus. The mechanism of staphopain inhibition by SCCA1 is apparently the same for serpin interaction with target serine proteases whereby the formation of a covalent complex result in cleavage of the inhibitory reactive site peptide bond and associated release of the C-terminal serpin fragment. Interestingly, the SCCA1 reactive site closely resembles a motif in the reactive site loop of native S. aureus-derived inhibitors of the staphopains (staphostatins). Given that S. aureus is a major pathogen of epithelial surfaces, we suggest that SCCA1 functions to temper the virulence of this bacterium by inhibiting the staphopains. 相似文献

Background: Physiotherapy of edema in cases with obstructed main lymphatics of lower limbs requires knowledge of how high external pressures should be applied manually or set in compression devices in order to generate tissue pressures high enough to move tissue fluid to nonswollen regions and to measure its flow rate. Methods: We measured tissue fluid pressure and flow in subcutaneous tissue of lymphedematous limbs stages II to IV at rest and during pneumatic compression under various pressures and inflation timing. An 8-chamber sequential compression device inflated to pressures 50-120?mmHg, for 50 sec each chamber, with no distal deflation, was used. Pressures were measured using a wick-in-needle and electronic manometer. Fluid flow was calculated from continuously recorded changes in limb circumference using strain gauge plethysmography. Results: Before massage, in all stages of lymphedema, stagnant tissue fluid pressures in subcutaneous tissue ranged between -1 and +10 mmHg and did not differ from those measured in normal subjects. Pressures generated in tissue fluid by pneumatic compression reached 40-100 mmHg and were lower than those in inflated chambers. High pressure gradient through the skin was caused by its rigidity (fibrosis) and dissipation of applied compression force to proximal noncompressed limb regions. The calculated volumes of displaced tissue fluid ranged from 10 to 30 ml per compression cycle, to reach in some cases 100 ml in the groin region. Conclusions: Tissue fluid pressures generated by a pneumatic device were found lower than in the compression chambers. The obtained results point to the necessity of applying high pressures and longer compression times to generate effective tissue fluid pressures and to provide enough time for moving the stagnant fluid. 相似文献

Excessive consumption of highly palatable food has been linked to the development of eating disorders and obesity, and can
be modeled in non-food-deprived rats by offering them a limited (2-h daily) access to an optional dietary fat. Since the glutamatergic
system has recently emerged as a viable target for binge-eating medication development, we compared the effects of subchronic
treatment with glutamatergic receptor antagonists to the effects of a reference appetite-suppressing agent sibutramine on
highly palatable food (lard) and normal chow intake. In three separate experiments, the consumption of a standard laboratory
chow and lard were measured during 12 days of medication treatment and for 6 days afterwards. Generalized estimating equations
analysis demonstrated that sibutramine (7.5 mg/kg, PO) significantly decreased lard consumption, with a concurrent increase
in chow consumption. Sibutramine effects disappeared after treatment discontinuation. The NMDA receptor antagonist memantine
(5 mg/kg, IP) significantly decreased lard consumption and increased chow consumption, comparable to effects of sibutramine;
however, memantine’s effects persisted after treatment discontinuation. The effects of the mGluR5 antagonist MTEP (7.5 mg/kg,
IP) on food consumption were in the same direction as seen with memantine, but the observed differences were not significant.
In an additional control experiment, sibutramine and memantine reduced unlimited (24 h) chow intake during the treatment phase.
Present results provide evidence that glutamatergic neurotransmission might be involved in the regulation of excessive consumption
of highly palatable foods, and suggest that NMDA receptor may be an attractive target for developing obesity and disordered

In the aquatic environment, mercury is readily methylated into its most toxic form of methylmercury. In this form, it enters the aquatic food chain and its concentrations increase in subsequent links, which decreases the quality of fish meat and poses risks to consumer health. Concentrations of methylmercury (MeHg) and total mercury (THg) were determined in the muscle tissues of 64 eel specimens measuring from 59 to 95 cm in length as functions of specimen size and weight. Risks posed to consumers by eel from different length classes were also assessed. The mean concentration of THg in all of the eel examined was 0.179 mg kg?1, but the range was from 0.028 to 0.487 mg kg?1. The mean concentration of MeHg was 0.147 mg kg?1, and the range was also wide from 0.023 to 0.454 mg kg?1. Accumulated MeHg and THg increased with eel body length. The percentage share of MeHg in THg also changed with specimen length, and there was a positive correlation between the concentrations of MeHg and THg. Risk assessment was performed based on the doses of THg and MeHg ingested with fish for several specimen length classes. Consuming the meat of eel measuring 80 cm in length increased the estimated weekly intake (EWI) of THg and MeHg twofold in comparison to that from specimens 60 cm in length and fourfold in specimens exceeding 90 cm in length. The percentage shares of the EWI in the tolerable weekly intake and the target hazard quotient coefficient also increased proportionally. Generally, concentrations of MeHg and THg in eel are below current limits and pose no risk to consumer health as long as the consumption of larger specimens is avoided. 相似文献

The majority of male songbirds have small repertoires and sing with eventual variety; that is, they present one song type several times before switching to the next one. Several hypotheses have been proposed to explain this phenomenon. The antiexhaustion hypothesis argues that song‐type switching prevents muscle fatigue in the syrinx. The signal redundancy hypothesis suggests that repeating the same signal increases transmission success. Here, we have studied the song behaviour of the chaffinch, Fringilla coelebs, a common Eurasian species in which the males sing a few different song types and provide eventual variety. We tested different hypotheses to explain the temporal organisation of song output (repertoire size, song rate, bout duration, etc.) as a function of ambient noise by comparing birds from the same macrogeographic region in which the birds live either in a noisy town (n = 71) or in a quieter forest habitat (n = 68). Contrary to the prediction of the signal redundancy hypothesis and the results of earlier work on chaffinches living close to noisy streams, we found no significant differences in song characteristics between the town and forest populations. Our results support the antiexhaustion hypothesis because males with larger repertoires were able to sing with a significantly higher rate due to faster switching between different song types and producing shorter bouts. Sample size or population differences between our study and earlier investigations of the same species may explain the inconsistency with previous findings. Future studies should focus on determining the relations between song organisation and the directly measured quality of males and females' choice using, preferably, a longitudinal approach. 相似文献

Resveratrol is a stilbene present in different plant species and exerting numerous beneficial effects, including prevention of diabetes and attenuation of some diabetic complications. Its inhibitory effect on insulin secretion was recently documented, but the exact mechanism underlying this action remains unknown. Experiments employing diazoxide and a high concentration of K(+) revealed that, in depolarized pancreatic islets incubated for 90 min with resveratrol (1, 10, and 100 microM), insulin secretion stimulated by glucose and leucine was impaired. The attenuation of the insulin secretory response to 6.7 mM glucose was not abrogated by blockade of intracellular estrogen receptors and was found to be accompanied by diminished islet glucose oxidation, enhanced lactate production, and reduced ATP levels. Glucose-induced hyperpolarization of the mitochondrial membrane was also reduced in the presence of resveratrol. Moreover, in depolarized islets incubated with 2.8 mM glucose, activation of protein kinase C or protein kinase A potentiated insulin release; however, under these conditions, resveratrol was ineffective. Further studies also revealed that, under conditions of blocked voltage-dependent calcium channels, the stilbene reduced insulin secretion induced by a combination of glucose with forskolin. These data demonstrate that resveratrol 1) inhibits the amplifying pathway of insulin secretion, 2) exerts an insulin-suppressive effect independently of its estrogenic/anti-estrogenic activity, 3) shifts islet glucose metabolism from mitochondrial oxidation to anaerobic,4) fails to abrogate insulin release promoted without metabolic events, and 5) does not suppress hormone secretion as a result of the direct inhibition of Ca(2+) influx through voltage-dependent calcium channels. 相似文献

Proteins anchor copper(II) ions mainly by imidazole from histidine residues located in different positions in the primary protein structures. However, the motifs with histidine in the first three N-terminal positions (His1, His2, and His3) show unique Cu(II)-binding properties, such as availability from the surface of the protein, high flexibility, and high Cu(II) exchangeability with other ligands. It makes such sequences beneficial for the fast exchange of Cu(II) between ligands. Furthermore, sequences with His1 and His2, thus, non-saturating the Cu(II) coordination sphere, are redox-active and may play a role in Cu(II) reduction to Cu(I). All human protein sequences deposited in UniProt Knowledgebase were browsed for those containing His1, His2, or His3. Proteolytically modified sequences (with the removal of a propeptide or Met residue) were taken for the analysis. Finally, the sequences were sorted out according to the subcellular localization of the proteins to match the respective sequences with the probability of interaction with divalent copper. 相似文献

The covalent conjugate of oligosaccharide core of Escherichia coli type R4 with tetanus toxoid was prepared using reaction of reductive amination. The neoglycoconjugate was a good immunogen in rabbits yielding a high level of anti-lipopolysaccharide (LPS) antibodies of the IgG class. It was found that antiserum was able to react with the smooth LPS molecules of identical (R4) or related (R1) core type. The reactions were shown in the enzyme-linked immunosorbent assay and the immunoblotting test. Flow cytometry showed that anti-core antibodies reacted with LPS present on intact, live, smooth bacteria labelling more than 90% of cells. The anti-OS R4-TT serum used for in vitro studies showed high endotoxin neutralization activity. The serum inhibited endotoxin-induced tumor necrosis factor alpha and nitric oxide synthesis by the J-774A.1 cell line and attenuated pulmonary retention of YAC-1 cells. 相似文献

The majority of mammals are unable to digest lactose due to post-weaning deactivation of the LCT gene, which is responsible for encoding the enzyme lactase (i.e., adult-type hypolactasia). A substitution of C with T at position −13910 bp upstream of the LCT gene has been linked to the lactase persistence phenotype in European populations. We investigated the frequency of LCT-13910C>T
polymorphism in 223 blood donors from central Poland. All samples were genotyped by polymerase chain reaction and direct sequencing.
The LCT-13910 T allele (lactase persistence) was present in 51% of individuals sampled from the Polish population. We did
not find any non-European variants associated with lactase persistence (LCT-13907C>G, LCT-13913T>C, LCT-13915T>G), or any
new polymorphisms within the sequenced region. Allele frequencies obtained are in agreement with results from other countries
and confirm the unique pattern of distribution of the LCT-13910C>T genotype in Europe. 相似文献
