Data on seasonal dynamics of bat activity in central Europe are scarce, mostly restricted to either summer period or autumn swarming. Few studies provide extensive interspecific comparisons of temporal activity patterns. Through broadband ultrasound detection, temporal variation of flight activity was studied in seven insectivorous bat species in N Poland for the first time. Seasonal activity pattern ofMyotis daubentonii (Kuhl, 1817) was clearly bimodal with the higher peak of activity in April and the second, smaller peak in August–September. The first peak is possibly associated with post-hibernal restoration of fat reserves, while the second one might result either from pre-hibernal fat accumulation, increase in energy demands of males during spermatogenesis or from dispersal of newly weaned juveniles. The highest flight activity ofNyctalus, Eptesicus andPipistrellus bats was noted in summer months, reflecting mainly an increase in energy demands between pregnancy and lactation. However, a distinct, second peak in activity ofN. noctula appeared at the end of September, probably matching the late migration wave of individuals from E Europe. The cluster analysis of species’ activity patterns reflected division on different hunting tactics, but not migratory behaviour.  相似文献

Hepatitis C virus (HCV) was discovered in 1989. HCV is a positive single-strand RNA. We all have thought, that HCV can replicate only in liver tissue, but now we know, that HCV can replicate in extrahepatic tissue as well. In about 48-86% of HCV infected patients, chronic hepatitis C (CHC) has been noticed and eventually, after tens of years, liver insufficiency, cirrhosis or hepatocellular carcinoma. The current recommended treatment for CHC is a combination of pegylated-interferon alpha and Ribavirin. Presently it is known, that HCV infection can persist as an occult infection. RNA HCV can be detected in patients after successful treatment for CHC or spontaneous elimination. Persistent HCV replication in hepatocytes or lymphoid cells would likely lead to continuous antigenic stimulation of the immune system. This prolonged replication may contribute to the immune tolerance of HCV, impairment of immune response and even further virus persistence. This occult infection grows more important in transplantation.  相似文献

Lipids extracted from the shiitake mushroom Lentinus edodes contain dolichols composed of 15 up to 19 isoprene units with Dol-17 as the dominating prenologue. Identification of dolichols was achieved by the application of 2D-TLC, HPLC and electrospray ionization mass spectrometry. Additionally a family of polyprenols (-unsaturated counterparts) with the same chain-length was also detected. Dolichols comprised approximately 0.002% of the fresh weight of the mushroom. Dolichols accompanied by traces of polyprenols are for the first time found in the mushroom tissue.  相似文献

In the 1920s and 1930s, Terzaghi and coworkers realized that the failure of various porous geomaterials under internal pore pressure is given through evaluating the failure function for the same materials at zero pressure, with 'total stress plus pore pressure' instead of 'total stress alone' as argument. As to check, probably for the first time, the relevance of this ('Terzaghi's') failure criterion for trabecular bone, a series of poromechanical and ultrasonic tests was conducted on bovine and human trabecular bone samples. Evaluation of respective experimental results within the theoretical framework of microporomechanics showed that (i) Terzaghi's effective stress indeed governs trabecular bone failure, (ii) deviatoric stress states at the level of the solid bone matrix (also called tissue level) are primary candidates for initiating bone failure, and (iii) the high heterogeneity of these deviatoric tissue stresses, which increases with increasing intertrabecular porosity, governs the overall failure of trabecular bone. Result (i) lets us use the widely documented experimental results for strength values of bone samples without pore pressure, as to predict failure of the same bone samples under internal pore pressure. Result (ii) suggests a favorable mode for strength modeling of solid bone matrix. Finally, result (iii) underlines the suitability of microfinite element simulations for trabecular bone microstructures.  相似文献

Homoprotocatechuate (HPCA) dioxygenases are enzymes that take part in the catabolism of aromatic compounds in the environment. They use molecular oxygen to perform the ring cleavage of ortho-dihydroxylated aromatic compounds. A theoretical investigation of the catalytic cycle for HPCA 2,3-dioxygenase isolated from Brevibacterium fuscum (Bf 2,3-HPCD) was performed using hybrid DFT with the B3LYP functional, and a reaction mechanism is suggested. Models of different sizes were built from the crystal structure of the enzyme and were used in the search for intermediates and transition states. It was found that the enzyme follows a reaction pathway similar to that for other non-heme iron dioxygenases, and for the manganese-dependent analog MndD, although with different energetics. The computational results suggest that the rate-limiting step for the whole reaction of Bf 2,3-HPCD is the protonation of the activated oxygen, with an energy barrier of 17.4 kcal/mol, in good agreement with the experimental value of 16 kcal/mol obtained from the overall rate of the reaction. Surprisingly, a very low barrier was found for the O-O bond cleavage step, 11.3 kcal/mol, as compared to 21.8 kcal/mol for MndD (sextet spin state). This result motivated additional studies of the manganese-dependent enzyme. Different spin coupling between the unpaired electrons on the metal and on the evolving substrate radical was observed for the two enzymes, and therefore the quartet spin state potential energy surface of the MndD reaction was studied. The calculations show a crossing between the sextet and the quartet surfaces, and it was concluded that a spin transition occurs and determines a barrier of 14.4 kcal/mol for the O-O bond cleavage, which is found to be the rate-limiting step in MndD. Thus the two 83% identical enzymes, using different metal ions as co-factors, were found to have similar activation energies (in agreement with experiment), but different rate-limiting steps.  相似文献

The goal of this study, conducted in seven large woodlands and three areas with small woodlots in northeastern Poland in 2004–2008, was to infer genetic structure in yellow‐necked mouse Apodemus flavicollis population and to evaluate the roles of environmental and population ecology variables in shaping the spatial pattern of genetic variation using 768 samples genotyped at 13 microsatellite loci. Genetic variation was very high in all studied regions. The primal genetic subdivision was observed between the northern and the southern parts of the study area, which harbored two major clusters and the intermediate area of highly admixed individuals. The probability of assignment of individual mice to the northern cluster increased significantly with lower temperatures of January and July and declined in regions with higher proportion of deciduous and mixed forests. Despite the detected structure, genetic differentiation among regions was very low. Fine‐scale structure was shaped by the population density, whereas higher level structure was mainly shaped by geographic distance. Genetic similarity indices were highly influenced by mouse abundance (which positively correlated with the share of deciduous forests in the studied regions) and exhibited the greatest change between 0 and 1 km in the forests, 0 and 5 km in small woodlots. Isolation by distance pattern, calculated among regions, was highly significant but such relationship between genetic and geographic distance was much weaker, and held the linearity at very fine scale (~1.5 km), when analyses were conducted at individual level.  相似文献

We investigate the performance of a simple corrugated aperture scanning near-field optical microscope (SNOM) probe with various cladding metals. The probes have only one corrugation, however, they offer increased transmission over both uncorrugated probes and those with many grooves. Enhancement of light throughput results from excitation of surface plasmons at the corrugation at the core–cladding interface. We show how the choice of metal influences radiation properties of grooved probes.  相似文献

The X-ray diffraction analysis, (13)C CP MAS NMR spectra and powder X-ray diffraction patterns were obtained for selected methyl glycosides: alpha- and beta-d-lyxopyranosides (1, 2), alpha- and beta-l-arabinopyranosides (3, 4), alpha- and beta-d-xylopyranosides (5, 6) and beta-d-ribopyranoside (7) and the results were confirmed by GIAO DFT calculations of shielding constants. In X-ray diffraction analysis of 1 and 2, a characteristic shortening and lengthening of selected bonds was observed in molecules of 1 due to anomeric effect and, in crystal lattice of 1 and 2, hydrogen bonds of different patterns were present. Also, an additional intramolecular hydrogen bond with the participation of ring oxygen atom was observed in 1. The observed differences in chemical shifts between solid state and solution come from conformational effects and formation of various intermolecular hydrogen bonds. The changes in chemical shifts originating from intermolecular hydrogen bonds were smaller in magnitude than conformational effects. Furthermore, the powder X-ray diffraction (PXRD) performed for 4, 5 and 7 revealed that 7 existed as a mixture of two polymorphs, and one of them probably consisted of two non-equivalent molecules.  相似文献

Milk urea concentration (MU) used by dairy producers for management purposes can be affected by selection for milk traits. To assess this problem, genetic parameters for MU in Polish Holstein-Friesian cattle were estimated for the first three lactations. The genetic correlation of MU with milk production traits, lactose percentage, fat to protein ratio (FPR) and somatic cell score (SCS) were computed with two 5-trait random regression test-day models, separately for each lactation. Data used for estimation (159,044 daily observations) came from 50 randomly sampled herds. (Co)variance components were estimated with the Bayesian Gibbs sampling method. The coefficient of variation for MU in all three parities was high (40–41 %). Average daily heritabilities of MU were 0.22 for the first parity and 0.21 for the second and third lactations. Average genetic correlations for different days in milk in the first three lactations between MU and other traits varied. They were small and negative for protein percentage (from ?0.24 to ?0.11) and for SCS (from ?0.14 to ?0.09). The weakest genetic correlation between MU and fat percentage, and between MU and lactose percentage were observed (from ?0.10 to 0.10). Negative average genetic correlation with the fat to protein ratio was observed only in the first lactation (?0.14). Genetic correlations with yield traits were positive and ranged from low to moderate for protein (from 0.09 to 0.33), fat (from 0.16 to 0.35) and milk yield (from 0.20 to 0.42). These results suggest that the selection on yield traits and SCS tends to increase MU slightly.  相似文献

The determination of the oligomeric state of functional enzymes is essential for the mechanistic understanding of their catalytic activities. RecQ helicases have diverse biochemical activities, but it is still unclear how their activities are related to their oligomeric states. We use single-molecule multi-color fluorescence imaging to determine the oligomeric states of Werner syndrome protein (WRN) during its unwinding and replication fork regression activities. We reveal that WRN binds to a forked DNA as a dimer, and unwinds it without any change of its oligomeric state. In contrast, WRN binds to a replication fork as a tetramer, and is dimerized during activation of replication fork regression. By selectively inhibiting the helicase activity of WRN on specific strands, we reveal how the active dimers of WRN distinctly use the energy of ATP hydrolysis for repetitive unwinding and replication fork regression.  相似文献
