Several studies on adult fish movement from marine protected areas to zones open to fishing activity conclude spillover is present, but most of these investigations use indirect evidence and small-sized species of little commercial importance. This paper reports the effects of manipulating a density gradient on movements of large-sized and commercially-important fish across “Jardines de la Reina” Marine Reserve boundaries, using tagging methods and visual census. Tagging was carried out using dart tags and modified spearguns at an experimental and a control site. Density of fish was experimentally manipulated on the unprotected side of the boundary. Before experimental manipulation, fish density was similar in both experimental and control sites and on both sides of the boundaries. After manipulation, fish density in the unprotected side of experimental site declined dramatically and a strong gradient was established through the boundary. One month later, this forced gradient disappeared, returning to the situation at the beginning of the study. This last result is due to spillover effect: the mean distance traveled by fish increased 1.5 times (mean from below 200 m to more than 300 m), the mean emigration rate doubled and the immigration rate decreased, allowing density levels to recover after manipulation.  相似文献

An increasing trend in non albicans infections and various susceptibility patterns to antifungal agents implies a requirement for the quick and reliable identification of a number of medically important Candida species. Real-time PCR followed by high resolution melting analysis (HRMA) was developed, tested on 25 reference Candida collection strains and validated on an additional 143 clinical isolates in this study. All reference strains and clinical isolates inconclusive when using phenotypic methods and/or HRMA were analysed using ITS2 sequencing. Considering reference and clinical strains together, 23 out of 27 Candida species could be clearly distinguished by HRMA, while the remaining 4 species were grouped in 2 pairs, when applying the mean Tm ± 3 SD values, the shape of the derivative melting curve (dMelt curve) and, in some cases, the normalized and temperature—shifted difference plot against C. krusei. HRMA as a simple, rapid and inexpensive tool was shown to be useful in identifying a wide spectrum of clinically important Candida species. It may complement the current clinical diagnostic approach based on commercially available biochemical kits.  相似文献

Plants are able to deal with variable environmental conditions; when exposed to strong illumination, they safely dissipate excess energy as heat and increase their capacity for scavenging reacting oxygen species. Both these protection mechanisms involve activation of the xanthophyll cycle, in which the carotenoid violaxanthin is converted to zeaxanthin by violaxanthin de-epoxidase, using ascorbate as the source of reducing power. In this work, following determination of the three-dimensional structure of the violaxanthin de-epoxidase catalytic domain, we identified the putative binding sites for violaxanthin and ascorbate by in silico docking. Amino acid residues lying in close contact with the two substrates were analyzed for their involvement in the catalytic mechanism. Experimental results supported the proposed substrate-binding sites and point to two residues, Asp-177 and Tyr-198, which are suggested to participate in the catalytic mechanism, based on complete loss of activity in mutant proteins. The role of other residues and the mechanistic similarity to aspartic proteases and epoxide hydrolases are discussed.  相似文献

An SAR study of an HTS screening hit generated a series of pyridodiazepine amines as potent inhibitors of Helicobacter pylori glutamate racemase (MurI) showing highly selective anti-H. pylori activity, marked improved solubility, and reduced plasma protein binding. X-ray co-crystal E–I structures were obtained. These uncompetitive inhibitors bind at the MurI dimer interface.  相似文献

Lysinibacillus sphaericus strains belonging the antigenic group H5a5b produce spores with larvicidal activity against larvae of Culex mosquitoes. C7, a new isolated strain, which presents similar biochemical characteristics and Bin toxins in their spores as the reference strain 2362, was, however, more active against larvae of Culex mosquitoes. The contribution of the surface layer protein (S-layer) to this behaviour was envisaged since this envelope protein has been implicated in the pathogenicity of several bacilli, and we had previously reported its association to spores. Microscopic observation by immunofluorescence detection with anti S-layer antibody in the spores confirms their attachment. S-layers and BinA and BinB toxins formed high molecular weight multimers in spores as shown by SDS-PAGE and western blot detection. Purified S-layer from both L. sphaericus C7 and 2362 strain cultures was by itself toxic against Culex sp larvae, however, that from C7 strain was also toxic against Aedes aegypti. Synergistic effect between purified S-layer and spore-crystal preparations was observed against Culex sp. and Aedes aegypti larvae. This effect was more evident with the C7 strain. In silico analyses of the S-layer sequence suggest the presence of chitin-binding and hemolytic domains. Both biochemical characteristics were detected for both S-layers strains that must justify their contribution to pathogenicity.  相似文献

Background

Congenital transmission is a major source of new Trypanosoma cruzi infections, and as vector and blood bank control continue to improve, the proportion due to congenital infection will grow. A major unanswered question is why reported transmission rates from T. cruzi-infected mothers vary so widely among study populations. Women with high parasite loads during pregnancy are more likely to transmit to their infants, but the factors that govern maternal parasite load are largely unknown. Better understanding of these factors could enable prioritization of screening programs to target women most at risk of transmission to their infants.

Methodology/Principal Findings

We screened pregnant women presenting for delivery in a large urban hospital in Bolivia and followed infants of infected women for congenital Chagas disease. Of 596 women screened, 128 (21.5%) had confirmed T. cruzi infection; transmission occurred from 15 (11.7%) infected women to their infants. Parasite loads were significantly higher among women who transmitted compared to those who did not. Congenital transmission occurred from 31.3% (9/29), 15.4% (4/26) and 0% (0/62) of women with high, moderate and low parasite load, respectively (χx2 for trend 18.2; p<0.0001). Twin births were associated with higher transmission risk and higher maternal parasite loads. Infected women without reported vector exposure had significantly higher parasite loads than those who had lived in an infested house (median 26.4 vs 0 parasites/mL; p<0.001) with an inverse relationship between years of living in an infested house and parasite load.

Conclusions/Significance

We hypothesize that sustained vector-borne parasite exposure and repeated superinfection by T. cruzi may act as an immune booster, allowing women to maintain effective control of the parasite despite the down-regulation of late pregnancy.  相似文献

Ecosystem services provided by mobile agents are increasingly threatened by the loss and modification of natural habitats and by climate change, risking the maintenance of biodiversity, ecosystem functions, and human welfare. Research oriented towards a better understanding of the joint effects of land use and climate change over the provision of specific ecosystem services is therefore essential to safeguard such services. Here we propose a methodological framework, which integrates species distribution forecasts and graph theory to identify key conservation areas, which if protected or restored could improve habitat connectivity and safeguard ecosystem services. We applied the proposed framework to the provision of pollination services by a tropical stingless bee (Melipona quadrifasciata), a key pollinator of native flora from the Brazilian Atlantic Forest and important agricultural crops. Based on the current distribution of this bee and that of the plant species used to feed and nest, we projected the joint distribution of bees and plants in the future, considering a moderate climate change scenario (following IPPC). We then used this information, the bee’s flight range, and the current mapping of Atlantic Forest remnants to infer habitat suitability and quantify local and regional habitat connectivity for 2030, 2050 and 2080. Our results revealed north to south and coastal to inland shifts in the pollinator distribution during the next 70 years. Current and future connectivity maps unraveled the most important corridors, which if protected or restored, could facilitate the dispersal and establishment of bees during distribution shifts. Our results also suggest that coffee plantations from eastern São Paulo and southern Minas Gerais States could suffer a pollinator deficit in the future, whereas pollination services seem to be secured in southern Brazil. Landowners and governmental agencies could use this information to implement new land use schemes. Overall, our proposed methodological framework could help design novel conservational and agricultural practices that can be crucial to conserve ecosystem services by buffering the joint effect of habitat configuration and climate change.  相似文献
