Intestinal entodiniomorphid ciliates are commonly diagnosed in the feces of wild apes of the genera Pan and Gorilla. Although some authors previously considered entodiniomorphid ciliates as possible pathogens, a symbiotic function within the intestinal ecosystem and their participation in fiber fermentation has been proposed. Previous studies have suggested that these ciliates gradually disappear under captive conditions. We studied entodiniomorphid ciliates in 23 captive groups of chimpanzees, three groups of captive bonobos and six populations of wild chimpanzees. Fecal samples were examined using Sheather's flotation and Merthiolate‐Iodine‐Formaldehyde Concentration (MIFC) methods. We quantified the number of ciliates per gram of feces. The MIFC method was more sensitive for ciliate detection than the flotation method. Ciliates of genus Troglodytella were detected in 13 groups of captive chimpanzees, two groups of bonobos and in all wild chimpanzee populations studied. The absence of entodiniomorphids in some captive groups might be because of the extensive administration of chemotherapeutics in the past or a side‐effect of the causative or prophylactic administration of antiparasitic or antibiotic drugs. The infection intensities of ciliates in captive chimpanzees were higher than in wild ones. We suppose that the over‐supply of starch, typical in captive primate diets, might induce an increase in the number of ciliates. Seasonal and diurnal variations of in-stream NO3-N concentration oscillations were studied through high-frequency measurements of streamwater’s physical, chemical parameters (in-stream NO3-N concentration, water temperature, dissolved oxygen, pH) and hydrometeorological variables (stream discharge, solar radiation) under hydrologically stable conditions. The study was carried out in 2006, within the 42 km2 forested Pade? stream watershed in the southwestern part of Slovenia, which is characterized by distinctive hydrogeological settings (flysch) and climate conditions (transitional area between the Mediterranean and continental climate). Fine temporal data resolution (15-min interval) enabled identification of the factors responsible for seasonal variability in the diurnal pattern of the streamwater NO3-N concentrations versus seasonal and diurnal behavior of meteorological and other water chemistry constituents. The observed seasonal variability of in-stream NO3-N daily oscillations indicates the important role of primary production uptake, particularly during seasons when deciduous vegetation is dormant and light levels in the stream are high. Highest daily NO3-N concentration amplitudes (0.3 mg/l-N) and daily changes in the NO3-N flux (0.4–0.5 g/s-N) were observed in spring; the NO3-N concentration oscillations in summer showed a considerably smaller effect of the in-stream uptake (maximum NO3-N daily concentration amplitude 0.1 mg/l-N; daily change in the NO3-N flux 0.02 g/s-N). Seasonal shifts in the timing of daily maximum (up to 6 h) and minimum NO3-N concentrations (between 1 and 3 h) provided some additional indications of seasonal changes in the in-stream primary production uptake and its relation to the terrestrial component of the forested watershed.  相似文献

The pel1 mutation in Saccharomyces cerevisiae and the Cgpgs1Δ mutation in Candida glabrata result in deficiency of mitochondrial phosphatidylglycerolphosphate synthase and lack of two anionic phospholipids, phosphatidylglycerol and cardiolipin. DNA sequence analysis of the PCR-amplified pel1 mutant allele revealed that the pel1 mutation resulted from a single amino-acid substitution (Glu463Lys) in the C-terminal part of encoded enzyme. The CgPGS1 gene cloned in a centromeric pFL38 vector functionally complemented the pel1 mutation in S. cerevisiae. Likewise, the ScPGS1 gene cloned in pCgACU5 plasmid fully complemented the Cgpgs1Δ mutation in C. glabrata. This mutation increased the cell surface hydrophobicity and decreased biofilm formation. These results support a close evolutionary relatedness of S. cerevisiae and C. glabrata and point to the relationship between expression of virulence factors and anionic phospholipid deficiency in pathogenic C. glabrata.  相似文献

We studied phenotypic relationships among 13 samples of two rock mice species:Apodemus mystacinus (Danford and Alston, 1877) from Anatolia (n = 38) andA. epimelas (Nehring, 1902) from the Balkans (n = 71). Cartesian coordinates of landmarks were collected on the skull and on the occlusal projection of the upper molars (18 landmarks). Centroid size (a measure of overall size) suggested that molars vary independently of overall skull size in both species. Discriminant function analysis on relative warp scores classified >80% of specimens into the correct species, with the best results obtained for the ventral aspect of the skull and for molars. Projection of the 1st discriminant function scores against centroid size provided good separation between the two species. Analysis of vector displacements associated with extremes of variation suggested considerable phenetic differences on the ventral side of the skull and in the molar shape of the two species. The great majority of shifts in landmarks were in a longitudinal direction and the rearrangements of molar cusps were more complex than was the case with the cranium. A set of 21 1-phenyl-5-benzylsulfanyltetrazoles substituted on the phenyl ring as well as on the benzyl moiety was evaluated for in vitro antimycobacterial activity against Mycobacterium avium and two strains of M. kansasii. The distinguishing property of Sm protein associations is very high stability. In order to understand this property, we analyzed the interfaces and compared the properties of Sm protein interfaces with those of a test set, the Binding Interface Database (BID). The comparison revealed that the main differences between the interfaces of Sm proteins and those of the BID set are the content of charged residues, the coordination numbers of the residues, knowledge-based pair potentials, and the conservation scores of hot spots. In Sm proteins, the interfaces have more hydrophobic and fewer charged residues than the surfaces, which is also the case for the BID test set and other proteins. However, in the interfaces, the content of charged residues in Sm proteins (26%) is substantially larger than that in the BID set (22%). Hot spots are residues that make up a small fraction of the interfaces, but they contribute most of the binding energy. These residues are critical to protein–protein interactions. Analyses of knowledge-based pair potentials of hot spot and non-hot spot residues in Sm proteins show that they are significantly different; their mean values are 31.5 and 11.3, respectively. In the BID set, this difference is smaller; in this case, the mean values for hot spot and non-hot spot residues are 20.7 and 12.4, respectively. Hence, the pair potentials of hot spots differ significantly for the Sm and BID data sets. In the interfaces of Sm proteins, the amino acids are tightly packed, and the coordination numbers are larger in Sm proteins than in the BID set for both hot spots and non-hot spots. At the same time, the coordination numbers are higher for hot spots; the average coordination number of the hot spot residues in Sm proteins is 7.7, while it is 6.1 for the non-hot spot residues. The difference in the calculated average conservation score for hot spots and non-hot spots in Sm proteins is significantly larger than it is in the BID set. In Sm proteins, the average conservation score for the hot spots is 7.4. Hot spots are surrounded by residues that are moderately conserved (5.9). The average conservation score for the other interface residues is 5.6. The conservation scores in the BID set do not show a significant distinction between hot and non-hot spots: the mean values for hot and non-hot spot residues are 5.5 and 5.2, respectively. These data show that structurally conserved residues and hot spots are significantly correlated in Sm proteins.  相似文献

Copper is known to induce oxidative stress in a number of models. It was shown that many pathophysiological events were associated with oxidative stress. Further, oxidative stress can increase gene expression of cytokines and of metalloproteinases. We previously found that copper toxic effects in isolated perfused rat livers were associated with significant oxidative stress (as assessed by lipid peroxidation, protein oxidation and oxidative DNA damage, particularly at concentration of 0.03 mM of Cu2+ in the perfusate). Here we investigated gene expression of tumor necrosis factor-alpha (TNF-α), interleukin-10 (IL-10); matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in frozen liver tissue samples by the real-time PCR assay. Compared to controls, copper at concentration of 0.01 mM did not affect gene expression of TNF-α, IL-10, MMP-2 and MMP-9, whereas copper at concentration of 0.03 mM significantly decreased gene expression of all the four TNF-α, IL-10, MMP-2 and MMP-9 by 69%, 81%, 43%, and 62%, respectively. These results suggest that copper-induced oxidative stress in the isolated rat liver can lead to the suppression of gene expression. Because TNF-α and metalloproteinases are involved also in liver regeneration, the suppression of these genes by copper may be one of the mechanisms by which acute intoxication of animals and humans with copper may impair regenerative capability of the liver.  相似文献

Micromorphological investigation of the types, dimensions and distribution of characteristic trichomes in leaves and stems in Teucrium L. species (T. arduini L., T. chamaedrys L., T. flavum L., T. montanum L., T. polium L., and T. scordium L. subsp. scordioides Schreb.) distributed in Croatia was carried out as part of the taxonomical study of the genus Teucrium. Secretory types of hairs, peltate and capitate hairs were observed on the epidermis of stems and leaves of all investigated species. Non-secretory, acicular hairs were almost completely lacking on stems of T. scordium subsp. scordioides. Flagelliform hairs were not found in T. flavum and T. polium. Cladose hairs were present only in T. polium. The largest micromorphological variability was established between wild and cultivated samples of T. arduini and T. scordium subsp. scordioides, while cultivated and wild specimens of T. polium were almost identical. Differences were primarily observed in trichome dimensions and much less in micromorphological features.  相似文献
