The interaction between calf thymus HMG14 and rat liver chromatin components has been studied via reconstitution and chemical cross-linking. Selective labeling of HMG14 with photoactivable reversible heterobifunctional reagents has allowed a clear identification of the histones interacting with it (histones H2A, H2B and H1). These results are not dependent on whether the chromatin samples used were bulk chromatin, mononucleosomes, or core particles (for H2A and H2B). In addition to histone proteins, DNA also seems to be involved in HMG14 attachment to nucleosome. 相似文献

The number of structural parameters were determined for spin-labelled angiotensin in aqueous solution with the use of fluorescence spectroscopy and 1H NMR relaxation induced by the spin label. At the same time all measured parameters were estimated theoretically by means of energy calculations and Monte-Carlo techniques. The matching procedure for experimental and computational data allows one to suggest a dynamic equilibrium between conformers of the molecule in aqueous solution and to estimate the values of their weights. 相似文献

A single X-chromosome balancer-bearingCelegans ♂+, as a founder of a strain (AF1), was isolated directly from Fl progeny of irradiated+dpy-8unc-3/lon-2++ hermaphrodites on the basis of the absence of recombinant F2 categories. The balancer chromosome (Bal-X-1) suppresses recombination over a two-thirds section of the X chromosome (between genesdpy-8 andlet-2) and is associated with a reciprocal translocation between linkage groups (LG) X and I. Animals homozygous for the translocation (szT1(X:1)) are nonviable. Hermaphrodites heterozygous for the translocation segregate male selfprogeny at a frequency of 0.08-0.12.Bal-X-l carries the marker mutationlon-2(e678) and can be detected cytologically. Forty-five strains of Staphylococcus hyicus subsp. hyicus and 36 strains of S. hyicus subsp. chromogenes were examined for bacteriolytic activity with the same assay system previously used in taxonomic studies on staphylococci. The two subspecies differed from each other chiefly in that for optimal lytic activity S. hyicus subsp. hyicus strains required a higher salt concentration in the test medium than S. hyicus subsp. chromogenes strains. The lack of lytic activity on B15TP1 medium was a major difference between S. hyicus and S. aureus, and the lack of activity on TP2P medium was a major difference between S. hyicus and S. intermedius. Penicillin-binding proteins (PBPs) were studied in 40 S. hyicus strains. The S. hyicus subsp. hyicus strains had only one PBP (mol. wt 79 000) while the S. hyicus subsp. chromogenes strains had three distinct PBPs (mol. wts 84 000, 82 000 and 79 000). 相似文献

The study of Escherichia coli strain D-2, which harbours the ftsA2(ts) allele, has shown that temperature-induced filaments of this strain can divide, at 30 degrees C, in the absence of DNA replication and translation. Strain D-2 is thermosensitive during a period coincident with that in which the termination protein should be synthesized and exert its action. The ftsA gene product, which participates in the structure of the septum, needs for its synthesis a short period of DNA replication. The FtsA protein could be involved in a mechanism that coordinates chromosome replication and cell division by a pathway different from and independent of the SOS-induced response. 相似文献

Examinations of 297 sera for diphtheria antitoxin and 160 sera for tetanus antitoxin were carried out in 1981. All sera were obtained from the cord blood of mothers between 15 and 34 years of age. The mothers were divided into four age groups each of which was further subdivided into the primipara and multipara subgroups. The aim was to assess the age-specific variations in response to active immunization against diphtheria and tetanus. The protective level of diphtheria antitoxin (at least 0.01 I.U./ml) was recorded in the serum of 96.3% of examinees and the rates of seropositivity were found to fall with increasing age. The protective level of tetanus antitoxin (at least 0.1 I.U./ml) was found in the serum of 95.2% of mothers. The serologic response encountered in groups of older mothers was a clear-cut demonstration that the country-wide mass immunization against tetanus carried out between 1974 and 1975 was highly effective and fully justified. The variations in the diphtheria and tetanus antitoxin levels found in the primipara and multipara subgroups were not statistically significant. 相似文献

Inhibition of liver prenyltransferase by citronellyl and geranyl phosphonate and phosphonylphosphate
Citronellyl- and geranylphosphonic acids and the corresponding phosphonylphosphates were made and tested as inhibitors of liver prenyltransferase. Kinetic analysis showed that citronellyl- and geranylphosphonylphosphate were powerful inhibitors of the enzyme, and that they were competitive inhibitors with geranyl diphosphate and noncompetitive inhibitors with isopentenyl diphosphate. Two inhibition constants, representing the equilibria [E][I]/[EI] = K5 and [ES1][I]/[ES1I] = K9, have been defined for the inhibitors. For citronellylphosphonylphosphate, the value of K5 was 1.25 microM and K9 was 3.30 microM; for geranylphosphonylphosphate, K5 = 1.50 microM and K9 = 1.60 microM. The phosphonates were very poor linear mixed noncompetitive inhibitors with respect to both substrates of the transferase. 相似文献

When the RNA processing enzyme RNAase E is inactivated in an Escherichia coli strain carrying derivatives of the colicin E1 plasmid, a small RNA, about 100 nucleotides long, accumulates. Structural analysis of this RNA showed that it is RNA I, the RNA that inhibits plasmid DNA synthesis. RNA I is a specific substrate for RNAase E and the cleavage takes place between the fifth and sixth nucleotides from the 5' end of the molecule. This is only the second natural RNA substrate that has been found, so far, for the RNA processing enzyme ribonuclease E, the other being a precursor for 5 S ribosomal RNA. It is remarkable that nine nucleotides around the cleavage sites are identical in both substrates: (Formula: see text). Therefore, we suggest that at least part of the interaction between RNAase E and its substrate is controlled by these nine nucleotides. 相似文献

Host-parasite relationships of Pratylenchus zeae and Quinisulcius acutus, alone or in combination, were studied on sorghum in the greenhouse and laboratory. Q. acutus at 1,000 or 5,000 nematodes per 15-cm-d pot and P. zeae at 500 nematodes per pot significantly suppressed plant height and fresh and oven dry shoot and root weights. A mixture of 1,000 Q. acutus and 500 P. zeae per pot resulted in greatest suppression of growth. Roots of plants inoculated with Q. acutus alone were reduced in number and size and showed lesions and discoloration. Reproduction of this nematode 42 days after inoculation was much greater in treatments of 100 or 1,000 than 5,000 nematodes. The population density of the two species at 6 weeks after inoculation was significantly less when combined than for each species alone. When the two species were combined, reproduction of P. zeae was greater than that of Q. acutus, but the final populations per gram of root weight were the same. Q. acutus fed ectoparasitically on epidermal cells of sorghum roots in the zone of elongation and differentiation when observed under in vitro conditions. 相似文献

The concentration of ACTH and corticosterone in plasma were measured following peripheral administration of naloxone and naloxone methylbromide (quaternary derivative of opiate antagonist naloxone which is thought not to cross the blood brain barrier) in male rats. Subcutaneous administration of naloxone methylbromide in the dose range of 0.625 - 5.0 mg kg-1 resulted in a small but significant increase in plasma corticosterone levels. Both naloxone and its quaternary derivative injected via permanent intraperitoneal catheters to freely moving rats induced a highly significant increase in plasma ACTH levels measured in blood obtained via permanent tail artery catheters 30 min following injection. These results indicate that loci outside the blood brain barrier are, at least partially, involved in the naloxone-induced stimulation of ACTH release. 相似文献
