By superinfection of human immunodeficiency virus type 2 (HIV-2) strain HIV-2ben-infected macaques with simian immunodeficiency virus (SIV) strain SIVmac, we investigated the mutual influences of an apathogenic and a pathogenic virus in vivo. Four rhesus and two cynomolgus monkeys were infected with HIV-2ben in 1988 and 1989, respectively. Virus could be reisolated from five of six animals 6 weeks after infection. The monkeys remained healthy over the next 2 to 3 years. PCR for viral RNA became negative, and virus could no longer be reisolated by coculture. All six macaques were superinfected with the pathogenic SIVmac251/32H. Subsequently, five monkeys became persistently viremic, while one animal was protected against the SIVmac infection. In the peripheral blood mononuclear cells and cocultures of the five viremic animals, DNA from both HIV-2 and SIVmac was present. The plasma contained RNA from both viruses. Thus, superinfection with SIVmac activated HIV-2. A proliferative T-cell response against both HIV-2 and SIVmac was measured in all animals after superinfection. Such a response was regularly seen after infection with the apathogenic HIV-2 but never when the pathogenic SIVmac alone was administered. While naive control monkeys inoculated with SIVmac251/32H regularly develop AIDS-like symptoms soon after infection and have to be killed, none of the preinfected animals has developed AIDS-like symptoms, but two of six animals developed tumors. After the SIVmac challenge, however, apoptotic lymphocytes were detected in the peripheral blood mononuclear cells of all animals. Thus, the presence of an apathogenic viral variant seems to retard the disease occurring after infection with a pathogenic virus rather than to confirm total protection. This partial protection appears to depend on a specific proliferative T-cell response early after infection.  相似文献

Left ventricular stroke volume (LVSV) falls during obstructed inspiration in animals and normal human subjects through mechanisms that may be closely related to pleural pressure. In this study we postulated that a similar reduction in LVSV should occur in patients with obstructive sleep apnea (OSA). Daytime polysomnograms were performed in 10 patients with OSA. A noninvasive electrical impedance method was used to determine LVSV. Pleural pressure was measured by esophageal balloon. In comparison with awake values, during OSA we found reductions in LVSV, cardiac output, and heart rate of 18, 27, and 11%, respectively (P less than 0.01). We observed that systolic pleural pressure did not have a significant effect on LVSV (P greater than 0.05). However, at pleural pressures lower than 10 cmH2O below resting expiratory level, there was a linear relationship between falls in LVSV and falls in middiastolic pleural pressure (P less than 0.0001). We concluded that reduced LVSV shown in patients with OSA was significantly related to diastolic pleural pressure level. Our findings suggested reduced preload as the most likely mechanism for decreased cardiac output in OSA.  相似文献

Cardiac deaths are more likely to be unanticipated than cancer deaths by patients, their families, and their physicians. We hypothesized that differing physician attitudes toward dying patients may affect the degree of expectation of death. To evaluate differences in attitudes and behaviors among subspecialists, we surveyed a randomly selected population of California subspecialists; 44 of 136 (32%) of cardiologists and 91 of 167 (55%) of oncologists responded. Oncologists experienced three times as many deaths as cardiologists and reported having discussed code status more often with patients who died. Cardiologists'' patients'' deaths were more often unexpected and occurred more frequently in intensive care units. In addition, their patients were more likely to be given cardiopulmonary resuscitation. Oncologists reported being more comfortable dealing with dying patients and having less desire to avoid them. When presented with patient scenarios, however, cardiologists'' and oncologists'' responses were similar when discussing and estimating prognosis and likelihood of successful therapy.  相似文献

Background  

Long-Term Potentiation (LTP) of synapses is thought to be due in part to a change in AMPA Receptor trafficking leading to an increase in the number of AMPA Receptors at the synapse. LTP onset occurs within seconds after the induction signal. A particle-based stochastic simulation software is used to investigate the effect of Brownian diffusion of glutamate receptors on receptor incorporation into the synaptic specialisation and the time-course of LTP expression. The model of the dendritic spine includes receptors diffusing within the membrane, scaffold molecules within the synaptic specialisation capable of binding receptors and a molecular picket-fence surrounding the synaptic membrane area, all features found within the biological system.  相似文献

With the increasing demand for higher throughput single nucleotide polymorphism (SNP) genotyping, the quantity of genomic DNA often falls short of the number of assays required. We investigated the use of degenerate oligonucleotide primed polymerase chain reaction (DOP-PCR) to generate a template for our SNP genotyping methodology of fluorescence polarization template-directed dye-terminator incorporation detection. DOP-PCR employs a degenerate primer (5′-CCGACTCGAGNNNNNNATGTGG-3′) to produce non-specific uniform amplification of DNA. This approach has been successfully applied to microsatellite genotyping. We compared genotyping of DOP-PCR-amplified genomic DNA to genomic DNA as a template. Results were analyzed with respect to feasibility, allele loss of alleles, genotyping accuracy and storage conditions in a high-throughput genotyping environment. DOP-PCR yielded overall satisfactory results, with a certain loss in accuracy and quality of the genotype assignments. Accuracy and quality of genotypes generated from the DOP-PCR template also depended on storage conditions. Adding carrier DNA to a final concentration of 10 ng/µl improved results. In conclusion, we have successfully used DOP-PCR to amplify our genomic DNA collection for subsequent SNP genotyping as a standard process.  相似文献

Several sarcoma-inducing viruses encode protein kinases that phosphorylate tyrosine residues. Such enzymatic activities can be detected within the detergent-insoluble matrix of transformed fibroblasts. We have analysed the protein kinase activities in two murine lymphoma cell lines ( MBL2 and LSTRA) induced by Moloney murine leukemia virus (Mo-MuLV). After incubation of the detergent-insoluble matrix of these cells with [gamma-32P]ATP, several alkali-resistant phosphoproteins, including a very heavily labelled 55 000 mol. wt. protein ( p55 ), have been detected in LSTRA, reflecting the activity of a protein kinase specific to this cell line. This protein kinase activity shares some of the distinctive properties of the protein kinases of transforming viruses, i.e., specificity for tyrosine residues, association with membranous and/or cytoskeletal structures, and inhibition by a synthetic peptide derived from the phosphorylation site of pp60src. In view of the absence of a transforming gene in MoMuLV , it is likely that the high level of protein kinase detected in the LSTRA cell line arises from the expression of a cellular gene.  相似文献

Summary This paper describes the design, extraction, and maintenance of intact soil-core microcosms which accurately predicted biomass reduction, nutrient loss and trace element enrichment in field-grown crops amended with acidic precipitator fly ash. These agricultural microcosms sustain ecosystem-level interactions while offering a cost-effective, manageable, experimental unit which can be subjected to laboratory controls.  相似文献

The selection of nucleic acid aptamers is an increasingly important approach to generate specific ligands binding to virtually any molecule of choice. However, selection-inherent amplification procedures are prone to artificial by-product formation that prohibits the enrichment of target-recognizing aptamers. Little is known about the formation of such by-products when employing nucleic acid libraries as templates. We report on the formation of two different forms of by-products, named ladder- and non-ladder-type observed during repetitive amplification in the course of in vitro selection experiments. Based on sequence information and the amplification behaviour of defined enriched nucleic acid molecules we suppose a molecular mechanism through which these amplification by-products are built. Better understanding of these mechanisms might help to find solutions minimizing by-product formation and improving the success rate of aptamer selection.  相似文献

Three approaches recommended for characterization of toxicity impact potential in a life cycle impact assessment (LCIA) are tested on a case study and compared. The two equivalency factor methods are the Persistence, Bioaccumulation, and Toxicity (PBT) method and the Multimedia Fate Modeling (MFM) method using a Mackay Level III model with state-specific environmental data. The simplified risk assessment (SRA) method involved dispersion modeling using site-specific environmental data. The life cycle inventory information evaluated by all three methods was limited to manufacturing of the RDX-based explosive in Kingsport, Tennessee. The effort to collect site-specific environmental data and conduct air dispersion modeling for the SRA method required about 24 times more effort than the PBT method and about 4 times more effort than the MFM method. Direct comparison of impact potential scores for the three approaches were limited to inhalation toxicity scores for nine air pollutants modeled by SRA. Correlations were made on the rank order of the impact potential scores for the nine air emissions evaluated for all three LCIA methods. Although the number of chemicals compared is very limited, the best correlation coefficient (0.96) was between the rank orders for the MFM and the SRA methods. The minimal effort and reduced accuracy of the PBT approach make it best suited for screening large numbers of chemicals for further evaluation of the highest ranked chemicals. The intermediate effort and reasonable accuracy (includes transfers to other media) of the MFM approach make it well suited for LCIAs involving comparative assertions or governmental policy decisions. The maximum effort and assumption of highest accuracy make the SRA approach suitable only after limiting the locations of interest to a few sites by screening with the other two approaches.  相似文献
